Recently, we have identified the first inhibitors of Rad6B, an E2 enzyme essential for post-replication DNA repair and a potential new drug target for the treatment of breast cancer. We report two newly optimised synthetic routes to our [4-amino-6-(phenylamino)-1,3,5-triazin-2-yl]methyl 4-nitrobenzoate target compounds TZ8 and TZ9 with general applicability for further structure–activity relationship studies around this pharmacophore. The key step involved the condensation/cyclisation between phenylbiguanide and either ethyl bromoacetate or dimethyloxalate in good yield.  相似文献

The geothermal steam turbines are exposed to mechanisms of corrosion/erosion that weakens its components and reduces their useful life. Due to this problem work has been done in application and characterization of coating in solid state by means of the technique of high-velocity Oxygen Fuel (HVOF), evaluating the corrosion rate (Vcorr) at high temperature of MCrAlY and Diamalloy 4006 coatings deposited on stainless steel SS304. Test was performance in an Autoclave at 170 0C using a modified geothermal fluid as electrolyte. Open circuit potential was monitoring during 24 hours until the system reached the equilibrium. After that, Polarization and Impedance Spectroscopy techniques were used to evaluate the specimens. For microstructure characterization; X–ray Diffraction (XRD), electron sweep microscope (SEM) and Optical microscope were applied. Results show that both coatings (Diamalloy 4006 and MCrAlY), have low current density compare with the substrate, which is an indicative of a lower corrosion rate due to the passive behavior of the species deposited on the Surface of the coating.  相似文献

The spectroscopic properties of Nω-nitro-l-arginine were investigated by FT-IR, UV-VIS, and 1H NMR spectra. Geometrical parameters and energies were calculated using the density functional theory (DFT) B3LYP method with the 6-311G basis set. Geometrical optimization of the molecule has been performed, vibrational spectra have been calculated, and fundamental vibrations have been determined from the total energy distribution (TED) of the vibrational modes. The HOMO-LUMO analysis is carried out for various electric fields (0.0–0.025 A?1). The HOMO-LUMO gap is decreased while increasing the electric field. The calculated quantum chemical parameters are calculated and correlated to the inhibition efficiency, A Mullliken population was also important for determining local reactivity by indicating reactive centers and identifying potential nucleophilic and electrophilic attack sites. Charge transfer occurs inside the compound based on the HOMO LUMO gap. Calculations of DFT were evaluated in their ability to predict inhibition efficiency.  相似文献

The preparation of Ni–SiC coatings using magnetic field-assisted jet electrodeposition under various plating settings is described in this study. A RBF-BP composite neural network with 4 × 4 × 4 × 7 × 10 × 1 was used to predict the corrosion resistance of Ni–SiC coatings prepared by employing different plating parameters. The results show that the fitting degree between the expected value and the actual value of the RBF-BP composite neural network is 0.97497. Moreover, the hybrid neural network can accurately predict the corrosion resistance of Ni–SiC coatings prepared under different process parameters. The corrosion weight loss of the coating is the lowest at the current density of 4 A/dm2, a jet rate of 3 m/s, a SiC particle concentration of 8 g/L, and at a magnetic field intensity of 0.8 T, demonstrating its corrosion resistance under these conditions. According to the coating characterization analysis, the coating's grain size was significantly refined, and the surface was smoother with a high amount of uniformly sized SiC nanoparticles.  相似文献

The effectiveness of 1H?pyrazole?3,5?dicarboxylic acid 5?benzyl ester 3?phenyl ester (PCBPE) as a preventer for deterioration of IS 513 Gr. D steel in 1 M HCl medium is evaluated via weight loss, electrochemical impedance, and polarization techniques. Kinetic and thermodynamic parameters assessed the feasibility of the adsorption process at diverse temperatures. The inhibition action on mild steel has been enhanced with increasing PCBPE concentration. It is found from the polarization studies that PCBPE behaves as mixed type inhibitor in HCl medium. The adsorption process of PCBPE on mild steel surface from acid environment is favoured Langmuir adsorption isotherm. The shielding efficiency of PCBPE has been enhanced at elevated concentrations, and it has been diminished at amplified temperatures. The Atomic Force Microscope (AFM), Scanning Electron Microscope (SEM), and Energy Dispersive Spectrum (EDS) were used to establish a surface characterization of metal specimens. A quantum chemical analysis of electron density distributions in the highest occupied molecular orbital (HOMO) and lowest unoccupied molecular orbital (LUMO) demonstrated how the inhibitor undergoes adsorption on mild steel in 1 M HCl. All experimental findings substantiate the corrosion mitigation performance of PCBPE on mild steel in acidic environments.  相似文献

The conformational changes in a sugar moiety along the hydrolytic pathway are key to understand the mechanism of glycoside hydrolases (GHs) and to design new inhibitors. The two predominant itineraries for mannosidases go via OS2B2,51S5 and 3S13H41C4. For the CAZy family 92, the conformational itinerary was unknown. Published complexes of Bacteroides thetaiotaomicron GH92 catalyst with a S-glycoside and mannoimidazole indicate a 4C14H5/1S51S5 mechanism. However, as observed with the GH125 family, S-glycosides may not act always as good mimics of GH's natural substrate. Here we present a cooperative study between computations and experiments where our results predict the E5B2,5/1S51S5 pathway for GH92 enzymes. Furthermore, we demonstrate the Michaelis complex mimicry of a new kind of C-disaccharides, whose biochemical applicability was still a chimera.  相似文献

