Developing Bright Green Fluorescent Protein (GFP)-like Fluorogens for Live-Cell Imaging with Nonpolar Protein−Chromophore Interactions

Fluorescence-activating proteins (FAPs) that bind a chromophore and activate its fluorescence have gained popularity in bioimaging. The fluorescence-activating and absorption-shifting tag (FAST) is a light-weight FAP that enables fast reversible fluorogen binding, thus advancing multiplex and super-resolution imaging. However, the rational design of FAST-specific fluorogens with large fluorescence enhancement (FE) remains challenging. Herein, a new fluorogen directly engineered from green fluorescent protein (GFP) chromophore by a unique double-donor-one-acceptor strategy, which exhibits an over 550-fold FE upon FAST binding and a high extinction coefficient of approximately 100,000 M⁻¹ cm⁻¹, is reported. Correlation analysis of the excited state nonradiative decay rates and environmental factors reveal that the large FE is caused by nonpolar protein−fluorogen interactions. Our deep insights into structure-function relationships could guide the rational design of bright fluorogens for live-cell imaging with extended spectral properties such as redder emissions.     

Numerical Solution for Linear-Quadratic Control Problems of Markov Jump Linear Systems and Weak Detectability Concept

A method for solving the linear-quadratic problem of Markov jump linear systems is developed in this paper, relying on the assumption of weak detectability. The concept of weak detectability generalizes previous concepts relevant to this class of systems, and most importantly, it allows us to revisit the quadratic control problem. In the main result of the paper, we show that, for weakly detectable systems, the solution obtained with the new method converges to the solution of the coupled algebraic Riccati equation that arises in the control problem if and only if the system is mean-square stabilizable. The paper shows how the concepts and the method involved are applied by means of numerical examples and comparisons.

Digital PID controller design for multivariable analogue systems with computational input-delay

This paper presents a new methodology to design MIMO digitalPID controllers for multivariable analogue systems with computationalinput time-delay. The preliminarily designed analogue PID controlleris refined using a newly developed state-feedback and state-feedforwardLQR approach. The optimally designed closed-loop system withthe refined MIMO analogue PID controller has pre-assigned closed-loopeigenvalues. A prediction-based digital redesign technique isdeveloped to discretize the cascaded MIMO analogue PID controller,such that the states of the digitally redesigned closed-loopsampled-data system with the MIMO digital PID controller areclose to those of the analogously designed closed-loop systemwith the refined MIMO analogue PID controller. The aforementioneddigital redesign technique is further modified based on thepredictive control method to cope with MIMO analogue systemswith input delay.

多指标面板数据的聚类分析及其应用

多指标面板数据的多元统计分析在国内研究中尚属空白.本文分析了面板数据的数据格式和数字特征,根据聚类分析原理,重新构造了多指标面板数据的距离函数和离差平方和函数,在此基础上,说明了多指标面板数据的聚类分析过程.最后对我国各地区工业企业生产效率进行了聚类实证分析,显示了良好的效果。     

温度影响下基于应力波的钛合金试件疲劳裂纹监测实验研究

本文对温度影响下应用应力波监测钛合金CT试件中的疲劳裂纹进行了研究。建立了由MTS材料试验机和温控箱组成的实验装置,模拟温度变化对应力波在钛合金试件中传播的影响。采用PZT压电元件分别在试件中激励和接收应力波信号。通过连续小波分析,提取应力波信号的多种特征用于表征疲劳裂纹的发生与发展,并对信号特征进行了统计分析。为在温度变化下监测钛合金试件中的疲劳裂纹,对信号特征进行多元偏值分析,采用马氏平方距离融合多个信号特征,在概率统计意义下确定钛合金试件中是否存在疲劳裂纹。实验结果表明了所提出方法的可行性和有效性。     

On a multivariable extension of the Humbert polynomials

In this paper, we present a multivariable extension of the Humbert polynomials, which is motivated by the Chan-Chyan-Srivastava multivariable polynomials, the multivariable extension of the familiar Lagrange-Hermite polynomials and Erkus-Srivastava multivariable polynomials. We derive various families of multilinear and mixed multilateral generating functions for these polynomials. Other miscellaneous properties of these multivariable polynomials are also discussed. Some special cases of the results presented in this study are also indicated.
