Optimum conditions for transformed Panax ginseng hairy roots in flask culture

Panax ginseng hairy roots were transformed by Agrobacterium rhizogenes KTCT 2744. They showed an active branching pattern and fast growth in hormone-free medium, and good growth at 23°C, pH 5.8, 1/2 MS medium, and 3% sucrose. Sucrose provided the highest growth among seven carbon sources tested. Six complex media were also tested. In the combined sugar study, hairy roots grew better on sucrose without glucose or fructose than with glucose or fructose. In the 1/2 MS basal medium, 30 mM in nitrogen and 0.62 mM phosphate salt concentration was the optimum. The growth ratio was maximal at an inoculum size of 0.4% (w/v). Crude saponin and polysaccharide levels were also measured.     

Isotachophoretic determination of theanine

Free amino acids, the key quality components of tea, are contained in higher amounts in higher grade teas. Among free amino acids in tea, the content of theanine, which is the amino acid of the highest amount, shows high correlation to the price of green tea. A capillary isotachophoresis (CITP) method was developed for the analysis of L-theanine in tea and food supplements. The optimized electrolyte system was following: 0.01 mol/l HCl + 0.02 mol/l TRIS + 0.05% HEC (leading electrolyte), 0.01 mol/L-valine + barium hydroxide to pH 10 (terminating electrolyte). Good separation of L-theanine from other components of sample was achieved within 20 min. Method characteristics, i.e., linearity (0–200 mg/l), accuracy (99 ± 2 %), intra-assay (1.5 %), quantification limit (2 mg/l), and detection limit (0.7 mg/l) were determined. Sufficient sensitivity, low labouriousness (extraction only) and low running cost are important attributes of this method. It was proved that the developed method is suitable for the routine analysis of L-theanine in green tea and food supplements containing green tea extract.     

做青强度对岭头单枞茶香气成分影响的研究

用气相色谱－质谱法分析了岭头单枞乌龙茶不同做青强度的茶叶香气组分的变化。结果表明，岭头单枞宜用偏轻做青。偏轻做青时芳樟醇及其氧化物含量较高，香气指数（Ｏｗｕｏｒ指数ＦＩ值）较高（达１２９１）。成品经感官审评，蜜韵显著，有花香，符合该品种风格特征，做青强度系数，以机摇系数００３１、全程系数００３８为佳。     

红花中红花黄色素含量的反相高效液相色谱测定

本文分离纯化了红花（Carthamus tinctorius L）中Safflower yellow A，Safflomin A，Safflower yellow B，Safflomin C四种红花黄色素，并用所纯化的红花黄色素做标准品，用反相高效液相色谱法，梯度洗脱，同时测定了不同产地红花中四种红花黄色素的含量。该方的法准确度高，重现性好。     

Polysaccharides of Saponin-Bearing Plants. XIV. Structural Study of Glucoarabinogalactan from Acanthophyllum pungens Roots

The structure of the polysaccharide glucoarabinogalactan, which is a branched polysaccharide, was established by chemical and spectral methods. Its main chain consists of -16-bound galactopyranose units. Galactopyranose atoms C-2 and C-3 act as branching points. The side chains contain galactopyranose, glucopyranose, and chains of -(13)-bound arabinopyranose units. The linear structures of three galactooligosaccharides were identified and proved by partial acid hydrolysis.

偶氮胂酸型荧光试剂测定铝的研究

比较了５种新试剂及偶氮胂－Ｉ的铝铬合物的荧光性能，讨论了试剂结构与性能之间的关系。试验出７－〔（４－甲基－２－胂酸基苯）偶氮〕－８－羟基喹啉－５－磺酸为铝的优良荧光试剂，并用其测定茶叶、含茶叶饲料，以及大白鼠排泄物中的铝和水样中的铝，结果满意。     

氢化物发生-原子荧光光谱法测定黄磷中微量砷

采用氢化物发生-原子荧光光谱法测定黄磷中微量砷,研究了样品消化方法以及仪器工作条件、还原剂等对测定的影响,确定了最佳试验条件.在最佳试验条件下,荧光强度与砷(Ⅲ)的质量浓度在0.06～10μg·L-1范围内呈线性关系,相关系数为0.999 4,检出限为0.03μg·L-1,样品分析结果的相对标准偏差小于3.5 %,加标回收率在93.80～103.6%之间.     

波谱分析在沙棘果多糖Hn结构研究中的应用

以波谱分析为主要手段研究沙棘果多糖Hn的结构，GC分析表明Hn由Ara，Gal，Man和Glc组成，单糖物质的量比与利用GC分析Hn甲基化产物的结果一致．^13C—NMR分析表明Hn中存在α，β型糖苷键．利用GC—MS分析进一步确定Hn主链结构由β（1→4）Glc，β（1→6）Man和β（1→4）Gal构成，在3-O处有分支，支链由α（1→4）Glc，（1→6）Gal或末端Ara，Glc构成．     

鲍鱼多糖Hal-A的甲醇解研究

从鲍鱼中分离出一种鲍鱼多糖Hal-A，并用改进甲醇解方法研究了Hal-A的组成 ，该多糖甲醇解产物经三甲硅醚衍生后，进行GC/MS，结果表明鲍鱼多糖Hal-A主要 由葡萄糖、半乳糖、甘露糖和少量木糖、岩灌糖和半乳糖醛酸组成。
