Synthesis and applications of new functional nanoparticles are topics of increasing interest in many fields of nanotechnology. Chemical modifications of inorganic nanoparticles are often necessary to improve their features as spectroscopic tracers or chemical sensors, and to increase water solubility and biocompatibility for applications in nano-biotechnology. Analysis and characterization of structured nanoparticles are then key steps for their synthesis optimization and final quality control. Many properties of structured nanoparticles are size-dependent. Particle size distribution analysis then provides fundamental analytical information. Asymmetrical flow field-flow fractionation (AF4) with multi-angle light scattering (MALS) detection is able to size-separate and to characterize nanosized analytes in dispersion. In this work we focus on the central role of AF4-MALS to analyze and characterize different types of structured nanoparticles that are finding increasing applications in nano-biotechnology and nanomedicine: polymer-coated gold nanoparticles, fluorescent silica nanoparticles, and quantum dots. AF4 not only size-fractionated these nanoparticles and measured their hydrodynamic radius (rh) distribution but it also separated them from the unbound, relatively low-Mr components of the nanoparticle structures which were still present in the sample solution. On-line MALS detection on real-time gave the gyration radius (rg) distribution of the fractionated nanoparticles. Additional information on nanoparticle morphology was then obtained from the rh/rg index. Stability of the nanoparticle dispersions was finally investigated. Aggregation of the fluorescent silica nanoparticles was found to depend on the concentration at which they were dispersed. Partial release of the polymeric coating from water-soluble QDs was found when shear stress was induced by increasing flowrates during fractionation. 相似文献

An acetylcholinesterase (AChE) biosensor was constructed based on gold nanoparticles (AuNPs) using electroless plating on vertical nitrogen-doped single-walled carbon nanotubes (VNSWCNTs) for detecting organophosphorus pesticides (OPs). AChE was immobilised on AuNPs via Au–S bonding, and VNSWCNTs were produced by spontaneous chemical adsorption of NSWCNTs on gold electrode, also via Au–S bonding. This modified electrode exhibited excellent electron transfer capacity due to the synergy between AuNPs and VNSWCNTs. The developed biosensor showed good linear relations at concentrations of 10?5 – 1 ppb, and the detection limits were 3.04 × 10?6 ppb for methyl parathion, 1.96 × 10?6 ppb for malathion and 2.06 × 10?6 ppb for chlorpyrifos, respectively. The AChE biosensor revealed satisfactory stability, excellent sensitivity and good repeatability. Recently, water‐soluble gold nanoclusters (AuNCs) have attracted more and more attention due to their unique properties. In this study, penicillamine‐protected gold nanoclusters (Pen‐AuNCs) were synthesized and initially fractionated by sequential size‐selective precipitation (SSSP). The crude Pen‐AuNCs and SSSP fractions were separated by capillary zone electrophoresis (CZE) with a diode array detector. The effects of key parameters, including the concentration of phosphate buffer, pH value and the ethanol content were systematically investigated. The separation of water‐soluble poly‐disperse AuNCs were well achieved at 30 mM phosphate buffer with 7.5% EtOH, pH 12.0, and applied voltage of 15 kV. The linear correlation between AuNCs diameter and mobility was observed. This finding provides an important reference for CE separation and product purification of water‐soluble AuNCs or other nanomaterials. 相似文献

介绍了一种利用互补核酸杂交富集金胶实现信号扩增的蛋白质生物传感器. 以凝血酶蛋白为研究对象, 利用凝血酶蛋白相对应的两段核酸适配体, 将适配体Ⅰ固定在磁性颗粒上, 用于特异性地捕获蛋白, 将适配体Ⅱ标记金胶作为检测信标. 由凝血酶蛋白和相对应的两段核酸适配体构建三明治结构的凝血酶蛋白生物传感器. 另外, 再通过信标金胶上过剩的核酸适配体链与另一段标记有金胶的互补核酸进一步杂交, 获得金胶的选择性聚集, 实现了信号扩增. 通过信号扩增, 使此传感器的灵敏度大大提高, 对凝血酶蛋白的检测下限可达到4.52×10-15 mol/L. 平行测定浓度为7.47×10-14 mol/L的凝血酶8次, 其RSD为3.0%. 该生物传感器对凝血酶蛋白有很好的特异性, 其它蛋白如溶菌酶和牛血清白蛋白的存在对于检测没有影响. 相似文献

考察了海水养殖区域缺氧沉积物中重金属形态连续萃取法萃取过程中溶液pH值、温度变化和胶体形成对形态分离结果的影响。结果表明,连续萃取法Step1萃取酸溶态时,萃取液pH值的轻微升高使Pb和Zn的萃取结果明显降低;Step2萃取还原态时,萃取液pH值明显升高对重金属的萃取结果影响很小;Step3萃取有机质结合态时,萃取液pH值的降低对重金属萃取结果影响较小(除Pb外)。由于连续萃取法中各步萃取反应的热力学过程不同,Step1和Step2萃取后溶液温度轻微升高而在Step3中溶液温度稍微降低,溶液温度变化(<2.5℃)对重金属形态的影响很小。在Step1、Step2和Step4萃取过程中胶体的形成和存在,对萃取液Cd、Zn和Cu有明显的吸附影响;而在Step3萃取中萃取剂的分散作用有效控制了溶液中胶体的形成。因此采用连续萃取方法评价沉积物中的重金属形态时,控制萃取液pH值和胶体形成有利于提高萃取结果的稳定性和准确性。 相似文献
