A Crossed Immunoelectrophoretic Technique for the Detection of the Down's Syndrome Protein

Abstract

The sera of seventy-one women and twenty-six men of various ages were examined by a two dimensional (crossed) immunoelectrophoresis technique which was modified to detect a protein (the Down's Syndrome protein) originally found in the sera of mothers of trisomy 21 children. The sensitivity of the method is reported to be 0.1 to 1.0 mg/L. Modifications included use of a Tris-Tricine buffer, high voltage in the second dimension, and relatively low antibody concentration to allow easy detection of the Down's Syndrome (DS) protein.

Of twenty-eight mothers and twelve fathers of trisomy 21 children, eighteen mothers and five fathers were positive, while of forty-three women and fourteen men having unaffected or no children, only seven women and two men were positive for the DS protein.

Since the presence of the Down's Syndrome protein may indicate a propensity for giving birth to a trisomy 21 affected child, this crossed immunoelectrophoretic technique may be used as a screening procedure for the DS protein and, therefore, as a tool in genetic counseling.

Blind separation of fluorescence spectra using sparse non‐negative matrix factorization on right hand factor

Sparse non‐negative matrix factorization on right side factor (SNMF/R) has better performance in feature extraction than non‐negative matrix factorization. In this work, SNMF/R was first used to separate the overlapped three‐dimensional fluorescence spectra of polycyclic aromatic hydrocarbons mixtures in pure water, lake water, and river water, respectively. It is found that the similarity coefficients between the acquired three‐dimensional spectra and the corresponding reference spectra with random initials are all above 0.80; the recognition rate of SNMF/R is higher than that of PARAFAC and non‐negative matrix factorization algorithms, especially in the case of lake water and river water samples. In addition, SNMF/R does not need any initialization scheme designing during spectra separation. These results demonstrate that SNMF/R is an appropriate algorithm to separate the overlapped fluorescence spectra of polycyclic aromatic hydrocarbons in aquatic environment accurately and effectively. CuI‐Catalyzed Multicomponent Synthesis of Aminoindolizines from Aldehydes,Amines, and Alkynes under Solvent‐Free Conditions

A direct and efficient approach to 1‐aminoindolizines through three‐component one‐pot reaction of heteroaryl aldehydes, secondary amines and terminal alkynes catalyzed by CuI under solvent‐free conditions has been developed. This methodology provides a rapid access to substituted aminoindolizines with good yields (up to 97%).     

Synthesis of 2,3‐Dihydro‐4H‐furo[3,2‐c] chromen‐4‐ones and 2,3‐Dihydronaphtho[2,3‐b]furan‐4,9‐diones by the Radical Cyclizations of Hydroxyenones with Electron-Rich Alkenes using Manganese(III) Acetate

We have obtained dihydrofurans 3a–j in the radical cyclization of 4‐hydroxycoumarin 1a and 2‐hydroxy‐1,4‐naphtoquinone 1b with electron rich alkenes 2a–i by manganese(III) acetate. Methods A and B, which have different molar ratios were studied comparatively in these reactions, and we observed that method B (molar ratio 2:1:3) gave the best results. Treatment of 4‐hydroxycoumarin 1a and electron rich alkenes 2a–e gave 2,3‐dihydro‐4H‐furo[3,2‐c]chromen‐4‐ones 3a–e in 36–86% yields by the method B. Under the same conditions, the reactions of 2‐hydroxy‐1,4‐naphtaquinone 1b with conjugated alkenes 2b and 2f–i afforded 2,3‐dihydronaphtho[2,3‐b]furan‐4,9‐diones 3f–j in an excellent yields.     

One-Pot Etherification of Ketones and Aldehydes with Organic Halides Using Sodium Hydride as a Reductant

One-pot etherification reaction of aromatic and some aliphatic carbonyl compounds with organic halides in the presence of sodium hydride as a reducing reagent proceeded smoothly in dioxane, a polar solvent with higher boiling point, to provide desired ethers in moderate to high yields.     

One‐Pot Synthesis of 2‐Arylthio‐2‐cyclohexenone Derivatives by the Diels–Alder Reaction of 4‐Arylthio‐3‐hydroxy‐2‐pyrones

The base‐catalyzed Diels–Alder reactions of 4‐arylthio‐3‐hydroxy‐2‐pyrones are reported. Treatment of 4‐arylthio‐3‐hydroxy‐2‐pyrones and dienophiles with triethylamine gave 2‐arylthio‐2‐cyclohexenone derivatives by the Diels–Alder reaction involving a decarboxylation in excellent to reasonable yields.
