Biosynthesis of poly-3-hydroxybutyrate with a high molecular weight by methanotroph from methane and methanol

Poly-3-hydroxybutyrate (PHB) can be produced by various species of bacteria. Among the possible carbon sources, both methane and methanol could be a suitable substrate for the production of PHB. Methane is cheap and plentiful not only as natural gas, but also as biogas. Methanol can also maintain methanotrophic activity in some conditions. The methanotrophic strain Methylosinus trichosporium IMV3011 can accumulate PHB with methane and methanol in a brief nonsterile process. Liquid methanol (0.1%) was added to improve the oxidization of methane. The studies were carried out using shake flasks. Cultivation was performed in two stages: a continuous growth phase and a PHB accumulation phase under the conditions short of essential nutrients (ammonium, nitrate, phosphorus, copper, iron (Ⅲ), magnesium or ethylenediamine tetraacetate (EDTA)) in batch culture. It was found that the most suitable growth time for the cell is 144 h. Then an optimized culture condition for second stage was determined, in which the PHB concentration could be much increased to 0.6 g/L. In order to increase PHB content, citric acid was added as an inhibitor of tricarboxylic acid cycle (TCA). It was found that citric acid is favorable for the PHB accumulation, and the PHB yield was increased to 40% (w/w) from the initial yield of 12% (w/w) after nutrient deficiency cultivation. Lattice rules with the trigonometric d-property that are optimal with respect to the number of points are constructed for the approximation of integrals over an n-dimensional unit cube. An extreme lattice for a hyperoctahedron at n = 4 is used to construct lattice rules with the trigonometric d-property and the number of points (d = 2Δ ? 1 ≥ 3 is an arbitrary odd number). With few exceptions, the resulting lattice rules have the highest previously known effectiveness factor.

In this paper, we study the p-ary linear code Ck(n,q), q=ph, p prime, h1, generated by the incidence matrix of points and k-dimensional spaces in PG(n,q). For kn/2, we link codewords of Ck(n,q)Ck(n,q) of weight smaller than 2qk to k-blocking sets. We first prove that such a k-blocking set is uniquely reducible to a minimal k-blocking set, and exclude all codewords arising from small linear k-blocking sets. For k<n/2, we present counterexamples to lemmas valid for kn/2. Next, we study the dual code of Ck(n,q) and present a lower bound on the weight of the codewords, hence extending the results of Sachar [H. Polyethylenimine (PEI) is a commonly used cationic polymer for small-interfering RNA (siRNA) delivery due to its high transfection efficiency at low commercial cost. However, high molecular weight PEI is cytotoxic and thus, its practical application is limited. In this study, different formulations of low molecular weight PEI (LMW-PEI) based copolymers polyethylenimine-g-polycaprolactone (PEI–PCL) (800 Da–40 kDa) and PEI–PCL–PEI (5–5–5 kDa) blended with or without polyethylene glycol-b-polycaprolactone (PEG–PCL) (5 kDa-4 kDa) are investigated to prepare nanoparticles via nanoprecipitation using a solvent displacement method with sizes ≈100 nm. PEG–PCL can stabilize the nanoparticles, improve their biocompatibility, and extend their circulation time in vivo. The nanoparticles composed of PEI–PCL–PEI and PEG–PCL show higher siRNA encapsulation efficiency than PEI–PCL/PEG–PCL based nanoparticles at low N/P ratios, higher cellular uptake, and a gene silencing efficiency of ≈40% as a result of the higher molecular weight PEI blocks. These results suggest that the PEI–PCL–PEI/PEG–PCL nanoparticle system could be a promising vehicle for siRNA delivery at minimal synthetic effort. 相似文献

For quantum fluids governed by the compressible quantum Navier-Stokes equations in $\Bbb R^3$ with viscosity and heat conduction, we prove the optimal $L^p-L^q$ decay rates for the classical solutions near constant states.