In this study, α-glucosidase was successfully immobilized on cellulose filter paper and further applied to screening inhibitors from traditional Chinese medicines combined with capillary electrophoresis analysis. For α-glucosidase immobilization, a cellulose filter paper was used as the carrier and grafted with amino groups by coating chitosan, then α-glucosidase was covalently bonded on the amino-modified carrier via epoxy ring-opening reaction using polyethylene glycol diglycidyl ether as the crosslinker. Several parameters influencing the enzyme immobilization were optimized and the optimal values were enzyme concentration of 4 U/mL, polyethylene glycol diglycidyl ether concentration of 1.25%, chitosan concentration of 7.5 mg/mL, immobilization pH 7.0, crosslinking time of 4 h and immobilization time of 2 h. The immobilized α-glucosidase exhibited good batch-to-batch reproducibility (RSD = 2.1%, n = 5), excellent storage stability (73.5% of its initial activity after being stored at 4°C for 15 days), and reusability (75% of its initial activity after 10 repeated cycles). The Michaelis constant of immobilized α-glucosidase and half-maximal inhibitory concentration of acarbose were calculated to be 1.12 mM and 0.38 μM, respectively. Finally, the immobilized α-glucosidase was used for screening inhibitors from 14 kinds of Traditional Chinese Medicine extracts, and Sanguisorbae Radix showed the strongest inhibitory effect on α-glucosidase.  相似文献

Saxifraga atrata is an important traditional Tibetan medicine used to treat cough and pneumonia, and has tremendous medicinal potential. In this study, we devised a technique to separate 1,1-diphenyl-2-picrylhydrazyl inhibitors from a methanol extract of S. atrata. The material was first processed using MCI GEL CHP20P medium-pressure liquid chromatography, yielding 1.1 g of the target fraction Fr2. Subsequently, online hydrophilic interaction liquid chromatography-1,1-diphenyl-2-picrylhydrazyl assay was used to identify prospective 1,1-diphenyl-2-picrylhydrazyl inhibitors, and two 1,1-diphenyl-2-picrylhydrazyl inhibitor fractions (Fr24 and Fr25) were identified from Fr2. Then, medium-pressure preparation was continued using an XIon column to separate two 1,1-diphenyl-2-picrylhydrazyl inhibitor fractions (Fr24 and Fr25). The target compound was concentrated in fractions Fr24 and Fr25 using reverse-phase liquid chromatography during further separation procedures. Finally, the purity, structure, and 1,1-diphenyl-2-picrylhydrazyl inhibitory activity of the isolated 1,1-diphenyl-2-picrylhydrazyl inhibitors were determined. Two 1,1-diphenyl-2-picrylhydrazyl inhibitors (adenosine with the half maximal inhibitory concentration of 66.87 ± 14.33 μM and (-)-4-O-(E)-caffeoyl-l -threonic acid with the half maximal inhibitory concentration of 59.06 ± 5.02 μM) were isolated with purities exceeding 95%. The results showed that this technology is effective in the targeted separation of antioxidants from natural products.  相似文献

The extraction of Scutellaria baicalensis Georgi was investigated using the response surface methodology-genetic algorithm mathematical regression model, and the extraction variables were optimized to maximize the flavonoid yield. Furthermore, a simple and efficient ultrafiltration–liquid chromatography–mass spectrometry and molecular docking methods were developed for the rapid screening and identification of acetylcholinesterase inhibitors present in Scutellaria baicalensis Georgi. Subsequently, four major chemical constituents, namely baicalein, norwogonin, wogonin, and oroxylin A, were identified as potent acetylcholinesterase inhibitors. This novel approach, involving the use of ultrafiltration–liquid chromatography–mass spectrometry and molecular docking methods combined with stepwise flow rate counter-current chromatography and semi-preparative high-performance liquid chromatography, could potentially provide a powerful tool for the screening and extraction of acetylcholinesterase inhibitors from complex matrices and be a useful platform for the production of bioactive and nutraceutical ingredients.  相似文献

Natural-based corrosion inhibitors have gained great research interest thanks to their low cost and higher performance. In this work, the chemical composition of the methanolic extract of Ammi visnaga umbels (AVU) was evaluated by gas chromatography (GC) coupled with mass spectrometry (MS) and applied for corrosion inhibition of carbon steel (CS) in 1.0 mol/L HCl using chemical and electrochemical techniques along with scanning electron microscope (SEM) and theoretical calculations. A total of 46 compounds were identified, representing 89.89% of the overall chemical composition of AVU extract, including Edulisin III (72.88%), Binapacryl (4.32%), Khellin (1.97%), and Visnagin (1.65%). Chemical (Weight loss) and electrochemical (potentiodynamic polarization curves (PPC), and electrochemical impedance spectroscopy (EIS)) techniques revealed that investigated extract can be used as an effective corrosion inhibitor for carbon steel in 1.0 mol/L HCl solution. At a low dose of 700 ppm, the inhibitory action of AVU extract reached an inhibition efficiency of 84 percent. According to polarization tests, the investigated extract worked as a mixed inhibitor, protecting cathodic and anodic corrosion reactions. The EIS test showed that upon the addition of AVU extract to HCl solution, the polarization resistance increased while the double layer decreased. SEM images showed a protected CS surface in the inhibited solution. Quantum chemical calculations by Density Functional Theory (DFT) for the main components confirmed the major role of heteroatoms and aromatic rings as adsorption sites. Molecular dynamics (MD) simulation was used to study the adsorption configuration of the main components on the Fe(1 1 0) surface. Outcomes from this study further confirmed the significant advantage of using plant-based corrosion inhibitors for protecting metals and alloys.  相似文献
