Opuntia dillenii Ker Gawl. is one of the medicinal plants used for the prevention and treatment of diabetes mellitus (DM) in Morocco. This study aims to investigate the antihyperglycemic effect of Opuntia dillenii seed oil (ODSO), its mechanism of action, and any hypoglycemic risk and toxic effects. The antihyperglycemic effect was assessed using the OGTT test in normal and streptozotocin (STZ)-diabetic rats. The mechanisms of action were explored by studying the effect of ODSO on the intestinal absorption of d-glucose using the intestinal in situ single-pass perfusion technique. An Ussing chamber was used to explore the effects of ODSO on intestinal sodium-glucose cotransporter 1 (SGLT1). Additionally, ODSO’s effect on carbohydrate degrading enzymes, pancreatic α-amylase, and intestinal α-glucosidase was evaluated in vitro and in vivo using STZ-diabetic rats. The acute toxicity test on mice was performed, along with a single-dose hypoglycemic effect test. The results showed that ODSO significantly attenuated the postprandial hyperglycemia in normal and STZ-diabetic rats. Indeed, ODSO significantly decreased the intestinal d-glucose absorption in situ. The ex vivo test (Ussing chamber) showed that the ODSO significantly blocks the SGLT1 (IC50 = 60.24 µg/mL). Moreover, ODSO indu\ced a significant inhibition of intestinal α-glucosidase (IC50 = 278 ± 0.01 µg/mL) and pancreatic α-amylase (IC50 = 0.81 ± 0.09 mg/mL) in vitro. A significant decrease of postprandial hyperglycemia was observed in sucrose/starch-loaded normal and STZ-diabetic ODSO-treated rats. On the other hand, ODSO had no risk of hypoglycemia on the basal glucose levels in normal rats. Therefore, no toxic effect was observed in ODSO-treated mice up to 7 mL/kg. The results of this study suggest that ODSO could be suitable as an antidiabetic functional food.  相似文献

The wine industry generates a large amount of wastes, including grape marc and vinification lees. These substances can be used to produce enzymes or other food additives. Composting could be a successful strategy for the sustainable and complete recycling of grape marc. In this work, the mesophilic biodegradation of grape marc during 60 d under microaerobic conditions using several ratios of seeds, skin, and stem were studied. The presence of Penicillium spp. was detected at the beginning of the composting. Changes in chemical and biological parameters were evaluated. Biodegraded grape marc with stem showed the best organic matter properties (C/N ratio of 14 and N content of 37 g/kg) and a germination index of 155% for the growth of ray grass seeds. The results suggest that the biodegraded of grape marc could be used as fertilizer, especially for ray grass crops.  相似文献

以丝光沸石(MOR)分子筛为催化剂,甲苯与三甲苯烷基转移反应为探针反应,系统考察了晶种数量变化对分子筛物化性能和催化性能的影响。研究表明,合成过程中晶种添加量的不同会显著影响催化剂的酸性、比表面积和孔体积,进而影响其催化剂活性和稳定性。当晶种添加量为8%时,MOR分子筛具有最多的B酸含量、最大的比表面积和孔体积,同时对应的催化剂活性和稳定性也最好。本研究还对甲苯和三甲苯烷基转移反应机理进行了深入研究,结果表明,发生在MOR分子筛上的烷基转移反应通过双分子中间体机理进行,并应用实验手段捕捉到了中间体的存在,证实了双分子中间体机理的合理性,同时推导出可能的反应机理路线图。  相似文献

发展了离线二维反相液相色谱/超临界流体色谱(2D RPLC/SFC)分离瓜蒌子的方法。实验在第一维采用反相色谱,按色谱峰收集从瓜蒌子样品中制备得到的12个组分(F_1~F_(12)),并将得到的组分在第二维使用SFC分离。这些组分在RPLC和SFC的分离对比说明,该二维方法具有良好的分离正交性,可至少检测到150个色谱峰,对于解决结构相似物质的分离、微量成分的富集表现出了明显的优势。SFC方法采用了乙醇-正己烷(3∶7,v/v)的混合溶剂作为改性剂,既提供了适当的洗脱能力,也保证了在上样量增加时满足样品溶解的要求。此二维分离体系可放大到制备水平用于化合物的制备,为瓜蒌子化学成分的纯化制备提供技术支持,为其物质基础研究提供参考。  相似文献

Ricin, (Ricinus communis agglutinin, RCA) is one of the most poisonous of naturally occurring substances and has great potential for bioterrorism because no antidote exists. Fast detection at low concentrations is a challenge, and vital to the development of proper countermeasures. In this study, a square wave adsorptive stripping voltammetric (SWAdSV) method for determining RCA using a cathodically polarized boron‐doped diamond (BDD) electrode is presented. An irreversible electrochemical RCA oxidation peak was identified on the BDD electrode by different voltammetric techniques using both direct and adsorptive stripping modes. An adsorption‐controlled (slope log Ip vs log v of 0.80) pH‐dependent process was observed. For values of 1.0≤pH≤9.0, the numbers of protons and electrons associated with the oxidation reaction were estimated (ca. 1.0) by differential pulse voltammetry. The RCA oxidation step may correspond to the oxidation of tryptophan amino acid residues, and occurs in a complex mechanism. The excellent analytical performance of the cathodically polarized BDD electrode in combination with the stripping mode ramp was verified with RCA by using a short deposition time in an open circuit potential (120 s). Under optimized analysis conditions, a linear response in the range of (3.3–94.0)×10−9 mol L−1 (r2=0.9944) and a limit of detection of 6.2×10−10 mol L−1 were estimated. This LOD is lower than several methods found in the literature. For example, it is 168 times lower than that obtained by using square wave voltammetric with a glassy carbon electrode. Moreover, an even lower LOD might be achieved by using the SWAdSV method with a higher pre‐concentration time. In addition, trace levels of RCA were successfully determined in different castor seed cultivars with an overall average recovery from 99.2±1.6 % for the three different RCA‐A concentration levels. The high accuracy of the analytical data highlights the use of the proposed method for determining RCA in other samples.  相似文献

