采用人肠内细菌和乌头碱体外温孵的方法, 探讨去氧乌头碱在人肠内的生物转化. 利用离子阱和傅里叶变换离子回旋共振质谱直接分析去氧乌头碱的转化产物. 乌头类生物碱及其代谢产物在正离子电喷雾质谱条件下形成质子化分子([M+H]+), 通过多级串联质谱进行结构表征. 去氧乌头碱可被人肠内细菌转化, 通过脱酰基、脱甲基脱羟基以及酯化反应产生新型的单酯型、双酯型和脂类生物碱等10余种代谢产物. 双酯型的去氧乌头碱的毒性较高, 当它被肠内细菌转化为单酯型和脂类生物碱时会使其毒性降低.  相似文献

The utilization of fermented foods with health-promoting properties is becoming more popular around the world. Consequently, kefir, a fermented milk beverage made from kefir grains, was shown in numerous studies to be a probiotic product providing significant health benefits. Herein, we assessed the antibacterial and antifungal potential of kefir against a variety of pathogenic bacteria and fungi. This study also showed the effectiveness of kefir in healing wounds in human gastric epithelial cells (GES-1) by (80.78%) compared with control (55.75%) within 48 h. The quantitative polymerase chain reaction (qPCR) results of kefir-treated HCV- or HBV- infected cells found that 200 µg/mL of kefir can eliminate 92.36% of HCV and 75.71% of HBV relative to the untreated infected cells, whereas 800 µg/mL (the highest concentration) completely eradicated HCV and HBV. Moreover, the estimated IC50 values of kefir, at which HCV and HBV were eradicated by 50%, were 63.84 ± 5.81 µg/mL and 224.02 ± 14.36 µg/mL, correspondingly. Kefir can significantly suppress the elevation of TNF-α and upregulate IL-10 and INF-γ in both treated HCV- and HBV-infected cells. High-performance liquid chromatography (HPLC) and gas chromatography-mass spectrometry (GC-MS) analysis of kefir revealed the presence of numerous active metabolites which mainly contribute to the antimicrobial, antiviral, and immunomodulatory activities. This study demonstrated, for the first time, the anti-HBV efficacy of kefir while also illustrating the immunomodulatory impact in the treated HBV-infected cells. Accordingly, kefir represents a potent antiviral agent against both viral hepatitis C and B, as well as having antimicrobial and wound healing potential.  相似文献

Fucosidases are associated with several pathological conditions and play an important role in the health of the human gut. For example, fucosidases have been shown to be indicators and/or involved in hepatocellular carcinoma, breast cancer, and helicobacter pylori infections. A prerequisite for the detection and profiling of fucosidases is the formation of a specific covalent linkage between the enzyme of interest and the activity-based probe (ABP). The most commonly used fucosidase ABPs are limited to only one of the classes of fucosidases, the retaining fucosidases. New approaches are needed that allow for the detection of the second class of fucosidases, the inverting type. Here, we report an ortho-quinone methide-based probe with an azide mini-tag that selectively labels both retaining and inverting bacterial α-l-fucosidases. Mass spectrometry-based intact protein and sequence analysis of a probe-labeled bacterial fucosidase revealed almost exclusive single labeling at two specific tryptophan residues outside of the active site. Furthermore, the probe could detect and image extracellular fucosidase activity on the surface of live bacteria.  相似文献

Development of novel derivatives to rein in and fight bacteria have never been more demanding, as microbial resistance strains are alarmingly increasing. A multitude of new fluoroquinolones derivatives with an improved spectrum of activity and/or enhanced pharmacokinetics parameters have been widely explored. Reporting novel antimicrobial agents entails comparing their potential activity to their parent drugs; hence, parent fluoroquinolones have been used in research as positive controls. Given that these fluoroquinolones possess variable activities according to their generation, it is necessary to include parent compounds and market available antibiotics of the same class when investigating antimicrobial activity. Herein, we provide a detailed guide on the in vitro biological activity of fluoroquinolones based on experimental results published in the last years. This work permits researchers to compare and analyze potential fluoroquinolones as positive control agents and to evaluate changes occurring in their activities. More importantly, the selection of fluoroquinolones as positive controls by medicinal chemists when investigating novel FQs analogs must be correlated to the laboratory pathogen inquest for reliable results.  相似文献

海洋发光细菌是一类在正常的生理条件下能够发射可见光的异养细菌,其发光特性与细菌体内的物质结构、海洋环境要素、水中污染物等多种因素有关。通过对海洋发光细菌发光特性及其应用可能性的论述,如利用发光细菌的发光强度与水中毒物的浓度、毒性的关系检测污染物;利用潜艇航行时激发的生物发光勾画出潜艇涡动的光尾流可跟踪探测潜艇;海洋发光细菌的发光特性还可用以改进水下光通讯与探测、海洋水色遥感、海洋发光细菌分类、发光免疫和抗菌素浓度测定等方面。阐明海洋发光细菌发光特性的重要研究意义并为进一步开展应用研究提供借鉴。  相似文献

