Plasticized protein films were prepared by the casting method from water solution of sodium caseinate and plasticizers with the aim to obtain environmentally friendly materials for packaging applications. Mechanical properties (tensile strength, elongation and Young’s modulus) of caseinate based films were determined versus ratio of protein to plasticizer, plasticizer type and relative humidity conditions. Among the different polyol-type plasticizers tested, glycerol (Gly) and triethanolamine (TEA) were the most efficient for the improvement of mechanical properties (high strains for low stresses). Further, chemical crosslinking between formaldehyde (HCHO) and free amino groups (ε-NH2) of sodium caseinate was performed to increase water resistance of TEA plasticized films. Optimal mechanical properties, i.e. elastic modulus of 105 MPa, tensile strength of 8-9 MPa for elongation at break about 110-125% were obtained for HCHO/ε-NH2 ratios higher than 1.35. Protein specific water solubility was determined from a 280 nm absorbance. For convenient crosslinker (HCHO) content sodium caseinate solubility can be lowered to less than 5 wt% after 24 h immersion in water.  相似文献

The transport of Na+ out of the cell and K+ into the cell against a concentration gradient is catalyzed by a (Na+ + K+)-activated ATPase. The way in which the cations pass through the cell membrane has not yet been elucidated. Studies on the ATP hydrolysis revealed a Na+-dependent phosphorylation of the enzyme protein; the conformation of the enzyme also appears to change. The energy required for transport of the cations against their concentration gradients is probably provided by K+-dependent hydrolysis of the enzyme-bound phosphate. The enzyme can synthesize ATP from inorganic phosphate and ADP on reversal of the cation concentration gradient. By keeping the enzyme in a particular conformation, the cardiac glycoside ouabain specifically inhibits the Na+ pump.  相似文献

It is difficult to quantify NH4+ by ion chromatography in the presence of high concentrations of Na+ due to peak overlap. The Dionex IonPac CS15 column, which contains phosphonate, carboxylate, and 18-crown-6 functional groups, was originally developed to overcome this problem. We have found that the addition of 18-crown-6 to the eluent promotes improved peak resolution between Na+ and NH4+ even at concentrations as high as 60,000 to 1 using this column. Its use also improves the separation of alkali and alkaline earth metal and amine cations. Mobile phase 18-crown-6 increased the retention times of CH3NH3+, NH4+, and K+, and decreased the retention time of Sr2+. The retention times of Li+, Na+, Mg2+, Ca2+, (CH3)2NH2+, and (CH3)3NH+ were not affected. This method makes possible the direct analysis of ammonia from nitrogenase, the enzyme responsible for biological nitrogen fixation. The resolution of the NH4+ peak from the Na+ and Mg2+ peaks improved from zero resolution to values of 6.19 and 5.65, respectively. This technique considerably reduces the analysis time of NH4+ in the presence of high concentrations of Mg2+ and Na+ over traditional indophenol measurements.  相似文献

Surface properties of the binary mixed systems of decyl- and dodecylpyridinium chloride or bromide and sodium pentyl- and heptylsulfonate have been investigated. The surface tension of solutions of equimolar mixtures of surfactants and individual surfactants was measured, and the composition of mixed monolayers and surface interaction parameter β were calculated with the regular solution theory. Our results indicate that the properties of mixed films depend on both ionic strength and the kind of added inorganic electrolyte. With the increase of inorganic electrolyte concentration, the content of more surface active ions in the adsorption films enhances and is the highest in the presence of NaI and the smallest when solutions contain NaCl. Mutual interactions in mixed adsorbed films were found to be attractive. However, the strength of interaction weakens with the increase of ionic strength and depends on the kind of inorganic ions in the order: Cl>Br>I.  相似文献

Sodium insertion in the tetrahedral layer structure of the ferrites Ba2−xSrxFe4O8 was performed by solid state reaction at 1220 K in air. Superstoichiometric oxides with the actual formula (Ba2−xSrx)1−y/4NayFe4O8y0.56; 0.60Ba/Sr1.67—were characterized by X-ray and neutron powder diffraction. The hexagonal unit-cell volume shows an increasing dependence on the sodium insertion when the Ba/Sr ratio reaches the largest values. The marked expansion of the c parameter is the likely signature of the location of the inserted sodium cations within the interlayer space. One-half of the sodium cations partly sits on the Sr(Ba) sites in octahedral coordination and the other half occupies extra octahedral and tetrahedral sites. ac conductivity measurements point to a cationic conductivity whose thermally activated regime—Ea 0.7 eV—evidenced from 570 K, is unsensitive to the sodium content. The bottleneck of the 2D sodium mobility regards the crossing of the oxygen triangular faces shared by the different polyhedra within the interlayer space.  相似文献

