In this paper, fluorescence correlation spectroscopy (FCS) is used for investigation of homogeneous immune reaction using synthetic peptide as antigen. The binding process of CA125 peptide antigen and its antibody was systematically investigated. The dissociation constant and dissociation rate for antigen-antibody complex were determined, which were kdiss = 0.94 ± 0.05 nM and koff = 0.00215 ± 0.0001 s−1, respectively. Under optimal conditions, the detection limit of the competitive immunoassay was 4 × 10−10 M (S/N = 3). The good recoveries were obtained with human serum samples. Our preliminary results demonstrated that the homogeneous competitive immunoassay based on FCS is simple, rapid, sensitive and small sample and reagent requirement, and this method maybe possess great potential applications in clinical diagnosis, food and environmental analyses and biological and biomedical studies. 相似文献

Electrochromic properties of chemically bath deposited nanoporous NiO thin films were investigated as a function of film thickness using Ni sulphate precursor, aqueous ammonia and potassium persulphate as complexing and oxidizing agents respectively. The films were characterized for their structural, morphological, optical and electrochromic properties using X-ray diffraction, scanning electron microscopy, FT-IR spectroscopy, cyclic voltammetry, chronoamperometry and optical transmittance studies. X-ray diffraction patterns show that the films are polycrystalline, consisting of NiO cubic phase. Infrared spectroscopy results show the presence of free hydroxyl ion and water in NiO thin films. SEM micrographs revealed nanoporous nature composed of interconnected nanoporous network, forming well defined 3D nano envelopes. The optical band gap energy was found to be decreased from 3.22 to 2.80 eV with increasing film thickness. The electrochromic properties of all the films were investigated in aqueous (KOH) and non aqueous (LiClO4-PC) electrolyte by means of cyclic voltammetry (CV), chronocoulometry (CC) and optical studies. The transmittance modulations or optical density differences during the coloring/bleaching process were found to be increased with the film thickness. This increment in optical differences led to an increase in coloration efficiency (CE) to about 95 cm2/C, which is two times more than that observed in KOH and response time of 2.9 s for bleaching (reduction) and 3.5 s for coloration (oxidation) observed for the film deposited at 60 min with excellent electrochemical stability up to 3000 c/b cycles in LiClO4-PC electrolyte. 相似文献

Modeling walking behavior of pedestrian groups with floor field cellular automaton approach 下载免费PDF全文
Walking in groups is very common in a realistic walking environment. An extended floor field cellular automaton(CA)model is therefore proposed to describe the walking behavior of pedestrian groups. This model represents the motion of pedestrian groups in a realistic way. The simulation results reveal that the walking behavior of groups has an important but negative influence on pedestrian flow dynamics, especially when the density is at a high level. The presence of pedestrian groups retards the emergence of lane formation and increases the instability of operation of pedestrian flow. Moreover,the average velocity and volume of pedestrian flow are significantly reduced due to the group motion. Meanwhile, the parameter-sensitive analysis suggests that pedestrian groups should make a compromise between efficient movement and staying coherent with a certain spatial structure when walking in a dense crowd. 相似文献

Treatment of 1,2-(E,E)-di(1-buta-1,3-dienyl)cyclohexanol (21) with potassium hydride in tetrahydrofuran at room temperature resulted in the facile formation of a 14-membered ring dienolate which on kinetic protonation provided cyclotetradeca-3,5,7-trien-1-one. This novel rearrangement which provides the basis for an efficient, eight unit ring expansion method was also observed when 5,8-dimethyl-5-hydroxy-1,3,7,9-decapentaene (28), the acyclic analogue of 21, was treated with potassium hydride in tetrahydrofuran. The effects of dissolved electrolytes on the structure of liquid formamide have been investigated by Raman spectroscopy. The spectral features are dependent on the concentration and nature of the dissolved electrolyte and are discussed in terms of direct electrolyte formamide interactions. Two vN—H bands, arising from ion—formamide species, have been observed superimposed on the spectrum of residual liquid formamide. The results lend further support to the interaction model for electrolytes in liquid formamide previously proposed by us on the basis of nuclear magnetic resonance and infrared spectral data. 相似文献
