A fast and inexpensive sample preparation procedure based on the matrix solid-phase dispersion (MSPD) technique is proposed for the isolation of several organophosphate esters (mainly employed as flame retardants and plasticizers) from indoor dust samples. Extraction and clean-up were carried out in a single step and target compounds were determined by gas chromatography (GC) with nitrogen-phosphorus detection (NPD). The main parameters affecting extraction yield and selectivity, such as type and amount of dispersant material, clean-up co-sorbent and extraction solvent, were evaluated and optimised. Under final conditions, 0.5 g of dust were dispersed with equal amounts of anhydrous sodium sulphate and Florisil, and loaded on the top of a polypropylene cartridge containing 0.5 g of alumina. The dispersed sample was washed with 2 mL of n-hexane to remove the least polar interferences and analytes were eluted with 3 mL of acetone. Recoveries of the proposed method for spiked samples ranged from 80 to 116%, and the day-to-day variability remained between 5 and 10%. Data on levels of organophosphate species in dust from private houses and vehicle cabins are provided. In both cases, the lowest concentrations corresponded to the short chain, non-chlorinated, alkyl organophosphates, whereas mean values above 1 μg g−1 were measured for the rest of analytes.  相似文献

选用液相基质制样,考察了激光强度、回旋池开门时间等因数对基质辅助激光解吸电离-傅里叶变换离子回旋共振质谱(MALDI—FT—ICR—MS)检测结果的影响,优化了实验条件。使用液相制样方法对5类实际样品进行了MALDI—FT-ICR—MS检测,结果表明:液相基质具有很好的通用性,质谱信号稳定、持久。利用FT-ICR—MS特有的超高分辨率与准确度,能够很准确地测定化合物组成。  相似文献

In this paper, we propose the construction of merging arrangements for combining the information from various runs as a powerful approach to improve the resolution. The bacteriorhodopsin (bR) photocycle has been chosen in this study as an example dealing with the protein dynamics monitored by means of time-resolved step-scan FT-IR spectroscopy. The possibilities of matrix merging are evaluated and results are compared with those from the analysis of individual and augmented matrices. As a conclusion, this strategy provides excellent results for the analysis of this type of time-resolved FT-IR data.  相似文献

Liquid chromatography–tandem mass spectrometry (LC-MS/MS) is a versatile coupling system which combines both selectivity and sensitivity and certainty. Hence, it is generally considered as the most reliable technique to quantify chemical compounds in complex matrices. In the present paper, we evaluate the performance of LC-MS/MS methods for the quantification of 3-nitrotyrosine in human urine in order to point out its dependence on the design of the quantification method, and emphasize the role of matrix effects in the performance. We compare external and internal calibrations, isotope dilution and isotopomer-based exact matching. The role of both sample preparation and multiple transitions monitoring is particularly addressed.  相似文献

We analyzed ABC transporter solute-binding proteins (SBPs) of the Bacillus subtilis membrane using a proteomic approach. We prepared a washed cell membrane fraction that was insoluble in 134 mM nondetergent sulfobetaine and then extracted proteins using mixtures of detergents in a stepwise manner. The membrane proteins were resolved by three two-dimensional polyacrylamide gel electrophoresis (2-D PAGE) or two one-dimensional (1-D) PAGE procedures, electroblotted, and digested in the presence of 5% or 80% acetonitrile. Thereafter, matrix-assisted laser desorption/ionization-time of flight-mass spectrometry (MALDI-TOF MS) identified 637 proteins corresponding to 15.9% of the total cellular proteins. We predicted that among these, 256 were membrane proteins, 101 were lipoproteins or secretory proteins and 280 were soluble proteins containing peripheral proteins that function in both the cytoplasm and the cell membrane such as SecA and FtsY. Among the 637 proteins, we identified 30 SBPs among 38 importers predicted by a bioinformatic search of the genome. We confirmed expression of the genes for the 30 SBPs using DNA microarray analysis. We compared the 2-D gel separation profiles of submembrane fractions solubilized by 1% n-dodecyl-beta-D-maltoside from cells cultured on Luria Bertani (LB), S7, and S7 medium without glutamate as well as DNA microarray data on LB and S7. The results suggested that YcdH, YtmK and YurO are binding proteins for Mn(++), glutamate and glucose, respectively, and that YqiX and YxeM are binding proteins for amino acids (tryptophan in S7 medium).  相似文献

