This work presents a sensitive voltammetric method for determination of the flavonoid baicalein by using a thermally reduced graphene oxide (TRGO) modified glassy carbon electrode (GCE) in 100 mM KCl‐10 mM sodium phosphate buffer solution (pH 7.40). The surface morphology and structure of TRGO investigated by atomic force microscopy, FT‐IR spectroscopy and Raman spectroscopy reveal that the TRGO prepared maintained as single or bilayer sheets and with significant edge‐plane‐like defect sites. The TRGO/GCE modified electrode shows more favorable electron transfer kinetics for potassium ferricyanide and potassium ferrocyanide probe molecules, which are important electroactive compounds, compared with bare GCE and GO/GCE electrodes. The electrochemical behaviors of baicalein at the TRGO/GCE were investigated by cyclic voltammetry, suggesting that the TRGO/GCE exhibits excellent electrocatalytic activity to baicalein. Under physiological conditions, the modified electrode showed linear voltammetric response from 10 nM to 10 µM for baicalein, with a detection limit of 6.0 nM. This work demonstrates that the graphene‐modified electrode is a promising tool for electrochemical determination of flavonoid drugs. 相似文献

以聚酰胺为柱层析吸附剂,以乙醇-氯仿混合溶剂梯度洗脱,从中药黄芩的甲醇提取液中分离出有效成分黄芩素的粗品。 通过对有效成分进行重结晶等纯化处理,得到了一种黄色短棱柱晶状产物。 利用红外光谱(FTIR)、紫外-可见光谱(UV-Vis)、质谱(MS)、氢谱(1H-NMR)及碳谱(13C-NMR)等分析测试手段,对产物的组成和结构进行了表征。 红外光谱测定结果表明,产物的结构中含有共轭芳香体系和单取代苯;紫外-可见光谱和质谱表明,产物为黄酮而非黄酮苷化合物,A环上含有三个酚羟基, B环无羟基和其他基团取代;核磁共振氢谱及碳谱表明,产物的结构中含15个碳原子, 三组羟基质子。 同时,文章还对所有1H-NMR和13C-NMR信号进行了归属,对FTIR光谱特征吸收峰所对应的官能团及其振动形式进行了指认,给出了化合物及碎片离子在质谱中的裂解方式,最终将黄色短棱柱晶状产物鉴定为5,6,7-三羟基黄酮,即黄芩素,分子式为C15H10O5。 相似文献

Glioblastoma multiforme (GBM) is the most aggressive malignant tumor of the brain. It has different glutamate receptor types. So, these receptors can be a suitable target for GBM treatment. The current study investigated the anticancer effects of bovine serum albumin (BSA)-Baicalein @Zn-Glu nanostructure mediated-GluRs in human glioblastoma U87 cells. BSA-Ba@Zn-Glu hybrid nanoparticles (NPs) were set and considered transporters for Baicalein (Ba) active compound delivery. BSA-Ba@Zn-Glu NPs were synthesized by a single-step reduction process. The successful production was confirmed through transmission electron microscopy (TEM), dynamic light scattering (DLS), Fourier transform infrared spectroscopy (FT-IR), and hemolysis test. The cytotoxic efficacy and apoptosis rate of the nanostructures on U87 glioblastoma cells were investigated by 3-(4,5-dimethylthialzol-a-yl)-2,5 diphenyltetrazolium bromide (MTT) and flow cytometry assays, respectively. The synthesized BSA-Ba@Zn-Glu nanostructures with a diameter of 142.40 ± 1.91 to 177.10 ± 1.87 nm and zeta potential of −10.57 ± 0.71 to −35.77 ± 0.60 mV are suitable for extravasation into tumor cells. The drug release from the BSA-Ba@Zn NPs showed controlled and pH-dependent behavior. In vitro results indicated that the BSA-Ba@Zn-Glu NPs significantly reduce cell viability and promote apoptosis of U87 cancer cells. It revealed the cytotoxic effect of the Baicalein and an increase in cellular uptake of nanoparticles by Glu receptors. Zn NPs were synthesized based on a green synthesis method. BSA NPs were used as a nano-platform for Glu conjugation and Ba drug delivery. BSA-Ba@Zn-Glu NPs induce cytotoxicity and apoptosis in human brain cancer cells (U87) in a dose-dependent manner. Finally, this nanostructure could be served in targeted drug delivery in vivo studies and applied along with other strategies such as X-ray irradiation as combinational therapies in future studies. 相似文献

