We prepared hybrid particle-monolithic polymethacrylate columns for micro-HPLC by in situ polymerization in fused silica capillaries pre-packed with 3–5 μm C18 and aminopropyl silica bonded particles, using polymerization mixtures based on laurylmethacrylate–ethylene dimethacrylate (co)polymers for the reversed-phase (RP) mode and [2-(methacryloyloxy)ethyl]-dimethyl-(3-sulfopropyl) zwitterionic (co)polymers for the hydrophilic interaction (HILIC) mode. The hybrid particle-monolithic columns showed reduced porosity and hold-up volumes, approximately 2–2.5 times lower in comparison to the pure monolithic columns prepared in the whole volume of empty capillaries. The elution volumes of sample compounds are also generally lower in comparison to packed or pure monolithic columns. The efficiency and permeability of the hybrid columns are intermediate in between the properties of the reference pure monolithic and particle-packed columns. The chemistries of the embedded solid particles and of the interparticle monolithic moiety in the hybrid capillary columns contribute to the retention to various degrees, affecting the selectivity of separation. Some hybrid columns provided improved separations of proteins in comparison to the reference particle-packed columns in the reversed-phase mode. Zwitterionic hybrid particle-monolithic columns show dual mode retention HILIC/RP behaviour depending on the composition of the mobile phase and allow separations of polar compounds such as phenolic acids in the HILIC mode at lower concentrations of acetonitrile and, often in shorter analysis time in comparison to particle-packed and full-volume monolithic columns.  相似文献

It is believed that the enantiorecognition mechanism based on macrocyclic antibiotics involves multimodal interactions via hydrogen bonding, π–π interaction, steric hindrance, hydrophobic interaction and so on. A variety of enantiomeric N-benzoylated amino acids were separated using balhimycin (A) or its analogues bromobalhimycin (B) and dechlorobalhimycin (C) as chiral mobile phase additive using a CE method, which combined the partial filling technique with the dynamic coating technique and the co-EOF electrophoresis technique. The enantioresolution and the migration time were highly relevant to the structure of analytes, especially to the substitutions on the N-tagged benzoyl moiety of the amino acids. A steric effect and π–π interaction based mechanism is proposed in order to explain some observed enantioresolution differences between positional isomers. Notably dechlorobalhimycin exhibited the best enantioresolution for several N-benzoylated derivatives of leucine, which was rarely observed for N-dansylated amino acid derivatives. The hydrophobicity difference of the aglycone pocket among three chiral selectors was assumed to account for this behaviour.  相似文献

建立了固相萃取-离子色谱(IC)/气相色谱-质谱(GC-MS)联合检测高矿化度油田水中低相对分子质量的有机酸和酚类化合物的方法。在中性pH条件下,样品经Waters Oasis HLB柱萃取后,萃取液经稀释、Ag2O沉淀和Ag-H柱处理除去大部分氯离子,再用IC测定有机酸;将萃取柱真空冷冻干燥,然后经甲基叔丁基醚/甲醇(9:1, v/v)脱附并用无水硫酸钠除水,再用GC-MS检测酚类化合物。在优化的实验条件下,4种低相对分子质量的有机酸以及5种酚类化合物的平均加标回收率达到80%以上,相对标准偏差(RSD, n=6)为2.38%~9.45%,定量限均低于88.9 μg/L。该方法测定结果准确可靠,适用于氯离子含量高达150 g/L左右水样中低相对分子质量的有机酸和酚类化合物的检测。  相似文献

Apparent molar adiabatic compressibilities (K?, s) of glycine, L-alanine, L-valine, and L-leucine have been determined in aqueous and mixed aqueous solutions of lactose (2 to 6 mass%) at T = (293.15, 298.15, 303.15, and 308.15) K. From these data partial molar adiabatic compressibilities at infinite dilution (K?, s0) have been evaluated to calculate corresponding transfer function. The transfer partial molar adiabatic compressibilities at infinite dilution (ΔK?, s0) are found to be positive. The decrease in the magnitude of transfer partial molar adiabatic compressibilities from glycine to L-leucine indicates the dominance of hydrophobic-hydrophobic interactions between the increasing side chains of amino acids. Also, the contributions of NH3+COO , and CH2 groups have been calculated by the linear correlation of K?, s0 with number of carbon atoms in the alkyl chain of amino acids.  相似文献

Densities and viscosities of glycine and l-valine have been measured at 308.15 and 318.15 K in aqueous tripotassium citrate solutions ranging from 0.2 to 0.8 mol kg− 1 of tripotassium citrate. The viscosity data have been analyzed by Jones-Dole equation. The activation parameters of viscous flow have been obtained to throw light on the mechanism of viscous flow. The values of apparent molar volume, partial molar volume at infinite dilution and relative viscosities of each amino acid in various aqueous tripotassium citrate solutions have been evaluated from the density and viscosity data. The partial molar volumes of transfer from water to aqueous tripotassium citrate solution at infinite dilution have also been calculated. Transfer volume data have been used to calculate the pair and triplet interactions. The results have been discussed in terms of solute-solute and solute-solvent interactions and the structural changes of the solutes in solutions.  相似文献

A method for fast sensitive ultraviolet detection of amino acids was developed with a disposable electrophoresis microdevice. The microdevice was conveniently constructed by fixing a fused-silica capillary with a sampling fracture to a printed circuit board. During the separation process, the on-column conjugation of amino acids with cupric cation led to the ultraviolet absorption at 232 nm that could be directly used for fast analysis of amino acids. Using 20 mM boric acid (pH 5.3) containing 5 mM cupric cation and 0.015% Tween 20 as running buffer, this method could completely separate lysine, glutamine and serine at a sampling time of 2 s at +210 V and a separation voltage of +1800 V (240 V/cm). The theoretical plate numbers were from 140,000 to 205,000 plates/m. The linear ranges were from 10 to 500 μM for lysine, 20-1000 μM for glutamine and serine. The novel protocol had been successfully used to detect amino acids in beverage samples with recovery more than 85.0%, indicating its advantages and potential analytical application in different fields.  相似文献
