An HPLC assay for hydroxyl radicals is described. The hydroxyl radical was trapped by terephthalic acid (non-fluorescent), and 2-hydroxyl terephthalic acid (fluorescent) was quantitated by HPLC-fluorescence detection. At a terephthalic acid concentration of 4.25 mmol/L, the hydroxyl radical formed in the Fenton reaction was successfully assayed in the concentration range of hydrogen peroxide of 2.5-50 micro mol/L, where the concentration of Fe(II) was 50 micro mol/L. The fluorescence of 2-hydroxy terephthalate was stable at 24 h, and its detection limit by this method was 5 nmol/L (100 fmol).  相似文献

The attractive features of a boron-doped diamond (BDD) thin-film detector for microchip capillary electrophoretic (CE) separations of dye-related amino-substituted aromatic compounds are described. The diamond electrode was employed in the end-column amperometric detection of 4-aminophenol (4-AP), 1,2-phenylenediamine (1,2-PDA), 2-aminonaphthalene (2-AN), 2-chloroaniline (2-CA), and o-aminobenzoic acid (o-ABA), and its attractive behavior was compared to commonly used screen-printed carbon and glassy-carbon electrodes. These conventional electrode materials exhibit a significant degree of passivation and low sensitivity to the above-mentioned environmental pollutants. The diamond-based electrochemical detection system displayed a favorable analytical performance, including lower noise levels, higher peak resolution with enhanced sensitivity, and improved resistance against electrode passivation. Factors influencing the on-chip analysis were assessed and optimized. The diamond detector displayed detection limits of 2.0 and 1.3 microM for 4-AP and 2-AN, respectively, and a wide linear response for these compounds over the 2-50 microM range. The enhanced stability was demonstrated by relative standard deviation (RSD) values of 1.4% and 4.7% for 100 microM 1,2-PDA and 200 microM 2-CA, respectively, for repetitive detections (n = 7). Besides, the simultaneously observed current decrease was 2.4 and 9.1% for 1,2-PDA and 2-CA, respectively (compared to 21.8 and 41.0% at the screen-printed carbon electrode and 28.3 and 34.1% at the glassy carbon electrode, respectively). The favorable properties of the diamond electrode indicate great promise for environmental applications in CE and other microchip devices.  相似文献

Separation and detection of proteins have been realized on nonionic surfactant-modified poly(dimethylsiloxane) (PDMS) microfabricated devices with end-column amperometric detection. The hydrophobic PDMS channels are turned into hydrophilic ones after being modified with Brij35 and facilitate the separation of proteins. The coating can remarkably reduce the adsorption of large protein molecules and is stable in the range of pH 6-12. The detection of proteins in such channels needs less rinsing time and thus efficiency is raised. Even large molecules of proteins can also be detected with better reproducibility and enhanced plate numbers. The relative standard deviation (RSD) of the migration time for glucose oxidase (GOD) is 2.2% (n = 19). Separation of GOD and myoglobin has been developed in modified channels. Predominant operational variables, such as the coating conditions, the concentration of surfactant and buffer, are studied in detail.  相似文献

We report the design and validation of a fast empirical function for scoring RNA-ligand interactions, and describe its implementation within RiboDock, a virtual screening system for automated flexible docking. Building on well-known protein-ligand scoring function foundations, features were added to describe the interactions of common RNA-binding functional groups that were not handled adequately by conventional terms, to disfavour non-complementary polar contacts, and to control non-specific charged interactions. The results of validation experiments against known structures of RNA-ligand complexes compare favourably with previously reported methods. Binding modes were well predicted in most cases and good discrimination was achieved between native and non-native ligands for each binding site, and between native and non-native binding sites for each ligand. Further evidence of the ability of the method to identify true RNA binders is provided by compound selection ('enrichment factor') experiments based around a series of HIV-1 TAR RNA-binding ligands. Significant enrichment in true binders was achieved amongst high scoring docking hits, even when selection was from a library of structurally related, positively charged molecules. Coupled with a semi-automated cavity detection algorithm for identification of putative ligand binding sites, also described here, the method is suitable for the screening of very large databases of molecules against RNA and RNA-protein interfaces, such as those presented by the bacterial ribosome.  相似文献

A simple, rapid and sensitive high-performance liquid chromatographic method with fluorescence detection for the simultaneous determination of oxytetracycline, doxycycline, tetracycline and chlortetracycline was developed, and successfully applied to the analysis of commercial tetracycline antibiotics. The separation was performed on a reverse-phase C18 column with a gradient elution composed of methanol and sodium acetate buffer (containing disodium ethylenediaminetetraacetate and calcium chloride, pH 8.10) as the mobile phase, and fluorescence detection at 532 nm (excitation at 380 nm). The detection limits for oxytetracycline, doxycycline, tetracycline and chlortetracycline were 0.1, 0.5, 0.3 and 0.4 g L–1, respectively. Data with respect to precision and accuracy were reported and discussed.  相似文献

