An electrophoretic microchip integrated with a Pd-film decoupler and a series-dual electrode was proven practical (200-800 V/cm) for routine amperometric detection. In fluidic systems, amperometric enhancement of parallel-opposed dual-electrode detection is due to redox cycling of analytes between the electrodes. We, however, found that the oxidation current of catecholamines was enhanced significantly (1.9-3.8 folds) by switching from the single electrode mode to dual-series mode. This novel finding was unexpected because the unidirectional flow characteristic of the microfluidic system should eliminate the possibility for analytes physically migrating back and forth between the upstream and downstream electrodes. We attribute the enhancement to turbulence generated by impinging of the flow onto the edge of the downstream electrode. The linear range, sensitivity, limit of detection (S/N = 3) and number of theoretical plates for DA and CA are, respectively, 0.5-50 microM, 47 pA/microM, 0.25 microM, 7000 m(-1) and 1.0-100 microM, 28 pA/microM, 0.49 microM, 15,000 m(-1).  相似文献

This paper gives an overview of some of the recent developments in the area of chip calorimetry. Using several chip calorimeters developed at Xensor Integration and tested by users, examples of chip calorimetry and its applications will be given. Examples of chip calorimeters developed at various universities are given to widen the overview. The coupling of a capillary electrophoresis (CE) microchip with electrochemical detection with a chemometric approach was used for the classification of wines. Cabernet Sauvignon wine samples from 3 different geographical regions were used for verifying the discriminating power of the microchip electrophoretic technique using high separation voltage (3000 V) with analysis time of only 60 seconds. Electrophoretic data were transformed into initial 14 variables, concerning peak's area and peak's height. Principal component analysis was applied to optimize the information given by the electrophoretic data sets. Results of classification using LDA and CART were promising and the preliminary model was able to classify samples on the basis of their geographical origin. While the coupling of microchips with chemometrics approaches is illustrated for a former tentative of the classification of wines it could be readily extended to a wide range of other important applications.  相似文献

将刻有微沟道的芯片固定在旋转圆盘电极上,在旋转对流条件下于微沟道中电沉积铜.微沟道深度为1μm,宽度分别为0.35μm,0.50μm,0.70μm.研究了芯片的旋转、电流密度以及Cu2+浓度等对微沟道中铜沉积的影响.实验表明,在旋转对流传质下,铜在微沟道中的沉积速率比静止芯片时的约快2~3倍.较低的Cu2+浓度和适中的沉积电流密度更有利于超等厚沉积的形成.  相似文献

考察了聚二甲基硅氧烷(Polydimethylsiloxane,PDMS)预聚体与固化剂间的配比、固化温度及固化时间对PDMS芯片封接强度的影响,得出PDMS芯片封接的最佳条件基片和盖片所用PDMS预聚体与固化剂质量配比分别为10∶1与5∶1,固化温度为75℃,固化时间分别为35~50min和25~40min,封接后继续加热60min.在该条件下封接制作的微芯片历经半年50多次的分析、冲洗及抽液后未见明显损坏,足以满足一般分析任务的要求,并将芯片成功用于两种氨基酸的快速毛细管电泳分离.  相似文献

A simple and powerful microfluidic array chip-based electrophoresis system, which is composed of a 3-D microfluidic array chip, a microvacuum pump-based negative pressure sampling device, a high-voltage supply and an LIF detector, was developed. The 3-D microfluidic array chip was fabricated with three glass plates, in which a common sample waste bus (SW(bus)) was etched in the bottom layer plate to avoid intersecting with the separation channel array. The negative pressure sampling device consists of a microvacuum air pump, a buffer vessel, a 3-way electromagnet valve, and a vacuum gauge. In the sample loading step, all the six samples and buffer solutions were drawn from their reservoirs across the injection intersections through the SW(bus) toward the common sample waste reservoir (SW(T)) by negative pressure. Only 0.5 s was required to obtain six pinched sample plugs at the channel crossings. By switching the three-way electromagnetic valve to release the vacuum in the reservoir SW(T), six sample plugs were simultaneously injected into the separation channels by EOF and electrophoretic separation was activated. Parallel separations of different analytes are presented on the 3-D array chip by using the newly developed sampling device.  相似文献

Analytical potentialities of a chip-based CE in determination of ammonium in wastewaters were investigated. CZE with the electric field and/or ITP sample stacking was performed on a column-coupling (CC) chip with integrated conductivity detectors. Acetate background electrolytes (pH ~3) including 18-crown-6-ether (18-crown-6) and tartaric acid were developed to reach rapid (in 7-8 min) CZE and ITP-CZE resolutions of ammonium from other cations (sodium, potassium, calcium and magnesium) present in wastewater samples. Under preferred working conditions (suppressed hydrodynamic flow (HDF) and EOF on the column-coupling chip), both the employed methods did provide very good repeatabilities of the migration (RSD of 0.2-0.8% for the migration time) and quantitative (RSD of 0.3-4.9% for the peak area) parameters in the model and wastewater samples. Using a 900-nL sample injection volume, LOD for ammonium were obtained at 20 and 40 μg/L concentrations in CZE and ITP-CZE separations, respectively. Very good agreements of the CZE and ITP-CZE determinations of ammonium in six untreated wastewater samples (only filtration and dilution) with the results obtained by a reference spectrometric method indicate a very good accuracy of both the CE methods presented.  相似文献
