The viscosity of PEG-modified urethane acrylate (PMUA) showed peculiar behavior in the course of soap-free emulsification. Moreover, the viscosity change with added amounts of water was influenced by the reaction molar ratio of polyethylene glycol (PEG). The rate of increase in viscosity slowed and the ratio of increase in viscosity increased as the reaction molar ratio of PEG increased. This peculiar viscosity behavior was due to the microphase separation between hydrophilic and hydrophobic segments of PMUA, and the orientation of polyoxyethylene groups at O/W interface which influenced droplet size of the soap-free PMUA emulsion. The location of polyoxyethylene groups of this resin at O/W interface was confirmed using the adsorption isotherm measurement of PMUA molecules containing polyoxyethylene groups at water/benzene interface. The microphase separation behavior of PMUA between hydrophilic and hydrophobic segments could apply to the preparation of the PMUA gels containing peculiar structure. PMUA gels were prepared using dioxane (UAG) and the swelling behavior of these gels were compared to that of gels prepared using water (UAHG) in the same medium. In the same medium, the swelling behavior of UAHG gels differed from that of UAG gels because of the difference in the microstructure of gel due to the microphase separation between hydrophilic and hydrophobic segments. This phase separation in the course of gelation in water could be confirmed using contact angle measurement.  相似文献

A study has been undertaken of stress relaxation in ovalbumin thermotropic gels with a concentration of 8–20%, depending on time and temperature of heating (respectively, 20–60 min, 70°–110°C), at pH 2.5–10.0. In all instances, the dependence of the initial gel elasticity modulus on heating has a single maximum. Gelation conditions corresponding to this maximum are considered optimal. Optimal gelation time is 30 min, regardless of pH. On the other hand, the optimal heating temperature depends on pH. To the right and left of the isoelectric point of protein (2.5pH<4.0 and 5.5G) of gels on heating conditions, pH and protein concentration (X 1,X 2,X 3,X 4), as well as on time of relaxation (t) may be generally described asG(X 1,X 1,X 1,X 1,t)=G e(X 1,X 2,X 3,X 4)f(t), whereG e is the equilibrium value of the elasticity modulus, and f(t) the relaxation function. Thus, a change in the parameters only affects the value of the equilibrium elasticity modulus, and exerts no effect on the relaxation time spectrum. For this reason, all the relaxation curves obtained may be transformed into two normalized relaxation functions:f(t)=f(t)/f(1)=G(X 1,X 2,X 3,X 4,t)/G(X 1,X 2,X 3,X 4, 1)Each of these normalized functions corresponds to one of the homologous groups. Rheological similarity of gels in each homologous group evidently points to their structural similarity. Invariance of the gel relaxationproperties with regard to protein concentration, leads to a concentration dependence of the equilibrium modulus at various pH values. These dependences are curvilinear on a double logarithmic scale. The slope of the curve exceeds 2 in the entire concentration interval studied. In other words, the dependences obtained cannot be described by the usual law of squares. On the other hand, they adequately match Hermans theoretical relation for a network formed by random association of identical polyfunctional particles without cyclization. This simple model evidently gives a true picture of the major regularities of thermotropic gelation for ovalbumin. An agreement between this theory and experiment was achieved for a protein concentration ofC *=6.0±1.0% at the gel point regardless of pH. A review is given on the main problems associated with the determination and interpretation of molecular diffusion in zeolites. It is shown that the diffusivities may most decisively depend on the relevant space and time scales of observation, as well as on the physical state under which the measurements are carried out. While there is a considerable interest in the food industry in determining various analytes using ion-selective electrodes (ISEs), only few reports describe their use for direct measurements in food. In this study, the suitability of glass electrodes and ionophore-based solvent polymeric ISEs for the determination of pH in Process cheese, Cheddar cheese and milk was investigated. The liquid junction potential between a 3 M KCl bridge electrolyte and diluted as well as undiluted Process cheese was found to be negligible. Reference electrodes with ceramic plug and sleeve-type junctions performed well, although precautions needed to be taken to prevent plugging at the junctions. While the protein rennet casein posed no problems in pH measurements, the extraction of neutral lipophilic compounds or hydrophobic peptides into solvent polymeric membranes was evident, resulting in some loss of selectivity for monovalent cations upon exposure to cheese. However, it was found that ISEs based on tridodecylamine (R3N) as ionophore and o-nitrophenyl octyl ether (oNPOE) as plasticizer can be used to accurately measure the pH of milk and, after desensitization of the electrodes in a cheese emulsion, of diluted Process cheese. Since pH measurements with a glass electrode showed that emulsions of cheese moderately diluted to a cheese content of 70% have the same pH as undiluted cheeses, it is possible to determine the pH in cheese with ionophore-based ISEs. R3N membranes also performed well in undiluted milk.  相似文献

A knowledge-based method for calculating the similarity of functional groups is described and validated. The method is based on experimental information derived from small molecule crystal structures. These data are used in the form of scatterplots that show the likelihood of a non-bonded interaction being formed between functional group A (the `central group') and functional group B (the `contact group' or `probe'). The scatterplots are converted into three-dimensional maps that show the propensity of the probe at different positions around the central group. Here we describe how to calculate the similarity of a pair of central groups based on these maps. The similarity method is validated using bioisosteric functional group pairs identified in the Bioster database and Relibase. The Bioster database is a critical compilation of thousands of bioisosteric molecule pairs, including drugs, enzyme inhibitors and agrochemicals. Relibase is an object-oriented database containing structural data about protein-ligand interactions. The distributions of the similarities of the bioisosteric functional group pairs are compared with similarities for all the possible pairs in IsoStar, and are found to be significantly different. Enrichment factors are also calculated showing the similarity method is statistically significantly better than random in predicting bioisosteric functional group pairs.  相似文献

Electric cell-substrate impedance sensing (ECIS) was applied to assess the structure-function of α2β1 integrin, receptor for collagen and laminin. On collagen-coated gold electrodes, expression of this integrin on human rhabdomyosarcoma (RD) cells (RDX2C2) yielded a five-fold increase in resistance when compared with mock transfected RD (RDpF) cells (34.5±5.2 versus 6.5±0.8 Ω/cell). An intermediate level of 16±2 Ω/cell was measured upon expression of an α2β1 mutant that lacked the α2 cytoplasmic domain (RDX2CO). On laminin, the resistance measured for RDX2C2 cells was also higher but only twice that of RDpF cells at 71±4 and 37±4 Ω/cell, respectively. In comparison, RDX2CO cells (38±4 Ω/cell), exhibiting no enhanced adhesive function, yielded a similar result to that of RDpF cells. On fibronectin, RDX2C2 and RDpF cells, exhibiting comparable levels of adhesion, were similar in resistance measurements at 85±5and 89±7 Ω/cell, respectively. It has been shown that deletion of α2 cytoplasmic domain results in dysregulated recruitment of the α2β1 mutant to focal adhesion complexes that mediate binding of fibronectin. RDX2CO cells on fibronectin, exhibiting reduced adhesive function, was associated with noticeably lower resistance (60±4 Ω/cell). Monitoring electroporation of the RD plasma membrane also indirectly validated cell attachment as reflected by the resistance measured. Results from this study demonstrated the potential of ECIS for study of the structure-function of βl integrin adhesion receptors.  相似文献
