Proteasome inhibitors have shown relevant clinical activity in several hematological malignancies, namely in multiple myeloma and mantle cell lymphoma, improving patient outcomes such as survival and quality of life, when compared with other therapies. However, initial response to the therapy is a challenge as most patients show an innate resistance to proteasome inhibitors, and those that respond to the therapy usually develop late relapses suggesting the development of acquired resistance. The mechanisms of resistance to proteasome inhibition are still controversial and scarce in the literature. In this review, we discuss the development of proteasome inhibitors and the mechanisms of innate and acquired resistance to their activity—a major challenge in preclinical and clinical therapeutics. An improved understanding of these mechanisms is crucial to guiding the design of new and more effective drugs to tackle these devastating diseases. In addition, we provide a comprehensive overview of proteasome inhibitors used in combination with other chemotherapeutic agents, as this is a key strategy to combat resistance. 相似文献

Auranofin (AF, hereafter) is an orally administered chrysotherapeutic agent approved for the treatment of rheumatoid arthritis that is being repurposed for various indications including bacterial infections. Its likely mode of action involves the impairment of the TrxR system through the binding of the pharmacophoric cation [AuPEt3]+. Accordingly, a reliable strategy to expand the medicinal profile of AF is the replacement of the thiosugar moiety with different ligands. Herein, we aimed to prepare the AF analogue bearing the acetylcysteine ligand (AF-AcCys, hereafter) and characterize its anti-staphylococcal activity. Biological studies revealed that AF-AcCys retains an antibacterial effect superimposable with that of AF against Staphylococcus aureus, whereas it is about 20 times less effective against Staphylococcus epidermidis. Bioinorganic studies confirmed that upon incubation with human serum albumin, AF-AcCys, similarly to AF, induced protein metalation through the [AuPEt3]+ fragment. Additionally, AF-AcCys appeared capable of binding the dodecapeptide Ac-SGGDILQSGCUG-NH2, corresponding to the tryptic C-terminal fragment (488–499) of hTrxR. To shed light on the pharmacological differences between AF and AF-AcCys, we carried out a comparative experimental stability study and a theoretical estimation of bond dissociation energies, unveiling the higher strength of the Au–S bond in AF-AcCys. From the results, it emerged that the lower lipophilicity of AF-AcCys with respect to AF could be a key feature for its different antibacterial activity. The differences and similarities between AF and AF-AcCys are discussed, alongside the opportunities and consequences that chemical structure modifications imply. 相似文献

Electro-conducting doped polypyrrole was deposited by in situ oxidative polymerisation on PET non-wovens. Thermal properties were evaluated by means of DSC and TG in nitrogen and air.
Flame resistance tests reveal that coated PET fibres resist to direct contact with fire. By thermal analysis, it was found
that PPy reduces the temperature at which thermo-oxidative degradation of PET occurs. Polypyrrole-coated PET non-wovens were
heated above the melting point of PET for 30 min. After the heating the fibres become brittle and frail, but SEM observations

Persistent organic pollutants (POPs) are greatly noxious chemicals in environment, and they can cumulate in organisms and transfer between different species. Therefore, it is significant to detect POPs for both environmental evaluation and further treatment. However, developing facile approach for the detection of POPs still remains a challenge so far. In this paper, we report an innovative method for facile detection of POPs using gas sensor for the first time. Porous SnO2 nanostructures with a special tri-walled structure prepared via hydrothermal route and annealing process, were employed as gas-sensing materials. Through gas measurements, it was revealed that the as-fabricated gas sensor exhibited highly sensitive performance towards target POPs, including methoxychlor, mirex, p,p′-DDT, and aldrin. Moreover, we found that target POPs were distinguishable by extracting characteristics in kinetic curves of gas adsorption-desorption. As the presented detecting approach is facile without the requirements of complex operations, expensive and bulky instruments, it is expected that it would be developed as a promising method for the detection of POPs, and thereby showing its significance for environmental monitoring. 相似文献