采用人肠内细菌和乌头碱体外温孵的方法,探讨乌头碱的代谢产物16-O-去甲基去氧乌头碱在人肠内的生物转化。利用离子阱电喷雾串联质谱(ESIMS/MSn)方法直接分析16-O-去甲基去氧乌头碱的代谢产物。乌头类生物碱在ESI正离子模式条件下形成质子化分子[M H] 。16-O-去甲基去氧乌头碱可被人肠内细菌转化,通过脱乙酰基、脱苯甲酰基、脱甲基、脱羟基以及酯化反应产生新型的单酯型、双酯型和脂类生物碱等10余种代谢产物。双酯型生物碱具有较高的毒性,相对应的单酯型和脂类生物碱毒性较低。16-O-去甲基去氧乌头碱被肠内细菌转化为单酯型和脂类生物碱会使其毒性降低。  相似文献

The essential oils were obtained by hydrodistillation from aerial parts of Mentha pulegium L. (M. pulegium L.) and Artemisia herba alba (A. herba alba) Asso. and analyzed by gas chromatography–flame ionization detector chromatograpy (GC–FID) and gaz chromatography–mass spectrometry (GC–MS). The antibacterial activities of the oils were determined by the disk diffusion method and a microdilution broth assay against six bacteria stains. The combinations of these essential oils with antibiotics were evaluated against two multi-drug-resistant bacteria strains: imipenem-resistant Acinetobacter baumannii (IRAB S3310) and methicillin-resistant Staphylococcus aureus (MRSA S19). The chemical analysis of M. pulegium essential oil revealed the presence of pulegone (74.8%) and neoisomenthol (10.0%). A. herba alba essential oil was characterized by camphor (32.0%), α-thujone (13.7%), 1,8-cineole (9.8%), β-thujone (5.0%), bornéol (3.8%), camphene (3.6%), and p-cymene (2.1%). All strains tested except Pseudomonas aeruginosa were susceptible to these oils. The combinations of essential oils with antibiotics exerted synergism, antagonism, or indifferent effects. The best effect was observed with A. herba alba essential oil in association with cefoxitin (CX) against MRSA S19. However, for IRAB S3310, the strongest synergistic effect was observed with M. pulegium in association with amikacin (AK). This study demonstrated that M. pulegium and A. herba alba essential oils have antibacterial activities which could be potentiated by antibiotics especially in the case of IRAB S3310.  相似文献

There is growing interest in the detection of bacteria in consumables, for example, in the food and water sectors. In this study, the aim was to produce a polymer-based bacteria biosensor via ROMP (ring opening metathesis polymerization). In the first part of the study, block and random copolymers were synthesized, and their biocidal activities were tested on the glass surface. Interdigitated electrode arrays coated with the polymers possessing the highest activity were used to screen the affinity towards different bacterial strains by monitoring impedance variations in real-time. The polymer-coated electrode could detect gram-positive and gram-negative bacteria strains at a concentration of 107 cfu/mL. The results show that ROMP-based polymer offers bacterial detection and can be used in developing biosensor devices for efficiently detecting pathogenic bacteria.  相似文献

This study aimed to define a consortium of lactic acid bacteria (LAB) that will bring added value to dried fresh cheese through specific probiotic properties and the synthesis of bioactive peptides (biopeptides). The designed LAB consortium consisted of three Lactobacillus strains: S-layer carrying Levilactobacillus brevis D6, exopolysaccharides producing Limosilactobacillus fermentum D12 and plantaricin expressing Lactiplantibacillus plantarum D13, and one Enterococcus strain, Enterococcus faecium ZGZA7-10. Chosen autochthonous LAB strains exhibited efficient adherence to the Caco-2 cell line and impacted faecal microbiota biodiversity. The cheese produced by the LAB consortium showed better physicochemical, textural and sensory properties than the cheese produced by a commercial starter culture. Liquid chromatography coupled with matrix-assisted laser desorption/ionization-time of flight tandem mass spectrometry (LC-MALDI-TOF/TOF) showed the presence of 18 specific biopeptides in dried fresh cheeses. Their identification and relative quantification was confirmed by liquid chromatography-tandem mass spectrometry (LC-MS/MS) using multiple reaction monitoring (MRM). The results also showed that their synthesis resulted mainly from β-casein and also α-S1 casein degradation by proteolytic activities of the LAB consortium. The designed LAB consortium enhanced the functional value of the final product through impact on biopeptide concentrations and specific probiotic properties.  相似文献

用微量热法研究了Na~2SeO~3和箬叶多糖对大肠杆菌和产气肠杆菌作用的热功率输出曲线,得到了大肠杆菌和产气肠杆菌在不同条件下的生长速率常数k等参数。结果表明:Na~2SeO~3在低浓度下对大肠杆菌生长有促进作用,在高浓度时对两种细菌均有很强的抑制作用,而箬叶多糖对大肠杆菌和产气肠杆菌生长代谢未表现出抑制作用。这为揭示硒的生物效应、进行药理学和毒理学等的研究、寻找高效低毒的新药提供了可靠的信息。  相似文献