The salt effects on molecular orientation at air/liquid methanol interface were investigated by the polarization-dependent sum frequency generation vibrational spectroscopy(SFG-VS). We clarified that the average tilting angle of the methyl group to be u = 308 58 at the air/pure methanol surface assuming a d-function orientational distribution. Upon the addition of 3 mol/L Na I, the methyl group tilts further away from the surface normal with a new u = 418 38. This orientational change does not explain the enhancement of the SFG-VS intensities when adding Na I, implying the number density of the methanol molecules with a net polar ordering in the surface region also changed with the Na I concentrations. These spectroscopic findings shed new light on the salt effects on the surfaces structures of the polar organic solutions. It was also shown that the accurate determination of the bulk refractive indices and Raman depolarization ratios for different salt concentrations is crucial to quantitatively interpret the SFG-VS data.  相似文献

采用薄膜分散法合成磷脂微囊,根据胶粒的双电层理论,通过在微囊中加入氯化锰、氯化钙和氯化镁电解质溶液,使微囊处于相对稳定的状态.研究发现加入氯化锰和氯化钙溶液,微囊胶体的粒径没有明显的变化,但加入一定浓度氯化镁溶液,其粒径明显变大.为了进一步增加磷脂微囊稳定性,将氯化锰、氯化钙、氯化镁磷脂微囊胶体分别与海藻酸钠(SA)溶液混合.结果表明,氯化镁与SA几乎不能形成水凝胶,氯化钙与SA形成水凝胶能力强于氯化锰.微囊胶体溶液中的磷脂酰丝氨酸(PS)可以与Ca~(2+)和Mg~(2+)键合形成PS-Ca~(2+)和PS-Mg~(2+),但不能与Mn~(2+)键合形成PS-Mn~(2+).对氯化钙磷脂微囊与海藻酸钠合成的复合水凝胶的形貌、溶胀率及细胞毒性进行了表征,结果表明,氯化钙与SA形成的水凝胶可以捕获胶体中磷脂微囊,且形貌规整,结构稳定,无细胞毒性.  相似文献

This work explored the possibility of coupling the toxin receptor-binding principle with the piezoelectric transduction principle. The sensing component of the saxitoxin biosensor involves a piezoelectric quartz crystal that was coated with sodium channel receptors. The sodium channel receptors were isolated from the electroplax organ of Electrophorus electricus. Binding of the sodium channel extracts to the quartz crystal was facilitated by pre-coating the gold electrode with a hydrophobic self-assembled monolayer of dodecanethiol. The instrumentation system consisted of a flow cell that held the quartz crystal, an oscillator circuit, an injection port, and a frequency counter that was connected to a personal computer. The various immobilization and measurement parameters were optimized. Binding of saxitoxin standards with the immobilized sodium channels was monitored through the decrease in the crystal oscillation frequency readings (ΔF) upon the introduction of saxitoxin into the flow cell. A calibration curve for saxitoxin was constructed by plotting the ΔF values vs. saxitoxin concentrations in the range from 0.1 to 2.0 μg/mL. A correlation coefficient of 0.9653 was obtained. The saxitoxin biosensor developed has the potential to be applied to the rapid screening of total paralytic shellfish poisoning toxins.  相似文献

目的以重组人钠/碘转运体(hNIS)基因转染结肠癌SW480细胞并检测hNIS mRNA及蛋白的表达,为放射性碘治疗非甲状腺肿瘤提供新思路。方法将构建好的重组质粒(pcDNA3.1+-hNIS)进行酶切、测序鉴定并扩增、提取。SW480细胞分为重组质粒(pcDNA3.1+-hNIS)转染组、空白质粒(pcDNA3.1+)转染组、空白对照组,转染后以RT-PCR和Western blot检测各组细胞hNIS mRNA和蛋白的表达。结果酶切和测序结果显示插入的hNIS基因大小和方向均正确。RT-PCR和Western blot显示重组质粒转染组SW480细胞可见hNIS mRNA和蛋白的表达,而空白对照组和空白质粒转染组均未检测到hNIS mRNA和蛋白的表达。结论脂质体法可有效地将hNIS基因转染至SW480细胞并成功表达hNIS蛋白。  相似文献

The novel heteronuclear complexes [Na2Zn(dipic)2(H2O)] n (1) and [Na2Mn(dipic)2(H2O)2] n (2) (H2dipic?=?pyridine-2,6-dicarboxylic acid) were synthesized and characterized by single-crystal X-ray diffraction, analysis, IR spectroscopy and thermogravimetric analysis. Complex 1 consists of infinite 2D double layers, which are further attached into a 3D network through π–π stacking. All sodium ions are 3-coordinate. Complex 2 has an intricate 3D structure, in which two sodium ions are differently involved in forming polymeric coordination units, and are 3- and 7-coordinate, respectively. Dipic anions act as multidentate ligands showing novel and unprecedented bridging coordination modes. One ligand coordinates to three metal ions and with five bonds; the other coordinates five metal ions with nine bonds.  相似文献