A graphene‐based electrochemical sensing platform for sensitive determination of baicalein was constructed by means of pulsed potentiostatic reduction of graphene oxide (GO) on a glassy carbon electrode (GCE). The resulting electrode (ERGO/GCE) was characterized by cyclic voltammetry (CV) and scanning electron microscopy (SEM). The electrochemical behaviors of baicalein at the ERGO/GCE were investigated in detail by CV, chronoamperometry (CA) and chronocoulometry (CC). The experimental results demonstrated that the ERGO/GCE exhibited excellent response toward the redox of baicalein as evidenced by the significant enhancement of redox peak currents (ip) and the decreased peak‐to‐peak separation (ΔEp) in comparison with a bare GCE. Under the optimum experimental conditions, the reduction peak cureent was proportioanal to the baicalein concentration in the range of 5.0 × 10‐9 ~ 5.0 × 10‐7 mol L‐1 with the detection limit of 2.0 × 10‐9 mol L‐1. The proposed method was also applied successfully to determine baicalein in spiked human blood serum samples. 相似文献

A modified electrode Ni(II)-BA-MWCNT-PE has been fabricated by electrodepositing nickel(II)-baicalein [Ni(II)-BA] complex on the surface of multi-wall carbon nanotube paste electrode (MWCNT-PE) in alkaline solution. The Ni(II)-BA-MWCNT-PE exhibits the characteristic of improved reversibility and enhanced current responses of the Ni(III)/Ni(II) couple compared with Ni(II)-BA-CPE. It also shows good electrocatalytic activity toward the oxidation of hydrazine. Kinetic parameters such as the electron transfer coefficient α, rate constant ks of the electrode reaction, the diffusion coefficient D of hydrazine and the catalytic rate constant kcat of the catalytic reaction are determined. Moreover, the catalytic currents present linear dependence on the concentration of hydrazine from 2.5 μM to 0.2 mM by amperometry. The detection limit and sensitivity are 0.8 μM and 69.9 μA mM−1, respectively. The modified electrode for hydrazine determination is of the property of simple preparation, good stability, fast response and high sensitivity. 相似文献

The present paper describes to modify a double stranded DNA-octadecylamine (ODA) Langmuir-Blodgett film on a glassy carbon electrode (GCE) surface to develop a voltammetric sensor for the detection of trace amounts of baicalein. The electrode was characterized by atomic force microscopy (AFM) and cyclic voltammetry (CV). Electrochemical behaviour of baicalein at the modified electrode had been investigated in pH 2.87 Britton-Robinson buffer solutions by CV and square wave voltammetry (SWV). Compared with bare GCE, the electrode presented an electrocatalytic redox for baicalein. Under the optimum conditions, the modified electrode showed a linear voltammetric response for the baicalein within a concentration range of 1.0 × 10−8-2.0 × 10−6 mol L−1, and a value of 6.0 × 10−9 mol L−1 was calculated for the detection limit. And the modified electrode exhibited an excellent immunity from epinephrine, dopamine, glucose and ascorbic acid interference. The method was also applied successfully to detect baicalein in the medicinal tablets and spiked human blood serum samples with satisfactory results. 相似文献

The effects of the different combined manner of ultrasound and covalent reaction between polyphenol and myofibrillar protein (MP) from chicken were studied. More so, antioxidant activities, digestive properties, and potential mechanism of ultrasound-assisted oxidation system of hydrophilic ((−)-Epicatechin gallate, ECG) and hydrophobic (Baicalein, BN) polyphenols was also analyzed in this study. Among all the combined treatments, surface hydrophobicity (SUH), active sulfhydryl contents (ASC), and specific surface area (SSA) of ultrasonic assisted ECG oxidation group (T6) was relatively apparent, indicating that a more unfolding MP structure was obtained. Furthermore, ultrasonic assisted ECG oxidation group showed the highest antioxidant activities compared with other combined treatments on the basis of the results of DPPH free radical scavenging activities, metal ion chelating activities, and hydroxyl radicals (OH·) scavenging activities. The results of simulated digestion system and kinetic analysis also verified that ultrasonic assisted ECG oxidation had higher MP bio-accessibility than the control group. In contrast, a lower digestibility was displayed in ultrasonic assisted BN oxidation group. In summary, the ultrasound-assisted covalent reaction of MP and ECG might be a desirable approach for industrial production of MP from chicken with better antioxidant activities and digestive properties. 相似文献