An analytical protocol combining a headspace technique with gas chromatography and detection by photoionization detector and flame ionization detector (HS-GC-PID-FID) was developed. This procedure was used to measure volatile organic compounds (VOCs) in environmental aqueous matrices and was applied in determination of VOCs on the coast of Fortaleza, Brazil. At optimum operating conditions, analytical figures of merit such as linearity (R ranged from 0.9983 to 0.9993), repeatability (5.62 to 9.63% and 0.02 to 0.19% for the quantitative and qualitative analyses, respectively), detection limits (0.22 to 7.48 μg L1) and sensibility were estimated. This protocol favors a fast sampling/sample preparation (in situ), minimizes the use of laboratory material, eliminates the matrix effect from environmental samples, and can be applied to river, estuarine and oceanic waters. The advantage of detectors in series is that a low sensitivity in detection in one is compensated by the other. Toluene was the most abundant VOC in the studied area, with an average concentration of 1.63 μg L1. It was followed by o-xylene (1.15 μg L1), trichloroethene (1.08 μg L1), benzene (0.86 μg L1), ethylbenzene (0.74 μg L1), carbon tetrachloride (0.55 μg L1), m/p-xylene (0.48 μg L1) and tetrachloroethene (0.46 μg L1), compounds which are very commonly detected in urban runoff from most cities. The results of the VOC distribution showed that port activity was not the main source of VOCs along the Fortaleza Coast, but that the contribution from urban runoff seemed more significant.  相似文献

A simple, inexpensive sample preparation procedure, based on the matrix solid-phase dispersion (MSPD) technique, for the determination of six UV filters: 2-ethylhexyl salicylate (EHS), 3,3,5-trimethylcyclohexyl salicylate (Homosalate, HMS), 3-(4-methylbenzylidene) camphor (4-MBC), isoamyl-p-methoxycinnamate (IAMC), 2-ethylhexyl-p-methoxycinnamate (EHMC) and octocrylene (OCR), in dust from indoor environments is presented and the influence of several operational parameters on the extraction performance discussed. Under the final working conditions, sieved samples (0.5 g) were mixed with the same amount of anhydrous sodium sulphate and dispersed with 2 g of octadecyl bonded silica (C18) in a mortar with a pestle. This blend was transferred to a polypropylene solid-phase extraction cartridge containing 2 g of activated silica, as the clean-up co-sorbent. The cartridge was first rinsed with 5 mL of n-hexane and the analytes were then recovered with 4 mL of acetonitrile. This extract was adjusted to 1 mL, filtered and the compounds were determined by gas chromatography combined with tandem mass spectrometry (GC–MS/MS). Recoveries for samples spiked at two different concentrations ranged between 77% and 99%, and the limits of quantification (LOQs) of the method between 10 and 40 ng g−1. Analysis of settled dust from different indoor areas, including private flats, public buildings and vehicle cabins, showed that EHMC and OCR were ubiquitous in this matrix, with maximum concentrations of 15 and 41 μg g−1, respectively. Both UV filters were also quantified in dust reference material SRM 2585 for first time. EHS, 4-MBC and IAMC were detected in some of the analyzed samples, although at lower concentrations than EHMC and OCR.  相似文献

Recent studies have shown that amphetamines and other drugs of abuse residues occur in wastewater. Consequently, several methods have been developed for their determination by solid-phase extraction (SPE) and liquid chromatography–tandem mass spectrometry (LC–MS/MS). However, a major drawback of these methods is the lack of selectivity during SPE that results in reduced sensitivity, due to matrix effects, and in some cases in low precision and poor accuracy. In order to tackle this problem, three different SPE alternatives have been evaluated in this work for the determination of five amphetamines: common hydrophilic balance (Oasis HLB), mixed-mode (Oasis MCX) and molecularly imprinted polymers (MIPs) sorbents. Among them, Oasis HLB showed the worst performance, as three amphetamines (MDA, MDMA and MDEA) could not be determined because of interfering signals in the LC–MS/MS chromatogram, and amphetamine recoveries could not be corrected by the use of the deuterated analogue internal standard. Oasis MCX permitted the determination of all target analytes, but with still strong signal suppression: ca. 70% signal drop with wastewater samples, which could in this case be corrected by the internal standards providing acceptable trueness (overall recoveries: 101–137%), precision (RSD: 2.0–12%) and limits of detection (LOD: 1.5–4.4 ng/L). Alternatively, MIPs rendered cleaner extracts with less matrix effects (ca. 30% signal drop), and thus lower LODs (0.5–2.7 ng/L) and even better trueness (91–114% overall recovery) and precision (1.5–4.4%RSD). The final application of the method with MIP cartridges showed the presence of MDA and MDMA in the seven analysed wastewaters at the 4–20 ng/L level.  相似文献

基质金属蛋白酶(MMPs)是一族Zn2+依赖的蛋白水解酶. 该族酶的过度表达与多种病理过程密切相关, 因此其抑制剂可用于这些疾病的治疗. 本文设计合成了15个α-卤代丁二酰氧肟酸类新型基质金属蛋白酶抑制剂, 经核磁共振氢谱和质谱进行了结构表征, 并以伊洛马司他(Ilomastat)为阳性对照, 分别测定了它们对基质金属蛋白酶MMP-2和MMP-9的体外抑制活性. 结果显示, 4个化合物对MMP-2的抑制活性与阳性对照相当; 5个化合物对MMP-9的抑制活性与对照药相当.  相似文献
