We describe a novel method for rapid and ultrasensitive detection of intact glycoproteins without enzymatic pretreatment which was commonly used in proteomic research. This method is based on using gold nanoparticle (AuNP) as signal tag in laser desorption/ionization mass spectrometry (LDI-MS) analysis combined with boronic acid assisted isolation strategy. Briefly speaking, target glycoproteins were firstly isolated from sample solution with boronic acid functionalized magnetic microparticles, and then the surface modified gold nanoparticles were added to covalently bind to the glycoproteins. After that, these AuNP tagged glycoproteins were eluted from magnetic microparticles and applied to LDI-MS analysis. The mass signal of AuNP rather than that of glycoprotein was detected and recorded in this strategy. Through data processing of different standard glycoproteins, we have demonstrated that the signal of AuNP could be used to quantitatively represent glycoprotein. This method allows femtomolar detection of intact glycoproteins. We believe that the successful validation of this method on three different kinds of glycoproteins suggests the potential use for tracking trace amount of target glycoproteins in real biological samples in the near future.  相似文献

The matrix enhancement effect in gas chromatography (GC) has been a problem for the last decade and results in unexpected high recovery. Most efforts, including the use of different types of injectors/matrix simplification procedures, and further clean-up associated with removing this effect has focused on equalizing the response of the standard in the solvent and matrix. However, after eliminating the matrix enhancement effect, the sensitivity of GC remained unchanged. But, GC sensitivity can be increased by utilizing this matrix effect originating from a matrix-matched standard. Very few studies have highlighted utilizing the matrix effect but have rather advocated eliminating it. Analyte protectants (3-ethoxy-1,2-propanediol, gulonolactone and sorbitol) have been introduced as an alternative for GC–mass spectroscopy (GC–MS) (not examined for other GC detectors), as they equalize the response without removing the matrix effect, and, hence, increase sensitivity. Versatile applications of analyte protectants are not observed in practice. The European guidelines recommend the use of matrix-matched standard calibration for residue measurements. As a result, numerous applications are available for matrix-matched standards that compensate for the matrix effect. Moreover, the matrices (among them pepper leaf matrix) act as a protectant for thermolabile analytes in some cases. A lower detection limit should be achieved to comply with the maximum residue limits. Therefore, the matrix enhancement effect, which is considered a problem, can play an important role in lowering the detection limit by increasing the transfer of analyte from the injection port to the detector.  相似文献

Fluorescence correlation spectroscopy (FCS) data acquisition and analysis routines were developed and implemented in a home-built, multiparameter photon-counting microscope. Laser excitation conditions were investigated for two representative fluorescent probes, Rhodamine110 and enhanced green fluorescent protein (EGFP). Reliable local concentrations and diffusion constants were obtained by fitting measured FCS curves, provided that the excitation intensity did not exceed 20% of the saturation level for each fluorophore. Accurate results were obtained from FCS measurements for sample concentrations varying from pM to μM range, as well as for conditions of high background signals. These experimental constraints were found to be determined by characteristics of the detection system and by the saturation behavior of the fluorescent probes. These factors actually limit the average number of photons that can be collected from a single fluorophore passing through the detection volume. The versatility of our setup and the data analysis capabilities were tested by measuring the mobility of EGFP in the nucleus of Drosophila cells under conditions of high concentration and molecular crowding. As a bioanalytical application, we studied by FCS the binding affinity of a novel peptide-based drug to the cancer-regulating STAT3 protein and corroborated the results with fluorescence polarization analysis derived from the same photon data.  相似文献

A signal‐enhanced immunosensor has been developed by self‐assembling Au NPs onto a ferrocene‐branched poly(allylamine)/multiwalled carbon nanotubes (PAA‐Fc/MWNTs) modified electrode for the sensitive determination of hepatitis B surface antigen (HBsAg) as a model protein. The formation of PAA‐Fc/MWNTs composite not only effectively avoided the leakage of Fc and retained its electrochemical activity, but also enhanced the conductivity and charge‐transport properties of the composite. Further adsorption of Au NPs into the PAA matrix provided both the interactive sites for the immobilization of hepatitis B surface antibody (HBsAb) and a favorable microenvironment to maintain its activity. Tests performed with this immunosensor showed a specific response to HBsAg in the range of 0.1–350.0 ng mL?1 with a detection limit of 0.03 ng mL?1.  相似文献

A signal sequence detector in a high areal density recording channel is required to provide robust compensation against unexpected error events. Primarily, a number of error events are caused by media noise and nonlinear distortion. The same problem of signal sequence detection remains to be solved in a future magnetic recording system that comes in predisposed to trend for recording by large-sector size instead of existing single-sector one that consists of 512 information 8-bits bytes. For the above problem, this paper shows the signal estimation method based on statistical inference for such a finite mixture model with known number of degraded noise components. Our signal detection scheme with multivariate autoregressive models for total noise and the expectation maximization algorithm is applied to maximum a posteriori estimation for multivariate mixtures of noise. Furthermore, a non-binary low-density parity-check (LDPC) code is used for an error-correcting code that satisfies the specific run-length limited condition in the proposed system. It shows that the proposed error-correcting and signal detection methods are effective in estimating signal sequences degraded by media noise and in improving the error rate performances with respect to the conventional system using the binary LDPC code and univariate autoregressive model.  相似文献

拉曼光谱技术是一种高灵敏度、无损伤、振动分子光谱技术,在医药、生物、分析化学等诸多领域有着重要的作用。然而,由于拉曼散射强度低,实际测得的拉曼信号容易被噪声所污染。特别是在较短的曝光时间,收集到的拉曼光谱的信噪比很低。因此,提出了一种基于匹配追踪算法的信号重构方法,用于提取低信噪比的拉曼信号。该方法首先通过阈值循环迭代的方法在平均谱上找出特征峰的位置、估计峰的区间。根据峰的位置区间等信息,用高斯密度函数生成字典。在噪声谱上,根据特征峰位置和区间,将其区分为有信号区间和无信号区间,在有信号区间上利用匹配追踪算法重构被噪声所掩盖的拉曼信号。该算法不仅能够很好的逼近掩盖在噪声中的拉曼信号,且在重构信号的过程中也会对基线进行扣除,无须作基线校正处理。在仿真和实验中对该算法与常规算法进行了比较,结果证明,该算法在低信噪比条件下能够较好的恢复拉曼信号。该算法不同于传统光谱去噪算法,能同时对拉曼光谱进行了基线扣除以及噪声的处理,且能取得较为理想的结果,不需要使用不同的算法对基线和噪声分别处理。其次,在算法上我们创造性地将匹配追踪算法用于拉曼光谱信号的稀疏逼近求解。  相似文献

细胞内原位信号放大策略是检测低丰度内源性目标物的有效手段, 但多数信号放大策略依赖于外源性辅助物, 不可避免地改变细胞内微环境, 进而对机体造成一定干扰. 针对此问题, 可利用细胞内源性物质(如金属离子、 核酸、 蛋白酶等)实现原位荧光信号放大, 对不同生物标志物进行荧光成像, 此方法对低丰度靶分子检测及成像具有重要意义. 本文对内源性物质辅助信号放大及细胞内荧光成像相关研究进行了归纳整理, 介绍了内源性核酸、 酶、 蛋白质、 三磷酸腺苷(ATP)和金属离子辅助信号放大策略, 并探讨了其信号放大机理; 总结了内源性物质辅助信号放大探针在低丰度物质检测及成像方面的研究进展; 最后展望了该策略在细胞成像方面的优势及应用前景.  相似文献
