It is shown that pipe-flow turbulence consists of transients. The fractal dimensions of the dynamical process are thus all zero. Nevertheless, this is compatible with Grassberger-Procaccia analyses suggesting the existence of a high-dimensional strange attractor. The availability of sufficient quantities of DNA of adequate quality is crucial in polymerase chain reaction (PCR)-based methods for genetically modified food detection. In this work, the suitability of anion-exchange CIM (Convective Interaction Media; BIA Separations, Ljubljana, Slovenia) monolithic columns for isolation of DNA from food was studied. Maize and its derivates corn meal and thermally pretreated corn meal were chosen as model food. Two commercially available CIM disk columns were tested: DEAE (diethylaminoethyl) and QA (quaternary amine). Preliminary separations were performed with standard solution of salmon DNA at different pH values and different NaCl concentrations in mobile phase. DEAE groups and pH 8 were chosen for further isolations of DNA from a complex matrix-food extract. The quality and quantity of isolated DNA were tested on agarose gel electrophoresis, with UV-scanning spectrophotometry, and by amplification with real-time PCR. DNA isolated in this way was of suitable quality for further PCR analyses. The described method is also applicable for DNA isolation from processed foods with decreased DNA content. Furthermore, it is more effective and less time-consuming in comparison with the existing proposed methods for isolation of DNA from plant-derived foods. 相似文献

Using Monte Carlo molecular dynamics, a new, careful study is made of the approach of the trajectory of a typical particle in a hard sphere fluid to that of a Brownian particle, discussed before by Powles and Quirke and Rapaport. The impedance of zinc-rich polymer coatings on steel in a 3-% NaCl solution is studied. The electrochemical behavior of the electrode is satisfactorily described by an equivalent circuit that contains a constant-phase element, which reflects fractal properties of the zinc surface in the binder. From the results of calculations of the circuit elements for metal-rich electrodes with different zinc contents computed are fractions of the active area of the surface of the coating and its fractal dimensionality. 相似文献

Hydrogen peroxide in basic media is proposed as a means for dissolving whole blood samples to be analyzed by electrothermal atomization atomic absorption spectrometry, ET AAS. Approximately 2 g of the whole blood sample were directly weighed in a 150 mL volumetric flask; 3 mL of a NaOH 0.2 mol L−1 solution, two drops of 1-octanol, as an antifoaming agent, and 1 mL of 30% volume hydrogen peroxide were added to the flask to promote oxidation. The solution was then manually shaken and after approximately three minutes of shaking, a clear solution, with no apparent suspended solids or greasy layers, was obtained. Distilled-deionized water was used to complete the volume. Ten μL of the resulting solution along with 10 μL of a solution containing 5000 mg L−1 of NH4H2PO4 and 300 mg L−1 of Mg(NO3)2 as a modifier, were injected into transversely heated graphite tubes for lead determination. Both aqueous standards and standard addition calibration curves produced results not significantly different at a 95% confidence limit level. Accuracy of the measurements was assessed by analysis of the IAEA A-13 (concentration of trace and minor elements in freeze dried animal blood) standard reference material containing 0.18 mg L−1 lead on a dry basis and by means of recovery tests. Analysis of the IAEA A-13 standard produced 0.17 ± 0.02 mg L−1 lead on a dry basis; recovery tests afforded values from 95 to 105%. Ten consecutive measurements of a 5 ppb lead solution gave a characteristic mass of 47.2 pg and a (3S) detection limit of 1.77 μg L−1 Pb. Results obtained from analysis of whole blood samples of volunteer donors covered a lead concentration range between 8 and 21 μg L−1 with a mean value of 11.9 ± 4.7 μg L−1. 相似文献

The activity of cellulase has traditionally been described by pH and temperature; however, the buffering medium is also an important factor, Taking plain water as a reference medium, three kinds of buffer including KH2PO4/K5HPO4, citric acid/sodium citrate, and acetic acid/sodium acetate were adopted to survey their effects on the activity of cellulase. Chromatographic assays indicated that xylose, glucose, and cellobiose were the major products and that minor products such as cellotriose and cellotetraose were present in some cases. The activities of cellulase based on glucose production showed that the phosphate buffer acted as a deactivator for cellulase and each of the two organic acid buffers acted as activators for cellulase. The concentration of activation buffer should be high to reach a high cellulase activity; however, this effect would be compensated for by the product inhibition of cellulase. The highest activity obtained was 4.16 ± 0.08 (× 10?3) IU mg?1 for the citric acid/sodium citrate buffer under pH 4.80, 40 °C and an agitation speed of 150 rpm. 相似文献
