A thin-layer chromatographic procedure is reported for the separation of serotonin from epinephrine and norepinephrine. The method involves the use of ethylenediaminetetraacetic acid-impregnated silica gel G plates and n-butanol-ethanol-acetic acid-water (8:2:1:3) as developer. Serotonin is well separated from both epinephrine and norepinephrine. Epinephrine and norepinephrine overlapped slightly but were both detectable. The procedure is thus applicable to the separation of all three biogenic amines in spite of the absence of an absolutely clear separation of epinephrine from norepinephrine. 相似文献

of valerenic acid in Valeriana jatamansi and Valeriana officinalis which is one of the stable compounds of Valeriana officinalis and designated as a key marker compound. Valerenic acid makes substantial contribution to the sedative and spasmolytic activity
of the essential oil and extract of Valeriana officinalis. Separation and quantification was achieved by HPTLC using ternary mobile phase of hexane: ethyl acetate: acetic acid (80:20:0.5

An electrochemical method for the simultaneous determinations of HgII concentration and total AsIII and AsV concentration has been developed. The method does not require the additional preliminary step of the chemical reduction of AsV to AsIII, or oxidation of AsIII to AsV before stripping analysis takes place. Also, the method for the simultaneous determination of HgII concentration and AsIII concentration is described. Measurements were performed in 0.1 M HCl using a gold-plated graphite electrode as sensor. Detection limits for both methods are below 0.4 ppb. Relative standard deviation did not exceed 15%. The possible interference by other trace metals was investigated. Analyses of natural water and industrial solutions were made using proposed methods and AAS. The t-test demonstrates that there was no significant difference between the results obtained with these methods. Proposed methods decrease the time of analysis because concentrations of the HgII and arsenic ions were measured simultaneously. Also, the removal of the additional step of chemical reduction of AsV to AsIII or oxidation of AsIII to AsV decreases analysis time, and also reduces the chance of contamination due to the use of additional reagents. 相似文献

Cytochrome P-450 isozymes represent a critical component of nature’s spectrum of detoxification catalysts that could be exploited
for bioremediation. The ethanol-inducible human cytochrome P-450 2E1 serves as a model eukaryotic P-450 that complements the
bacterial P-450 cam in dehalogenation and detoxification of environmental pollutants. We explored the construction of novel
chimeric P-450s using cytochrome P-450 camC and 2E1 genes. For construction of chimera 1 (478 amino acids, 55.14 kDa), 145 amino acids from the N-terminus of P-450 2E1 protein
(493 amino acids, 56.84 kDa) were replaced with 130 amino acids from the N-terminus of P-450 camC protein (415 amino acids,
46.66 kDa). In chimera 2 (525 amino acids, 60.24 kDa) the strategy involves replacement of 28 amino acids in the C-terminus
of chimera 1 with 75 amino acids from the C-terminus of P-450 camC gene. Homology models of both the chimeric proteins were
developed using SWISS-MODEL based on the known crystal structure of cytochrome P-450 camC, BM-3, 1DT6A, and 2C17A. The models
indicated that the proposed heme-binding site was intact, which is inevitable for catalytic activity of cytochrome P-450s.
The expression of chimera 1 and 2 genes in Escherichia coli DH5α was evident from light-pink cell pellets, protein band in sodium dodecyl sulfate polyacrylamide gel electrophoresis,
and diagnostic carbon monoxide-difference spectra. Our studies show that strategies can be developed to exploit the natural
diversity of the P-450 superfamily to generate chimeric biocatalysts that would provide new templates amenable to directed

The formation of a new ternary ion-associate complex of vanadium(V) with 2,3-dihydroxynaphthalene and iodonitrotetrazolium
chloride with a composition ratio of 1:2:1 is reported. The complex is quantitatively extracted from water into chloroform.
The molar absorptivity (ɛ) of the extract at λ
max=340 nm is 2.5 × 104 dm3/mol cm, and Beer’s law is obeyed for concentrations ranging from 0.1 to 0.9 μg/cm3 V(V). The following constants are determined: the extraction constant, the association constant, the distribution constant,
and the recovery factor. The effects of foreign ions and reagents are studied. A selective and sensitive method is developed