This paper describes a rapid and sensitive method for determination of the hair dye Basic Blue 41 in wastewater samples using screen‐printed carbon electrodes modified with graphene (SPCE/Gr). The method is based on the reversible reduction of azo groups of the dye at potential of ?0.23 V/?0.26 V, where both the anodic and cathodic currents increased 1,300 % when compared to screen‐printed carbon (SPCE) and glassy carbon electrodes (GCE). The optimization of a square wave voltammetric method was performed by means of 23 factorial design, Doehlert matrix and multi‐response assays, and the best parameters were: frequency (54.8 Hz), step potential (6 mV), pulse amplitude (43.7 mV) and pH 4.5. The analytical curve was constructed from 3.00×10?8 to 2.01×10?6 mol L?1, with detection and quantification limits of 5.00×10?9 and 1.70×10?8 mol L?1, respectively. The repeatability of the method evaluated for 10 consecutive measurements at concentrations of 1.70×10?7 mol L?1 and 1.70×10?6 mol L?1, showed relative standard deviation of 3.56 and 0.57 %, respectively. The sensor based in SPCE/Gr was successfully applied in wastewater samples collected from a drinking water treatment plant and validated by comparison with HPLC‐DAD method with good accuracy. 相似文献

在高应力滚-滑动(滑动率约10%)条件下,利用自制的磨损试验机研究了高钒高速钢的磨损性能,并利用电子显微镜分析了失效行为.结果表明:高钒高速钢的相对耐磨性是高铬铸铁(Cr20)的2倍以上.磨损失效形式为显微切削与疲劳剥落的复合,兼有碳化物碎裂.碳化物对磨损失效有重要作用,高铬铸铁中的杆状M7C3型碳化物易于弯曲、碎裂而在其内部形成大量裂纹,促进磨损表面产生大块的疲劳剥落;高钒高速钢中团块状VC硬度高、形态好、具有精细亚结构、不易碎裂,可有效地抵御显微切削和疲劳剥落,是高钒高速钢耐磨性优良的原因. 相似文献

Electrochemical impedance spectroscopy is finding increasing use in electrochemical sensors and biosensors, both in their characterisation, including during successive phases of sensor construction, and in application as a quantitative determination technique. Much of the published work continues to make little use of all the information that can be furnished by full physical modelling and analysis of the impedance spectra, and thus does not throw more than a superficial light on the processes occurring. Analysis is often restricted to estimating values of charge transfer resistances without interpretation and ignoring other electrical equivalent circuit components. In this article, the important basics of electrochemical impedance for electrochemical sensors and biosensors are presented, focussing on the necessary electrical circuit elements. This is followed by examples of its use in characterisation and in electroanalytical applications, at the same time demonstrating how fuller use can be made of the information obtained from complete modelling and analysis of the data in the spectra, the values of the circuit components and their physical meaning. The future outlook for electrochemical impedance in the sensing field is discussed. 相似文献

This study used substituted barium hexaferrites, which were previously prepared and reported by the authors, to detect acetaminophen by the modification of a conventional glassy carbon electrode (GCE), which led to promising results. The synthesis of this electrode-modifying material was conducted using a citrate sol gel process. A test synthesis using glycerin and propylene glycol revealed that glycerin produced a better result, while less positive anodic potential values were associated with the electrooxidation of N-acetyl-p-aminophenol (NAP). Excellent electroactivity was exhibited by the cobalt-substituted barium-hexaferrite-nanomaterial-modified electrode. A good linear relationship between the concentration and the current response of acetaminophen (paracetamol) was obtained with a detection limit of (0.255 ± 0.005) µM for the Ba1.0Co1.22Fe11.41O18.11 GCE, (0.577 ± 0.007) µM for the Ba1.14Cu0.82Fe11.65O18.02 GCE, and (0.595 ± 0.008) µM for the bare GCE. The levels of NAP in a real sample of urine were quantitatively analyzed using the proposed method, with recovery ranges from 96.6% to 101.0% and 93.9% to 98.4% for the modified electrode with Cobalt-substituted barium hexaferrites (CoFM) and Copper-substituted barium hexaferrites (CuFM), respectively. These results confirm the high electrochemical activity of Ba1.0Co1.22Fe11.41O18.11 nanoparticles and thus their potential for use in the development of sensing devices for substances of pharmaceutical interest, such as acetaminophen (NAP). 相似文献

利用反射式红外传感器和少量的电子器件(单稳态触发器),研制了智能型节水控制系统。该系统能够识别是否有人入厕,酌情产生冲洗信号, 躯动进水电磁阀开启,向浮球式水箱内注水,待注满足以冲刷干净所需的两水箱水后,便自动地切断电磁阀中电源,达到了既卫生又节约的目的。在某一教学楼应用该系统一年多来,与无使用此系统的同规模另一教学楼相比,节水约70%。该系统元件少、成本低、结构合理、工作可靠、安装容易、维修方便,有广阔的应用前景。 相似文献
