The 5′-GCGTGGGCGT-3′ (T4) oligomer found in the zinc finger 268–DNA complex was mutated into the sequence 5′-GCGGGGGCGT-3′
(G4). A 3D model was constructed from the T4 sequence using an X-ray structure as a template. Molecular dynamics simulations
were used to test the thermal stability of the model. A 500-ps trajectory was obtained for the fully charged complex in water
using GROMOS87. The complex and the G4 sequence are found to have dynamically stationary behavior. Comparisons made with a
previous T4 sequence molecular dynamics simulation show both systems have similar thermal stability. The structure of DNA
appears to be maintained by its global interactions with the protein although the mutated site does not contribute with its
full potential for binding. The protein structure shows some small differences compared to the T4 simulation. The simulation
provided evidence for the role of a chloride ion interacting with the protein and helping in the recognition process.

Finger vein recognition has been adopted due to its high recognition rate and the invisibility of vein in visible light. However, because a finger vein pattern is not distinctive due to light scattering in the skin layer, the localization of a finger vein pattern region using an image processing algorithm is a difficult procedure. Also, optical blurring increases these difficulties. We propose a new finger vein image restoration method to deal with skin scattering and optical blurring. Our research is novel in three ways compared to previous studies. Firstly, the amount of optical blurring of a finger vein image is measured based on the average gradient of the orthogonal profile of a finger edge. Secondly, the accurate point spread function (PSF) of optical blurring is adaptively determined based on the orthogonal profile of a finger edge. Thirdly, using a constrained least square (CLS) filter and optimized parameters in terms of the lowest error of finger vein recognition, the restoration of a skin scattered and optically blurred finger vein pattern was performed. Experimental results show that the equal error rate (EER) of finger vein recognition with restoration was reduced by as much as 3.14% (5.05-1.91%) compared to the EER without restoration. 相似文献

Despite decades of investigations, it is not yet clear whether there are rules dictating the specificity of the interaction between amino acids and nucleotide bases. This issue was addressed by determining, in a dataset consisting of 100 high-resolution protein-DNA structures, the frequency and energy of interaction between each amino acid and base, and the energetics of water-mediated interactions. The analysis was carried out using HINT, a non-Newtonian force field encoding both enthalpic and entropic contributions, and Rank, a geometry-based tool for evaluating hydrogen bond interactions. A frequency- and energy-based preferential interaction of Arg and Lys with G, Asp and Glu with C, and Asn and Gln with A was found. Not only favorable, but also unfavorable contacts were found to be conserved. Water-mediated interactions strongly increase the probability of Thr-A, Lys-A, and Lys-C contacts. The frequency, interaction energy, and water enhancement factors associated with each amino acid-base pair were used to predict the base triplet recognized by the helix motif in 45 zinc fingers, which represents an ideal case study for the analysis of one-to-one amino acid-base pair contacts. The model correctly predicted 70.4% of 135 amino acid-base pairs, and, by weighting the energetic relevance of each amino acid-base pair to the overall recognition energy, it yielded a prediction rate of 89.7%. 相似文献

In this study, we fabricated non‐woven matrices using blends of polycaprolactone and gelatin with various spinning volumes to control the immobilized heparin content, which was ultimately intended to increase the immobilization efficiency of bFGF. The amount of bFGF on the heparin conjugated fibrous matrices depended on the thicknesses of the swollen matrices ranging from 35.4 ± 6.5 to 162.3 ± 14.0 ng and ≈90% of the bFGF was gradually released over a period of up to 56 d. The released bFGF enhanced the proliferation of human umbilical vein endothelial cells and human mesenchymal stem cells. In conclusion, our heparin‐conjugated fibrous matrices have the potential to be used as a growth factor delivery system in tissue engineering applications.
