An essential part of the modulation of protein‐binding capacity in hydrophobic interaction chromatography is the buffer‐salt system. Besides using “single” electrolytes, multicomponent electrolyte mixtures may be used as an additional tool. Both the protein solubility and the binding capacity depend on the position of a salt in the so‐called Hofmeister series. Specific interactions are observed for an individual protein‐salt combination. For salt mixtures, selectivity, recovery, and binding capacity do not behave like for the single salts that are positioned in between the two mixed components in the Hofmeister series, as the continuous correlation would suggest. Thus, finding strategies for mixed salts could potentially lead to improved capacities in hydrophobic interaction chromatography. Mixtures of ammonium sulfate, sodium citrate, sodium sulfate, sodium chloride, sodium acetate, and glycine were used to investigate the binding capacities for lysozyme and a monoclonal antibody on various hydrophobic resins. Resin capacity for two investigated proteins increases when mixtures consisting of a chaotropic and a kosmotropic salt are applied. It seems to be related to the rather basic isoelectric points of the proteins.  相似文献

Different methods for treatment of leachate from a municipal solid waste (MSW) landfill were tested in a pilot plant. Raw leachate was pre-treated with aeration and sedimentation, followed by several parallel individual steps such as bioremediation, chemical oxidation, ozonation, and geo-bed filters. The efficiency of different treatment steps was evaluated according to one previously developed protocol, which includes measurements of several parameters such as conductivity, pH, nutrients, chloride, metals, organic compounds, and acute toxicity before and after a treatment step. The treatment steps which showed the highest efficiency towards organic pollutants in leachate were ozone treatment and chemical oxidation. The use of an adsorption filter, a geo-bed with a mixture of peat and bottom ash with ca 10% remaining carbon, also had good effects. A combination of pre-treatment and a geo-bed filter with peat and carbon ash gave the best overall treatment results when water-quality parameters such as total organic carbon and ammonia-nitrogen were also considered.  相似文献

A capillary electrophoretic (CE) method for the determination of residual mid-chain alkyltrimethylammonium compounds in the pharmaceutical product Welchol™ (an alkylated, crosslinked polyallylamine) was developed, validated and compared with the existing ion chromatographic (IC) method with suppressed conductivity detection. Excellent reproducibilities of migration times (RSD<0.5% within a series of 55 sample injections) and relative peak areas (RSD<2%) make the method suitable for quality control as a real alternative to IC. Limits of quantification of 0.01% w/w of each impurity in the active substance were achieved. Buffer systems for indirect UV detection based on creatinine as visualization reagent with different inorganic and organic acids (phosphoric, sulfuric, formic, acetic, oxalic and citric acid) and their effect on selectivity to ten quaternary ammonium compounds were studied. Selectivity changes were observed for the di- and trivalent analytes depending on the buffer applied. Also, the influence of acetonitrile, methanol, 1,4-dioxane and tetrahydrofuran on selectivity was investigated. In addition, CE–MS experiments were carried out in order to identify several impurities in the product.  相似文献

Ammonium polyphosphate (APP) with crystalline form V (APP-V) was synthesized by heating a mixture of 85% food-grade phosphoric acid and melamine under dry ammonia atmosphere. Effects such as raw material ratios, charging methods, dehydration temperature, condensation temperature and condensation time were investigated. Its water solubility was tested, its XRD and FTIR were characterized and compared with those of industrial APP-I and APP-II. Results showed that the prepared APP-V was insoluble in water, its XRD spectrum was the same as APP-V in literature. The molecular structures of the prepared APP-V and APP-I/APP-II were speculated and discussed in detail.  相似文献

This review examines the most recent electrochemical developments for nitrate, nitrite and ammonium detection for on-site water monitoring. There remains a high demand for effective field-based detection of the dissolved inorganic nitrogen (DIN) analytes to aid in mitigating nitrogen loading. Electrochemical approaches show increasing potential to fill this role as advancements in nanotechnology continually improve analytical performance and on-site applicability. However, translating these improvements into the field still faces the resonating challenges of reaching analytical proficiency (selectivity, sensitivity, robustness, stability), practical end-user functionality, minimal matrix interferences and cost effectiveness. Herein, we elaborate on these challenges via a critical evaluation of current studies and examine how realistic the prospects of on-site nitrate, nitrite and ammonium are. We also present recommendations in addressing these gaps to conclude the review.  相似文献

Salt cavern gas storage is one of the vital strategic natural gas reserves and emergency peak shaving facilities all over the world. However, rock salt in China is primarily bedded salt, usually composed of many thin salt layers and interlayers (e.g., anhydrite, mudstone, and glauberite). During the water solution mining process of the cavern, the insoluble mudstones fall to the bottom and account for 1/3 up to 2/3 of the storage capacity. The bulk volume of the insoluble mudstones is almost twice its in-suit volume. It is of great urgency to investigate the swelling mechanisms of the bottom insoluble mudstones. Given this, we first analyzed the mineral composition of salt rock and insoluble mudstones by using XRD and SEM methods. Then, experimental studies were carried out considering both clay swelling and physical packing. At last, the zeta potential tests were conducted to reveal the swelling mechanisms of the bottom mudstones. Results show that the volumetric expansion of mudstones is made up of three parts: clay swelling, particle surface bound water volume, and pore space free water volume increase. Because the content of expansive clay in the bottom mudstones is less than 2%, and the high salinity brine in the cavern has excellent clay stability performance, clay swelling is not the main contributor to the volumetric expansion of the bottom mudstones. Measurement results show that the surface of the mudstones is negatively charged after hydration. Electrostatic repulsion can increase the spacing between small rock particles and creates approximately 47.6% of the pore space, which is the main factor in the volumetric expansion of mudstones. This study provides a theoretical basis for the mining solution and capacity enlargement during the construction of bedded salt cavern gas storage in China.  相似文献
