In continuation of the search for new anthelmintic natural products, the study at hand investigated the nematicidal effects of the two naturally occurring quassinoids ailanthone and bruceine A against the reproductive system of the model nematode Caenorhabditis elegans to pinpoint their anthelmintic mode of action by the application of various microscopic techniques. Differential Interference Contrast (DIC) and the epifluorescence microscopy experiments used in the presented study indicated the genotoxic effects of the tested quassinoids (c ailanthone = 50 µM, c bruceine A = 100 µM) against the nuclei of the investigated gonadal and spermathecal tissues, leaving other morphological key features such as enterocytes or body wall muscle cells unimpaired. In order to gain nanoscopic insight into the morphology of the gonads as well as the considerably smaller spermathecae of C. elegans, an innovative protocol of polyethylene glycol embedding, ultra-sectioning, acridine orange staining, tissue identification by epifluorescence, and subsequent AFM-based ultrastructural data acquisition was applied. This sequence allowed the facile and fast assessment of the impact of quassinoid treatment not only on the gonadal but also on the considerably smaller spermathecal tissues of C. elegans. These first-time ultrastructural investigations on C. elegans gonads and spermathecae by AFM led to the identification of specific quassinoid-induced alterations to the nuclei of the reproductive tissues (e.g., highly condensed chromatin, impaired nuclear membrane morphology, as well as altered nucleolus morphology), altogether implying an apoptosis-like effect of ailanthone and bruceine A on the reproductive tissues of C. elegans.  相似文献

Polyethylene based composites are attractive materials for advanced circuit board applications because of their unique combination of properties: low dielectric constant and loss factor, light weight, high flexural modulus and low thermal expansion coefficient controlled in all spatial directions. This investigation describes a process to consolidate chopped fibers of ultra‐high molecular weight polyethylene concurrently with its bonding to a copper foil. Bonding is affected by a thin sheet of low‐density polyethylene, incorporating a crosslinking agent with a concentration gradient across the sheets thickness. In this single step process, the composite material is formed and bonded to the metal foil, achieving good adhesion without the use of extraneous glue. The purpose of this paper is studying the effect of incorporation of Multiwall Carbon Nanotubes (MWCNT) into two different nanocomposites in poly vinyl alcohol (PVA)/polyvinylpyrrolidone (PVP), and PVA/Polyethylene glycol (PEG). MWCNT were synthesized by chemical vapor deposition (CVD) method using acetylene and Fe/Co/Al2O3 as carbon precursor and catalyst, respectively. Nitric acid and sulfuric acid were used for purification and functionalization of MWCNT. Afterward, highly pure and functionalized MWCNT (0, 0.02, and 0.05% w/w) were incorporated in PVA/PVP and PVA/PEG to synthesize PVA/PVP/MWCNT and PVA/PEG/MWCNT nanocomposites hydrogel membranes that cross-linked by freezing–thawing. PEG and PVP were selected in these nanocomposites as dispersion matrix for MWCNT as well as for increasing the elasticity of the nanocomposites membranes. The morphology of the hydrogels was characterized by SEM, FTIR, XRD, TGA, and the mechanical properties of the hydrogel membranes were investigated. The swelling behavior in different pH-buffer solutions was studied as well as studying weight loss percentage and swelling kinetic. The drug releasing process of the hydrogel membranes was investigated using salicylic acid as a model drug. It was found that MWCNT are dispersed well into the polymers and crystallinity, mechanical properties and thermal stability of the hydrogels contain MWCNT are better than that without MWCNT. Maximum degree of swelling was observed at pH 7 and swelling degree increases with increasing the ratio of MWCNT in the hydrogels from 0.02 to 0.05%. All hydrogel membranes followed non-Fickian mechanism and drug releasing were controlled by varying the pH and amount of MWCNT.  相似文献

In this work, biaxially stretched polymer foams with well‐defined cellular structures were prepared from polyethylene via blown‐film extrusion and subjected to corona charging to produce a piezoelectric response. The charging parameters were first optimized in terms of charging voltage and needle distance, as well as the gas type and pressure to investigate their effect on the piezoelectric coefficient (d33). The results show that samples charged under nitrogen (N2) at 100 kPa had better d33 coefficient than those charged under ambient air or N2 at 20 kPa. Moreover, 2 different thermal pressure treatments were imposed to obtain an optimized eye‐like cellular structure with different cell aspect ratios (AR). The results showed that when the cells were elongated in both the longitudinal and transverse directions (higher AR), higher d33 coefficients were achieved. From all the samples produced, the best results were obtained for a longitudinal aspect ratio (AR‐L) of 7.1, a transversal aspect ratio (AR‐T) of 4.6, and a relative foam density of 0.52 leading to a d33 coefficient of 935 pC/N. This coefficient was further increased using reverse charging and multilayered films, reaching a maximum of 2550 pC/N. This value is much higher than typical ones reported so far for any polyethylene and polypropylene ferroelectrets. These results could increase the use of polyethylene in piezoelectric applications as these materials are very attractive for the large‐scale production of electret‐based sensors and transducers due to their low cost and easy processing.  相似文献

A series of trichlorotitanium complexes containing 2‐(1‐(arylimino)propyl)quinolin‐8‐olates was synthesized by stoichiometric reaction of titanium tetrachloride with the corresponding potassium 2‐(1‐(arylimino)propyl)quinolin‐8‐olates and was fully characterized by elemental analysis, nuclear magnetic resonance spectroscopy, and by single‐crystal X‐ray diffraction study of representative complexes. All titanium complexes, when activated with methylaluminoxane, exhibited high catalytic activity toward ethylene polymerization [up to 1.15 × 106 g mol?1(Ti) h?1] and ethylene/α‐olefin copolymerization [up to 1.54 × 106 g mol?1 (Ti) h?1]. The incorporation of comonomer was confirmed to amount up to 2.82 mol % of 1‐hexene or 1.94 mol % of 1‐octene, respectively. © 2011 Wiley Periodicals, Inc. The effect of pan-milling on the rheological properties of high density polyethylene (HDPE) was studied. Aninnovative milling apparatus, viz. an inlaid pan-mill, was used. Melt indexer, capillary rheometer, Haake Rheocord 90 single-screw extruder and Brabender rheometer were used to evaluate the rheologieal properties of HDPE. HDPE with higher initialmolecular weight and larger particle size was easier to degrade under pan-milling stress, as indicated by the melt index.Pressure oscillation in capillary flow occurred at significantly higher shear stress and shear rate for milled HDPE than forunmilled HDPE. The apparent shear viscosity of HDPE decreased with increasing times of milling. After milling, the flowactivation energy decreased and thus the sensitivity of viscosity to temperature was reduced. Die pressure and torque duringsingle screw extrusion were reduced significantly after milling. Plasticizing time as measured in a Brabander mixer decreasedmarkedly with increasing milling times.  相似文献

A quantitative study was undertaken of the anisotropy of low‐strain mechanical behavior for specially oriented polyethylene with controlled crystalline and lamellar orientation. The samples were prepared by the die drawing of injection‐molded rods of polyethylene and annealing. This produced a parallel lamellar structure for which a simple, three‐dimensional composite laminate model could be used to calculate the expected anisotropy. Experimental data, including X‐ray strain measurements of the lateral crystalline elastic constants, showed good quantitative agreement with the model prediction. The X‐ray strain measurements confirmed that the amorphous regions exert large constraints on the crystalline phase in the lateral directions, where an order of magnitude difference was found between the measured apparent lateral crystalline compliances in the lamellar‐stack sample and the expected values for a perfect crystal. © 2000 John Wiley & Sons, Inc.
