凝胶过滤色谱法测定泥鳅多糖的组成及分子量   总被引:14,自引:0,他引:14  
从泥鳅粘液中分离提取到中性的粗多糖 ,采用凝胶过滤色谱柱在高效液相色谱仪上进行分离和测定 ,结果表明 ,该多糖由一个高聚糖 (MAP ,19 1% )和一个寡糖 (MAO ,80 9% )组成。用标准的Pullulan(P 112 0 0 0 ,P 4 730 0 ,P 2 2 80 0 ,P 5 90 0 ,P 2 70 0 )作标准曲线 ,测得其重均分子量分别为 130 2 5 2 (约 72 3个糖基 )和 15 39(约 9个糖基 )。粗多糖经SephadexG 10 0凝胶柱 (1cm× 5 0cm)过滤 ,分离得到纯化的MAP和MAO。  相似文献

平菇多糖属生物高分子, 文章对平菇多糖稀溶液性质进行了研究。实验系采用Elias浊点滴定法测定平菇多糖溶液的θ组成, 用快速动态膜渗透计和粘度法测定平菇多糖的数均分子量、维利系数与分子链的构型参数。探讨其在θ条件下的分子形态及用Tung函数法等处理分级数据, 从而获得平菇多糖的分子量分布。  相似文献

In this paper, superhigh reproductive rate strains of MEV with titre more than HA8192* or TCID50 log9.7 10 have been achieved both by cultivation in cell lines with different susceptibility to MEV and by isolating and identifying in field by the author. The systematic tests proved that S18 and L12 strains of MEV are the best strains for vaccine preparation. In this study, the best means for the tissue cultivation of MEV and the most advanced technological process for the production and detection of serum-free cell-cultured MEV fluids with super-high HA titre in batches in large quantities have been established for the first time. Optimum conditions for MEV inactivation were determined, and safe and effective inactivated vaccines with mineral oil or A1(OH)3 gel adjuvant were successfully prepared with serum-free cell-cultured MEV fluids. Both vaccines with different adjuvants can be manufactured in batches in large quantities and have been widely used all over China since 1986. The change laws of the imm  相似文献

本文对不同产地和市售的二十四种厚朴药材及饮片的萃取液进行了高效液相色谱、气相色谱及紫外光谱分析;对所得数据用主成分分析进行分类,概括出不同样品化学成分的异同,认为大叶木兰可做为厚朴的代用品,而在陕西安康购得的样品不应做厚朴使用,后者经生药学鉴定为厚朴伪品凹叶木兰。化学模式识别技术可为寻找药材的代用品、评价中药质量及鉴定伪劣药材提供依据。  相似文献

Differential scanning calorimetry (DSC) is the most widely used thermal analytical technique in food research and it has a great utility in quality assurance of food. Proteins are the most studied food components by thermal analysis including studies on conformation changes of food proteins as affected by various environmental factors, thermal denaturation of tissue proteins, food enzymes and enzyme preparations for the food industry, as well as effects of various additives on their thermal properties. Freezing-induced denaturation of food proteins and the effect of cryoprotectants are also monitored by DSC. Polymer characterization based on DSC of polysaccharides, gelatinization behaviour of starches and interaction of starch with other food components can be determined, and phase transitions during baking processes can be studied by DSC. Studies on crystallization and melting behaviour of fats observed by DSC indicate changes in lipid composition or help characterizing products. Thermal oxidative decomposition of edible oils examined by DSC can be used for predicting oil stability. Using DSC in the freezing range has a great potential for measuring and modelling frozen food thermal properties, and to estimate the state of water in foods and food ingredients. Research in food microbiology utilizes DSC in better understanding thermoadaptive mechanisms or heat killing of food-borne microorganisms. Isothermic microcalorimetric techniques provide informative data regarding microbial growth and microbial metabolism.  相似文献

Between 1992 and 1994, a new screening campaign for blood Pb monitoring in the Italian general population was carried out. Since the first campaign (started in 1978, in accomplishment of the European Community Directive 77/312/EEC) a working group of the Laboratory of Clinical Biochemistry at the Italian National Institute of Health (Istituto Superiore di Sanità), as the Reference Centre (RC), has coordinated the activity of various laboratories spread over the national territory. Appropriate quality assurance procedures, including an external quality assessment scheme (EQAS), were elaborated. Within the EQAS, three or four trials were carried out every year. Each laboratory participating in the trial analyzed eight control samples prepared from cow blood at different Pb concentrations. The results obtained by each peripheral laboratory and the RC between 1992 and 1994 have been compared by regression analysis. The same statistical method was adopted to compare the results obtained by each peripheral laboratory and the RC in the duplicate analysis of about 10 per cent of the human samples collected during the 1992–1994 monitoring campaign. There was no evidence of systematic differences between the regression lines obtained on control and human samples. In spite of the lower Pb concentration in the control samples analyzed during the 1992–1994 campaign, the analytical performance of the laboratories was better than that obtained in the previous screening campaign (1985–1986). Blood Pb levels observed in human samples collected between 1992 and 1994, confirm the downward time trend observed in the campaigns carried out in 1978–1979, 1980–1981 and 1985–1986. This study confirms that the results obtained in an EQAS are representative of the actual performance in the analysis of real (human) samples.  相似文献

