Mass determination by mass spectrometric methods (electrospray ionization mass spectrometry (ESI-MS), matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS)) of sodiumdodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE)-separated proteins is a well known procedure and reliable protocols are available. In our efforts to use the established methods to determine the molecular mass of the disulfide bridged, heterodimeric glycoprotein GP3 and to determine the carbohydrate content of each protein subunit we developed an in-gel chemical deglycosylation method. For this purpose we established experimental conditions that allow maximum extraction of the high molecular mass protein subunits and developed a routine method to apply the HF-pyridine deglycosylation protocol to proteins isolated from polyacrylamide gel pieces. The novel protocol and extraction procedure described can be used to analyze O-glycosylated proteins up to 150 kDa after SDS-PAGE separation.  相似文献

Methods derived from topology and graph theory indicate that the deltahedral boranes B n H n 2– and the corresponding carboranes C2Bn–2H n (6 n 12) may be regarded as three-dimensional delocalized aromatic systems in which surface bonding and core bonding correspond to -bonding and -bonding, respectively, in planar polygonal two-dimensional hydrocarbons CinnH n (n–6)+ (n=5/7). The two extreme types of topologies which may be used to model core bonding in deltahedral boranes and carboranes are the deltahedral (D n ) topology based on the skeleton of the underlying deltahedron and the complete (K n ) topology based on the corresponding complete graph. Analyses of the Hoffmann-Lipscomb LCAO extended Hückel computations, the Armstrong—Perkins—Stewart self-consistent molecular orbital computations, and SCF MOab initio GAUSSIAN-82 computations on B6H6 2– indicate that the approximation of the atomic orbitals by the sum of the molecular orbitals, as is typical in modernab initio computations, leads to significantly weaker apparent core bonding approximated more closely by deltahedral (D n ) topology than by complete (K n ) topology.This work was presented at the Workshop The Modern Problems of Heteroorganic Chemistry sponsored by the A. The analysis of the effect of the solvents on the proton chemical shifts in1H NMR spectra of 2-vinyloxypyridine indicates that the C—H...N interaction of weak intramolecular hydrogen bond type hinders the formation of intermolecular hydrogen C—H...X and C-H... bonds. The thermal decomposition of sodium nitrite or nitrate pre-adsorbed upon TiO2 surfaces has been investigated by employing several techniques as infrared spectroscopy (IR) and temperature programmed desorption in conjunction with mass spectrometry analysis (TPD-MS) to study the features observed during these thermal decompositions. Differential thermal analysis (DTA) in combination with X-ray diffraction analysis (XRD) were used to investigate the possibility of a solid state chemical reaction between the solid products originated from the thermal decomposition of the pre-adsorbed species and the TiO2. A substance on which X-rays fall emits photoelectrons and Auger electrons. The energy spectra of the electrons emitted provide information about the electronic structure in the specimen, ranging from the innermost atomic levels and their dependence on the chemical environment to the molecular orbitals of the valence electrons and the band structure in solids. Electron spectra of this nature can now be recorded with high-resolution instruments; their analysis offers new aspects for investigation of chemical composition. The method of electron spectroscopy developed for this purpose, which has now been developed to a high degree of perfection, will be referred to in the following discussion as ESCA (Electron Spectroscopy for Chemical Analysis).  相似文献

XPS was used to characterize the chemical changes occurring after drying or applying a heat‐treatment to beech wood samples. Our results indicate that the surface of this air‐exposed material could be strongly affected either by the ambient atmosphere during storage or by the complex atmosphere in the oven during drying or heat‐treatment. However, the O/C ratio measured after removal of a thin slice of a few millimetres of an untreated sample is in reasonable agreement with that calculated from the well‐established chemical composition of beech. Through this methodology (equivalent to scraping for hard materials) it is expected to get a realistic characterization of the wood. The reliability and repeatability of the XPS measurements have been checked and the method applied to the study of the chemical changes of the beech samples subjected to heat‐treatment. Heating at 240 °C induces a significant decrease of the O/C ratio from 0.55 before to 0.44 after the treatment. Heat‐treatment induces also a decrease of the C2 carbon contribution (carbon atom bound to a single non‐carbonyl oxygen) associated with an increase of the C1 carbon contribution (carbon atoms bound only to carbon or hydrogen atoms), in agreement with chemical modifications reported previously in the literature. Thanks to the small analysed area of the equipment used in this study, different spots were analysed to demonstrate the presence or absence of a gradient of chemical composition due to thermal degradation or migration of extractives from within the wood structure to its surface. Argania spinosa L. Skeels is an emblematic tree in Morocco, known worldwide for its medicinal and nutritional value. Its fruits contain kernels used to prepare an edible oil, the leaves are used to feed livestock, and its wood is used as fuel. If the oil acquires high importance, the other components of the fruit of the argan are undervalued. Our objective is to invest the waste of the argan industry. Particularly, our study aimed to assess the effect of thermal activation of argan pulp on its therapeutic value, its phenolic profile and its functional and physicochemical properties. After heat treatment, the HPLC analysis for the average total phenolic content varied from 2% to 37%, depending on temperature. The antioxidant activity was increased with heat treatment. Higher values of antioxidant activity, polyphenol and pigment content were recorded at 70 °C. Functional properties analysis indicated that water solubility index and water absorption capacity were significantly affected by heat stress. Physicochemical analysis showed that moisture content, titratable acidity and soluble solids were affected.  相似文献

微电子塑料封装中常用的聚合物材料因易于吸收周围环境中的湿气而对器件本身的可靠性带 来很大影响, 本文回顾了封装材料中湿气扩散、湿应力及蒸汽压力的产生这3个互相联系过 程的研究情况, 从理论分析、特征参数描述及其实验测定、有限元模拟分析的角度来分别予 以介绍.从已有的理论分析与实验结果中可以看出, 塑封材料吸收湿气会给器件的可靠性带 来诸多影响, 湿气的吸收、扩散、蒸发等过程, 实验测量, 以及由湿气带来的其它相关问题正 成为微电子封装可靠性研究领域中的新热点, 受到越来越多的关注与重视.  相似文献

This work aimed to evaluate the effect of partial replacement of semolina with 0, 1, 5, 10, 15, and 20% of ground buckwheat hull (BH) on the chemical composition, antioxidant properties, color, cooking characteristics, and sensory properties of wheat pasta. Pasta samples were prepared by dough lamination (tagliatelle shape) and dried at 55 °C until the moisture content was 11–12% (wet basis). Analyses of samples showed that the addition of BH caused an increase in fiber content in pasta from 4.31% (control pasta) to 14.15% (pasta with 20% of BH). Moreover, the brightness and yellowness of BH-enriched products were significantly decreased compared to the control sample, and the total color difference ranged from 23.84 (pasta with 1% of BH) to 32.56 (pasta with 15% BH). In addition, a decrease in optimal cooking time, as well as an increased weight index and cooking loss, was observed in BH-enriched pasta samples. Furthermore, BH-enriched cooked pasta had significantly higher total phenolic content and antioxidant activity but an unpleasant smell and taste, especially if the level of BH was higher than 10%.  相似文献
