Many patients with dysphonia do not need to modify their respiratory patterns. For those that do, a simple four-point program is offered that will usually aid the patient in developing better expiration control. First, the patient is instructed that inspiratory-expiratory respiration is a continuous ongoing movement. Imagery is used to show that there is little or no discernible gap between inspiration-expiration. The patient begins working on expiratory control by prolonging phonation with no instruction given for “taking in” a breath. The patient's task is to prolong a sound with no discernible inspiratory effort before the sound is made. The patient is then asked to match a verbal target model, making no special effort to take in a breath before producing the model. Rapid and highly reproducible nonreducing agarose gel electrophoresis (NRAGE) of von Willebrand Factor (vWF) multimers was performed using a thermostated minigel apparatus that monitors and precisely controls internal gel temperature. The substitution of lithium dodecyl sulfate (LiDS) for sodium dodecyl sulfate (SDS) allowed electrophoresis to be performed below the 16 degrees C Krafft point of SDS and facilitated NRAGE of vWF over the entire range of 0-35 degrees C. Internal gel temperature was regulated by a thermocouple probe inserted directly into the gel during electrophoresis which interfaced with a thermopilot that continually measures and adjusts temperature to within +/- 0.5 degrees C. At 10 degrees C operative temperature, NRAGE at 1.5% agarose concentration was completed in 20 min at 250 V. Electrophoresis could be performed in only 10 min at 500 V, but at such high voltages, localized temperature fluctuations as much as 6 degrees C resulted in perturbation of banding patterns in those vicinities. In the optimized method, both high molecular weight multimers and proteolytic fragments of vWF were separable suggesting clinical applicability of this system for the diagnosis of von Willebrand Disease and thrombotic thrombocytopenic purpura. 相似文献

Random walk on heterogeneous networks is a recently emerging approach to effective disease gene prioritization. Laplacian normalization is a technique capable of normalizing the weight of edges in a network. We use this technique to normalize the gene matrix and the phenotype matrix before the construction of the heterogeneous network, and also use this idea to define the transition matrices of the heterogeneous network. Our method has remarkably better performance than the existing methods for recovering known gene–phenotype relationships. The Shannon information entropy of the distribution of the transition probabilities in our networks is found to be smaller than the networks constructed by the existing methods, implying that a higher number of top-ranked genes can be verified as disease genes. In fact, the most probable gene–phenotype relationships ranked within top 3 or top 5 in our gene lists can be confirmed by the OMIM database for many cases. Our algorithms have shown remarkably superior performance over the state-of-the-art algorithms for recovering gene–phenotype relationships. All Matlab codes can be available upon email request. 相似文献

In this article, we wish to investigate the dynamical behaviour of an SIRVS epidemic model with time-dependent coefficients. Under the quite weak assumptions, we give some new threshold conditions which determine whether or not the disease will go to extinction. The permanence and extinction of the infectious disease is studied. When the system degenerates into periodic or almost periodic system, the corresponding sharp threshold results are obtained for permanent endemicity versus extinction in terms of asymptotic time. In recent years, Huanglongbing (HLB) also known as citrus greening has greatly affected citrus orchards in Florida. This disease has caused significant economic and production losses costing about $750/acre for HLB management. Early and accurate detection of HLB is a critical management step to control the spread of this disease. This work focuses on the application of mid-infrared spectroscopy for the detection of HLB in citrus leaves. Leaf samples of healthy, nutrient-deficient, and HLB-infected trees were processed in two ways (process-1 and process-2) and analyzed using a rugged, portable mid-infrared spectrometer. Spectral absorbance data from the range of 5.15-10.72 μm (1942-933 cm−1) were preprocessed (baseline correction, negative offset correction, and removal of water absorbance band) and used for data analysis. The first and second derivatives were calculated using the Savitzky-Golay method. The preprocessed raw dataset, first derivatives dataset, and second derivatives dataset were first analyzed by principal component analysis. Then, the selected principal component scores were classified using two classification algorithms, quadratic discriminant analysis (QDA) and k-nearest neighbor (kNN). When the spectral data from leaf samples processed using process-1 were used for data analysis, the kNN-based algorithm yielded higher classification accuracies (especially nutrient-deficient leaf class) than that of the other spectral data (process-2). The performance of the kNN-based algorithm (higher than 95%) was better than the QDA-based algorithm. Moreover, among different types of datasets, preprocessed raw dataset resulted in higher classification accuracies than first and second derivatives datasets. The spectral peak in the region of 9.0-10.5 μm (952-1112 cm−1) was found to be distinctly different between the healthy and HLB-infected leaf samples. This carbohydrate peak could be attributed to the starch accumulation in the HLB-infected citrus leaves. Thus, this study demonstrates the applicability of mid-infrared spectroscopy for HLB detection in citrus. 相似文献
