对数生长前期的大肠杆菌在生理盐水中用CaCl2可以直接诱导建立感受态并完成转化过程。利用H-8100透射电子显微镜观察用CaCl2直接在生理盐水中诱导建立感受态的大肠杆菌细胞,发现感受态细胞整体电子密度趋于均匀,但存在有电子密度较高颗粒,表明大肠杆菌感受态建立过程中,细胞整体对电子通透性增强,可能存在涉及细胞膜通透性的复合物重新形成相关的胞内物质代谢及其在细胞内定位的受控改变,暗示大肠杆菌可能具有建立自然感受态的能力。 相似文献

Using scanning tunneling microscopy (STM) and time of flight secondary ion mass spectrometry (TOF/SIMS), we observed radiation effects on a Si(1 1 1)-(7 × 7) surface in the collision of a single highly charged ion (HCI) with a charge state q up to q = 50. The STM observation with atomic resolution revealed that a nanometer sized crater-like structure was created by a single HCI impact, where the size increased rapidly with q. The secondary ion yields also increased with q in which multiply charged Si ions (Sin+) were clearly observed in higher q HCI-collisions. The sputtering mechanism is briefly discussed, based on the so-called Coulomb explosion model. 相似文献

Carbon NEXAFS and EELS spectra of soot, and NEXAFS spectra of soot extracts, are presented. The EELS spectra of solid soot particles from a TEM-EELS show fewer structures than the corresponding NEXAFS spectra obtained at two different synchrotron beamlines. We attribute radiation damage in the TEM-EELS to the failure at resolving structures of surface functional carbon groups in or on soot. NEXAFS spectra of soot extracts studied with a scanning transmission X-ray microscope show alterations during X-ray exposure, which can be explained by a simple chemical model where oxygen apparently reacts with the sample. When the same extract is studied in an ultrahigh-vacuum beamline, no such alterations are observed. 相似文献

Characterization and phase transitions in pure and 0.5% BaTiO3 nano-dispersed liquid crystalline (LC) N-(p-n-heptyloxybenzylidene)-p-n-nonyloxy aniline, 7O.O9, com-pounds are carried out using a polarizing microscope attached with hot stage and camera. We observed that when any of these images are distorted, different local structures suffer from various degradations in a gradient magnitude. So, we examined the pixel-wise gradient magnitude similarity between the reference and distorted images combined with a novel pooling strategy – the standard deviation of the GMS map – to determine the overall phase transition variations. In this regard, MATLAB software is used for gradient measurement technique to identify the phase transitions and transition temperature of the pure and nano-dispersed LC compounds. The image analysis of this method proposed is in good agreement with the standard methods like polarizing microscope (POM) and differential scanning calorimeter (DSC). 0.5% BaTiO3 nano-dispersed 7O.O9 compound induces cholesteric phase quenching the nematic phase, which the pure compound exhibits. 相似文献

A mapping technique has been developed where a sub‐micrometer focused polychromatic X‐ray beam is scanned across a stationary sample instead of scanning the sample in front of the X‐ray microbeam. This method is applied to a gold nanowire during its mechanical loading using the tip of an atomic force microscope. During the loading process, such a sample is `accelero‐phobic', i.e. the sample scanning stages must not to be moved to avoid parasitic additional load. Without beam scanning, only one single position within the sample can be probed during the test. The probed material point may even change because of drifts or movements induced by the test itself. The new scanning approach facilitates the in situ mapping of the entire wire giving access to the evolution of the wire shape as well as to the boundary conditions. This novel scanning technique opens promising perspectives for studies where sample motion is forbidden because of the sample environment. 相似文献