In this study, essential oils from Voacanga africana seeds at different extraction stages were investigated. In the chemical composition analysis, 27 compounds representing 86.69–95.03% of the total essential oils were identified and quantified. The main constituents in essential oils were terpenoids, alcohols and fatty acids accounting for 15.03–24.36%, 21.57–34.43% and 33.06–57.37%, respectively. Moreover, the analysis also revealed that essential oils from different extraction stages possessed different chemical compositions. In the antioxidant evaluation, all analysed oils showed similar antioxidant behaviours, and the concentrations of essential oils providing 50% inhibition of DPPH-scavenging activity (IC50) were about 25 mg/mL. In the antimicrobial experiments, essential oils from different extraction stages exhibited different antimicrobial activities. The antimicrobial activity of oils was affected by extraction stages. By controlling extraction stages, it is promising to obtain essential oils with desired antimicrobial activities.  相似文献

种子纯度反映种子品种在特征特性方面典型一致的程度,提高种子纯度检测的准确性和可靠性对保证种子的质量具有重要的意义。高光谱图像技术可以同时反映种子的内部特征和外部特征,在农产品无损检测中已经得到广泛应用。利用近红外高光谱图像实现农产品无损检测的实质就是建立光谱信息与农产品品质参数之间的数学模型关系。但光谱信息易受环境、时间的影响,当待测样本的产地或者年份发生改变时光谱信息也随之改变,导致建立的模型的稳定性变差、泛化能力减弱。针对这一问题,采用主动学习算法选择具有代表性的待测样本,最终以添加最少最优的样本数来扩大原模型的样本空间,从而实现模型的快速更新,提高模型的稳定性,同时与基于随机选择算法(RS)和Kennard-Stone算法(KS)的模型更新效果进行比较。实验结果表明:在不同样本集划分比例下(1∶1, 3∶1, 4∶1),利用主动学习添加40个新样本更新后的2010年的玉米种子纯度检测模型对2011年新样本的预测精度由47%,33.75%,49%提高到98.89%,98.33%,98.33%;利用主动学习添加56个新样本更新后的2011年的玉米种子纯度检测模型对2010年新样本的预测精度由50.83%,54.58%,53.75%提高到94.57%,94.02%,94.57%;同时基于主动学习算法的模型更新效果明显优于RS和KS。因此基于主动学习算法实现玉米种子纯度检测模型的更新是可行的。  相似文献

微区无损分析可提供物质组成元素的原位分布信息,以揭示物质形成条件、元素动态分布过程与相互作用机理、生物代谢作用等。文章报道了实验室型微区X射线荧光(μXRF)光谱仪的研发和元素生物地球化学动态分布过程研究结果。μXRF光谱仪采用15 μm光斑的聚束毛细管X射线透镜为激发源,选用分辨率为135 eV的硅漂移探测器(SDD),样品和探测器间角度可调,使之可进行异型样品如地质样品的原位分析,利用五轴自控实现样品时空四维元素分布测定。利用该μXRF光谱仪测定了矿物-生物膜间的元素迁移和玉米种发芽过程中的元素分布,发现(1)生物膜可吸附、富集毒性元素铅,是重金属的重要汇集地,最大富集系数1.7。(2)生物膜是金属从固态矿物相经水相进入生态系统的重要途径。(3)在玉米种子中,可检测到K,Ca,Mn,Fe,Cu,Zn和Pb。Zn主要在胚乳中分布,胚中有少量Zn存在;在胚乳和胚中存在微量Fe;胚乳中存在微量Pb,胚中未观测到Pb。(4)经含Pb溶液浸泡发芽后,K在玉米种中胚和胚乳中部分富集,Fe分布在种皮和胚乳中,Cu和Zn主要在胚乳中分布;Pb主要在胚根、胚轴和胚芽中分布,且Pb在新生根中高度富集。研究表明,在种子萌发阶段,Pb等毒性元素可被植物滞留于根部,制约了其向地上部的转移,从而揭示了植物对毒性元素的耐受机制。  相似文献

以早熟品种“清风103”青花菜为试验材料,研究青花菜绿体春化过程中光合特性的动态变化,结果表明: 低温春化促进了青花菜的光合作用,青花菜植株叶片中总叶绿素质量比、净光合速率(Pn)、气孔导度(Gs)和胞间CO2摩尔比(Ci)均随着春化的进程而增加,说明春化可以积累更多的碳水化合物,为春化结束后的花芽分化提供更多的能量.  相似文献
