Unraveling of all of the information contained in proteomes poses a tremendous chemical challenge, which is balanced by the promise of potentially transformational knowledge. Mass spectrometry offers an unprecedented arsenal of tools for diverse proteomic investigations. Recently, it was demonstrated that ultraviolet light can be utilized to initiate unique and potentially useful fragmentations in peptides and proteins. Either nonspecific dissociation or highly specific dissociation at engineered chromophoric sites is possible following photon absorption. The level of specificity and control over fragmentation in these experiments is greater than with other fragmentation methods. Novel techniques made possible by this technology are poised to make substantial contributions to the field of proteomics.  相似文献

The Raman spectrum of any molecule consists of two mirror-image signals, th e Stokes and anti-Stokes Raman spectra. In most cases, unless highly specific sampling conditions are used, the anti-Stokes signal is much weaker than that of the Stokes. The recent application of intensified diode array detectors to Raman spectroscopy has produced a marked increase in the sensitivity of the technique which makes a study of the anti-Stokes spectrum potentially more rewarding than it has been to date. The present study has shown that, although of limited use for general purposes, there are some specific instances where the anti-Stokes spectrum can be of considerable practical use. Such applications are to extend the operating range of the Raman spectrometer, to study photodegradable samples and for the analysis of samples which exhibit strong fluorescence.  相似文献

The eigen-oscillation mode of the Nd:YAP cw laser has been analysed. The influence of thermal effects arising from the pumping process on the output character of the 1.3414 μm Nd:YAP cw laser has been discussed. The crack problem of the reflective film at a 1.34 μm dielectric mirror has been solved. Based on the aforementioned work, we have been able to achieve an 82.8 W laser output at 1.3414 μm with a nearly polarized beam from a 5.8 mm diameter by 111 mm Nd:YAP rod. The overall efficiency and the slope efficiency are 1.15% and 2.02% respectively, and the fluctuation of the output power at 62 W is less than 1% during continuous operation for 45 min.  相似文献

With a high uniformity of x-ray preionization, a long pulse duration and a high specific energy (9 J l-1), an XeCl excimer laser output has been obtained by using a compact low inductance capacitor bank as the laser excitation circuit. We present here a method for selecting optical patterns in a passive semiconductor microresonator, by using a spatial perturbation. A pattern is spontaneously generated in the system, and a switching beam causes this pattern to rotate even if the power in the switching beam is much lower than the power in the pattern. Birefringent filters can be used for tuning lasers, with some advantages over prisms, gratings and tilted etalons. In this paper the transmission characteristics of a new mixed birefringent filter are investigated. The term ‘mixed’ is used because both circular birefringence and linear birefringence have been utilized in the fabrication of this filter.It has a loss-less core, formed by a cascaded system of alternate rotators and retarders, and a linear polarizer at each end. The performance of this filter is better because of the more effective suppression of side peaks. Unlike previous birefringent filters, this one may be used inside a linear resonator.  相似文献
