Adrenocortical carcinoma (ACC) is a rare tumor and prognosis is overall poor but heterogeneous. Mitotane (MT) has been used for treatment of ACC for decades, either alone or in combination with cytotoxic chemotherapy. Even at doses up to 6 g per day, more than half of the patients do not achieve targeted plasma concentration (14–20 mg L?1) even after many months of treatment due to low water solubility, bioavailability, and unfavorable pharmacokinetic profile. Here a novel MT nanoformulation with very high MT concentrations in physiological aqueous media is reported. The MT‐loaded nanoformulations are characterized by Fourier transform infrared spectroscopy, differential scanning calorimetry, and powder X‐ray diffraction which confirms the amorphous nature of the drug. The polymer itself does not show any cytotoxicity in adrenal and liver cell lines. By using the ACC model cell line NCI‐H295 both in monolayers and tumor cell spheroids, micellar MT is demonstrated to exhibit comparable efficacy to its ethanol solution. It is postulated that this formulation will be suitable for i.v. application and rapid attainment of therapeutic plasma concentrations. In conclusion, the micellar formulation is considered a promising tool to alleviate major drawbacks of current MT treatment while retaining bioactivity toward ACC in vitro. 相似文献

The phase behaviors in a binary mixture of diblock copolymers confined between two parallel walls are investigated by using a cell dynamics simulation of the time-dependent Ginzburg-Landau theory.The morphological dependence of the wall-block interaction and the distance between walls(confinement degree) has been systematically studied,and the effect of repulsive interactions between different monomers is also discussed.It is interesting that multiple novel morphological transitions are observed by changing these factors,and various multilayered sandwich structures are formed in the mixture.Furthermore,the parametric dependence and physical reasons for the microdomain growth and orientational order transitions are discussed.From the simulation,we find that much richer morphologies can form in a binary mixture of diblock copolymers than those in a pure diblock copolymer.Our results provide an insight into the phase behaviors under parallel wall confinement and may provide guidance for experimentalists.This model system can also give a simple way to realize orientational order transition in soft materials through confinement. 相似文献

Poly (methyl methacrylate/butyl acrylate) [P(MMA/BA)] copolymers (M η~2×105) with different mass percentages of PMMA (100/0, 90/10, 81/19, and 75/25), were synthesized by the method of solution polymerization. In addition to the normal α and ρ peak, a third τ peak is observed in thermally stimulated depolarization current (TSDC) spectra of the copolymers in the high temperature region. The α peak‐corresponds to the glass transition, the ρ peak originates from the detrapping of trapped carriers in the bulk amorphous structure related with flexible side groups, and the τ peak can be attributed to the charge detrapping related to the liquid–liquid transition of the copolymers. The three peaks all move to lower temperature with an increase of the BA component, indicating that the flexible side groups of butyl acrylate not only have an effect of plasticization on the glass transition and liquid–liquid transition, but also make the trap depth shallower and the detrapping process easier for the ρ and τ peaks. The experimental results confirm that TSDC analysis is very sensitive for investigating the liquid–liquid transition of polymers. The liquid–liquid transition temperature (T LL) of the copolymers follows a type of the Fox equation. Fitting the results gives a T LL of 102°C for polybutyl acrylate. 相似文献

The self-assembly of diblock copolymers confined around one square-shaped particle is studied systematically within two-dimensional self-consistent field theory(SCFT).In this model,we assume that the thin block copolymer film is confined in the vicinity of a square-shaped particle by a homopolymer melt,which is equivalent to the poor solvents.Multiple sequences of square-shaped particle-induced copolymer aggregates with different shapes and self-assembled internal morphologies are predicted as functions of the particle size,the structural portion of the copolymer,and the volume fraction of the copolymer.A rich variety of aggregates are found with complex internal self-assembled morphologies including complex structures of the vesicle,with one or several inverted micelle surrounded by the outer monolayer with the particle confined in the core.These results demonstrate that the assemblies of diblock copolymers formed around the square-shaped particle in poor solvents are of immediate interest to the assembly of copolymer and the morphology of biomembrane in the confined environment,as well as to the transitions of vesicles to micelles. 相似文献

Block copolymers are a class of soft matter that self-assemble to form ordered morphologies on the scale of nanometers, making them ideal materials for various applications. These applications directly depend on the shape and size of the self-assembled morphologies, and hence, a high degree of control over the self-assembly is desired. Grafting block copolymer chains onto a substrate to form copolymer brushes is a versatile method to fabricate functional surfaces. Such surfaces demonstrate a response to their environment, i.e., they change their surface topography in response to different external conditions. Furthermore, such surfaces may possess nanoscale patterns, which are important for some applications; however, such patterns may not form with spun-cast films under the same condition. In this review, we summarize the recent progress of the self-assembly of block copolymers grafted onto a flat substrate. We mainly concentrate on the self-assembled morphologies of end-grafted AB diblock copolymers, junction point-grafted AB diblock copolymers(i.e.,Y-shaped brushes), and end-grafted ABA triblock copolymers. Solid polymer electrolytes are attractive materials for use as battery separators. Here, a molecular weight series of polystyrene–polyethylene oxide (PEO) multiblock copolymers was synthesized by the thiol–norbornene click reaction. The subsequent materials were characterized both neat and with a lithium bis‐(trifluoromethane)sulfonimide salt loading [(Li)/(EO)] of 0.1. In general, neat samples demonstrated crystallinity scaling with PEO content. Lithium ion‐containing samples had broad scattering peaks, half of which displayed disordered scattering, even at the lowest block molecular weights (polystyrene = 1 kg/mol, PEO = 1 kg/mol). Fitting of disordered scattering data, using the random phase approximation, yielded χRPA and Rg values that were compared with recent predictive work by Balsara and coworkers. The predictions were accurate near the volume fraction fPEO = 0.5 but deviated symmetrically with volume fraction asymmetry. Samples were also analyzed by electrochemical impedance spectroscopy for their potential to conduct lithium ions. Samples with fPEO ≥ 0.5 demonstrated robust conductivity, whereas samples below this volume fraction conducted very poorly, with one exception (fPEO = 0.24). This work expanded upon our recently reported approach to multiblock copolymer synthesis, demonstrating the improved access of materials to further our fundamental understanding of multiblock copolymers. Using cyclic voltammetry method, the reduction of cationic η6-fluorene complexes of manganese [(η6-9-R-C13H9)Mn(CO)2L]PF6 (L = CO, R = H (1 +); L = CO, R = CH3 (2 +); L = PnBu3, R = H (3 +); L = CO, R = tBu (4 +)) is studied. It is shown that, depending on the nature of a substituent in the position 9 of the fluorene ligand, the reduction occurs either with the detachment of an H atom from position 9 to give zwitterion compounds (complexes 1 ±, 2 ±, 3 ±) or with the attachment of an H atom into the coordinated ring of the fluorene ligand to given η5-cyclohexadienyl complex (η5-9-tBu-C13H9)Mn(CO)3 (5). 相似文献

Copolymers of ethyl cellulose (EC) with polystyrene (PSt) were synthesized through atom transfer radical polymerization (ATRP). The molecular weight of graft copolymers increased without any trace of the EC macro-initiator, and the polydispersity of the side chains was low. The molecular weight of the side chains increased with the monomer conversion. Kinetic study indicated that the polymerization was first order. The micelle characteristics of the graft copolymer in acetone were investigated using dynamic light scattering (DLS), atom force microscopy (AFM) and transmission electron microscopy (TEM). With increasing the concentration, micelles were gradually formed from the solution. The TEM and AFM images indicated that the micelles had spherical shape and showed core-shell structure. 相似文献