A portable chip-CE system with potential gradient detection (PGD) was developed and applied to the determinations of alkali metals and alkaloids. The separation efficiency appeared to be satisfactory and nonaqueous capillary electrophoresis (NACE) proved to be applicable to PGD or conductivity detection. The power supplies, separation and detection were built on a device of 3 kg in weight. A branch channel near the end of the separation channel was designed to perform PGD and make the application of relatively high field strength possible. The study is the first report on the application of PGD on the microchip platform. The design of the chip-CE system shows several advantages, such as simplicity, miniaturization and wide applicability.  相似文献

Carnosine (beta-alanyl-L-histidine) is a dipeptide regarded as an important molecular marker of the presence of processed animal proteins including meat and bone meal in animal feed. For its identification and quantification a sensitive and selective method by high-performance anion-exchange chromatography coupled with integrated pulsed amperometric detection (HPAEC-IPAD) was developed. The assay is based on isocratic elution with 100 mM NaOH as the mobile phase. Interferences of real matrices were efficiently removed; carnosine could be determined at the concentration ranges 0.1-100 microM with a rather low detection limit of 0.23 ng. Unlike feeds for dogs and cats, no carnosine peak was observed in all examined feeds for ruminants. The good analytical characteristics allowed camosine determination down to 5 microg/g of feed.  相似文献

The use of composite biosensors for multianalyte detection strategies is discussed. Graphite–Teflon rigid composite biosensors offer the possibility of coimmobilization of several enzymes by simple physical inclusion in the bulk of the electrode matrix with no covalent linkages. A novel trienzyme graphite–Teflon–glucose oxidase (GOD)–alcohol oxidase (AOD)–peroxidase (HRP)–ferrocene bisosensor yielded amperometric steady-state currents similar to those obtained with graphite–Teflon–GOD–HRP–ferrocene and graphite–Teflon–AOD–HRP–ferrocene electrodes for the same concentration of glucose and ethanol, respectively. The performance of the trienzyme biosensor for multianalyte detection was evaluated with the simultaneous determination of glucose and ethanol after separation by HPLC, in samples of sweet wine. The simultaneous analysis of several analytes in the same sample should imply that, with an adequate dilution, the concentration levels of the analytes can be included within the ranges of linearity of the corresponding calibration plots. The use of two composite biosensors in a parallel configuration, so that different analytes can be simultaneously detected with no need of chromatographic separation, is also discussed. The usefulness of this approach was evaluated by the simultaneous analysis of glucose and ethanol in sweet wine, and of glucose and lactic acid in red wine.  相似文献

An accurate and precise high-performance liquid chromatographic method using diode array detection for the determination of the novel antiepileptic, Levetiracetam, has been developed. Three clean-up procedures for the analysis of Levetiracetam in human plasma were implemented and evaluated, namely solid-phase extraction, deproteinization by addition of organic solvents and formation of insoluble salts. Adenosine was used as the internal standard for all three sample pretreatment procedures. Among the several cartridges used for solid-phase extraction, the hydrophilic-lypophilic balance (Oasis) HLB) phase provides the best extraction yield of Levetiracetam, together with high precision. With the two other clean-up procedures involving plasma deproteinization by addition of methanol or zinc sulphate, lower sensitivity and precision of the assays were obtained. However, they are cheaper and faster when compared with the solid-phase extraction procedure.  相似文献

Based on an official standard method of lutein analysis, an improved high performance liquid chromatography (HPLC) method for simultaneously detecting lutein and zeaxanthin was developed as focusing on the sample preparation protocol. The optimal pretreatment conditions included a saponification in a water bath for 15?min at a constant temperature of 50?°C, using a 10?mL 60% (w/v) potassium hydroxide solution, followed by extraction using 100?mL mixture of n-hexane, ethyl ether and cyclohexane (40: 40: 20, v/v/v). A mixture of dichloromethane, acetonitrile and methanol (20: 30: 50, v/v/v) was validated to elute lutein and zeaxanthin on a C30 column (4.6?×?250?mm, 5?µm). The resolution between lutein and zeaxanthin is ≥2.5. A millet sample was used for methodological verification and the results showed that the linear relations for lutein and zeaxanthin were good in ranges of 0.23–9.37?μg/mL and 0.30–12.02?μg/mL, respectively. The relative standard deviations of lutein and zeaxanthin were 1.40% and 5.09%, respectively, and their spiked recoveries were between 86.60% and 98.75%. The lutein and zeaxanthin results from this modified HPLC method are superior to those from the Chinese official method and ultrasonic extraction method.  相似文献