The effect of catalyst dibutyltin dilaurate (DBTDL) on the kinetics of urethane formation reactions of α,ω‐bis(hydroxy)‐terminated fluoropolyethers Fomblin® Z‐DOL TXs (FPEs) of various molecular weights and poly(oxyethylene) glycol PEG‐400 with isophorone diisocyanate (IPDI) in hexafluoroxylene (HFX) and tetrahydrofuran (THF) at 40 °C and NCO:OH = 2:1 have been studied in a broad range of catalyst (0.10–9.00) ×10?4 M and total reagents (10.0–60.1 wt %) concentrations. The rate of tin‐catalyzed second‐order reactions (with respect to diol and diisocyanate) was found to be proportional to the square root of catalyst concentration [DBTDL]0.5 both in low polar (HFX) and polar (THF) solvents. Effect of catalyst saturation was revealed for all the reaction systems at higher DBTDL concentrations as well as the appearance of the limiting catalyst concentrations Clim below which the rates of reaction were close to zero. Based on these findings new effective rate coefficients have been derived k = kcat/(C ? C) that are independent of the total reagent concentration in the range of 10.0–60.1 wt % ([OH] = 0.10–0.91 equiv/L). This new approach highlights that the rate of the tin‐catalyzed urethane formation reactions of α,ω‐bis(hydroxy)‐terminated fluoropolyethers Z‐DOL TXs with IPDI in HFX at 40 °C and NCO:OH = 2:1 increases significantly with increasing MW of FPE from 776 up to 3405. © 2004 Wiley Periodicals, Inc. Explorative data analysis of two-dimensional electrophoresis gels   总被引:1,自引:0,他引:1  
Methods for classification of two-dimensional (2-DE) electrophoresis gels based on multivariate data analysis are demonstrated. Two-dimensional gels of ten wheat varieties are analyzed and it is demonstrated how to classify the wheat varieties in two qualities and a method for initial screening of gels is presented. First, an approach is demonstrated in which no prior knowledge of the separated proteins is used. Alignment of the gels followed by a simple transformation of data makes it possible to analyze the gels in an automated explorative manner by principal component analysis, to determine if the gels should be further analyzed. A more detailed approach is done by analyzing spot volume lists by principal components analysis and partial least square regression. The use of spot volume data offers a mean to investigate the spot pattern and link the classified protein patterns to distinct spots on the gels for further investigation. The explorative approach in analysis of 2-D gels makes it possible, in a fast and convenient way, to screen many gels in order to determine the protein patterns that form clusters and could be selected for further examination.  相似文献

 On September 1, 1992 all testing sites in the United States were required to comply with the Clinical Laboratory Improvement Amendments of 1988 (CLIA'88). These regulations, based on both total quality management (TQM) and continuous quality improvement (CQI) principles, reshaped the environment for more than 90% of laboratories. CLIA'88 represented a revolutionary change by imposing universal, uniform regulations based on test complexity for all sites examining materials derived from the human body for the purpose of providing information for the diagnosis, prevention, or treatment of disease. CLIA'88 specifies minimum requirements for personnel, quality control, and proficiency testing (PT). In addition, laboratories are required to follow manufacturers' directions and comply with other specified good laboratory practices. PT is mandated for most of the frequently run analyses and quality assurance requirements integrate the principles of CQI as well as TQM into the regulatory process. Biannual inspection is integral to CLIA'88, however, laboratories can choose other federally approved ("deemed") professional organizations, such as the Commission on Office Laboratory Accreditation, the College of American Pathologists, or the Joint Commission on Accreditation of Healthcare Organization, having standards that meet or exceed those of CLIA'88. Thermogravimetric analysis of hydroxyl terminated polybutadiene (HTPB) and its fractions of different molecular weights separated by preparative GPC shows two major stages of weight loss of different nature in a nitrogen atmosphere. The first stage is primarily depolymerisation, cyclisation and crosslinking of molecules and the second stage is mainly the decomposition of the residue from the first stage. The kinetic parameters, viz. activation energy E and pre-exponential factor A using four different non-isothermal integral equations show a systematic increase with increase in molecular weight for the first stage, whereas for the second stage, the effect of molecular weight on E and A values is not prominent. The increase in E and A values for the first stage is attributed to the formation of greater number of cyclised and crosslinked products from molecules of higher dimensions. Quantitative correlations between the kinetic constants and the molecular weight parameters were derived for the first stage as a quadratic curve following the equation: E or ln A = K1K2/M (where K1 and K2 are empirical constants whose values are different for the different molecular weight averages, viz. Mn, Mw and Mz and for the different equations).  相似文献
