A sample preparation method that combines a modified target plate with a nanoscale reversed-phase column (nanocolumn) was developed for detection of neuropeptides by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). A gold-coated MALDI plate was modified with an octadecanethiol (ODT) self-assembled monolayer to create a hydrophobic surface that could concentrate peptide samples into a approximately 200-500-microm diameter spot. The spot sizes generated were comparable to those obtained for a substrate patterned with 200-microm hydrophilic spots on a hydrophobic substrate. The sample spots on the ODT-coated plate were 100-fold smaller than those formed on an unmodified gold plate with a 1-microl sample and generated 10 to 50 times higher mass sensitivity for peptide standards by MALDI-TOF MS. When the sample was deposited on an ODT-modified plate from a nanocolumn, the detection limit for peptides was as low as 20 pM for 5-microl samples corresponding to 80 amol deposited. This technique was used to analyze extracts of microwave-fixed tissue from rat brain striatum. Ninety-eight putative peptides were detected including several that had masses matching neuropeptides expected in this brain region such as substance P, rimorphin, and neurotensin. Ir catalyst possesses a good electrocatalytic activity and selectivity for the oxidation of NH3 and/or NH4OH at Ir anode in the potential fixed electrochemical sensor with the neutral solution. Owing to the same electrochemical behavior of NH3 and NH4OH in a NaClO4 solution, NH4OH can be used instead of NH3 for the experimental convenience. It was found that the potential of the oxidation peak of NH4OH at the Ir/GC electrode in NaClO4 solutions is at about 0.85 V, and the current density of the oxidation peak of NH4OH is linearly proportional to the concentration of NHaOH. The electrocatalytic oxidation of NH4OH is diffusion-controlled. Especially, Ir has no electrocatalytic activity for the CO oxidation, illustrating that CO does not interfere in the measurement of NH4OH and the potential fixed electrochemical NH3 sensor with the neutral solution, and the anodic Ir catalyst possesses a good selectivity. Therefore, Ir may have practical application in the potential fixed electrochemical NH3 sensor with the neutral solution. 相似文献

A simple, high‐throughput and highly sensitive liquid chromatography coupled with tandem mass spectrometry (LC–MS/MS) method has been developed for the simultaneous estimation of rosuvastatin and free ezetimibe. Liquid–liquid extraction was carried out using methyl‐tert butyl ether after prior acidification from 300 μL human plasma. The recovery for both the analytes and their deuterated internal standards (ISs) ranged from 95.7 to 99.8%. Rosuvastatin and ezetimibe were separated on Symmetry C18 column using acetonitrile and ammonium formate buffer, pH 3.5 (30:70, v/v) as the mobile phase. The analytes were well resolved with a resolution factor of 3.8. Detection and quantitation were performed under multiple reaction monitoring using ESI(+) for rosuvastatin (m/z 482.0 → 258.1) and ESI(−) for ezetimibe (m/z 407.9 → 271.1). A linear response function was established in the concentration ranges of 0.05–50.0 ng/mL and 0.01–10.0 ng/mL for rosuvastatin and ezetimibe, respectively, with correlation coefficient, r2 ≥ 0.9991. The IS‐normalized matrix factors for the analytes ranged from 0.963 to 1.023. The developed method was successfully used to compare the pharmacokinetics of a fixed‐dose combination tablet of rosuvastatin‐ezetimibe and co‐administered rosuvastatin and ezetimibe as separate tablets to 24 healthy subjects. The reliability of the assay was also assessed by reanalysis of 115 subject samples. 相似文献

本文通过固结磨料球与KDP晶体对磨的单因素试验探究固结磨料球中反应物种类、磨粒浓度、反应物浓度、基体硬度对摩擦系数、磨痕截面积和磨痕处粗糙度的影响,试验结果表明:KHCO3固结磨料球对磨后磨痕对称性好,磨痕处的粗糙度值低;磨痕截面积随磨粒和反应物浓度的增加而增大,随基体硬度的增大而降低;磨痕处粗糙度随磨粒和反应物浓度的增加先降低后上升,随基体硬度的增大先上升后降低;摩擦系数受磨粒和反应物浓度影响不明显,随基体硬度的增大而降低。选择KHCO3作为反应物,Ⅰ基体,磨粒浓度为基体质量的100%,反应物浓度为15%制备固结磨料球与KDP晶体对磨后的磨痕轮廓对称度好且磨痕处粗糙度值低,以该组分制备固结磨料垫干式抛光KDP晶体,可实现晶体表面粗糙度Sa值为18.50 nm,材料去除率为130 nm/min的高效精密加工。 相似文献

制备了用于固定床反应器的负载型TS-1分子筛催化剂,并以苯酚羟基化为探针反应,研究了载体、钛含量和预处理剂等对负载型TS-1分子筛催化性能的影响,考察了固定床反应器中苯酚羟基化反应的工艺条件.结果表明,酸性较强的载体会加剧H2O2的分解,导致苯酚的转化率和H2O2的有效利用率下降;在所研究的ZSM-5,TiO2,Al2O3,ZrO2,SiO2和硅藻土载体中,硅藻土具有最好的催化性能;催化剂的活性与骨架钛含量有关;用HNO3,KAc,NaAc,NH4Ac和NH4Cl处理过的催化剂,活性和选择性得到提高,而用NH3·H2O,Na2CO3和Na3PO4等碱性物质处理后,催化剂的活性则下降;溶剂、反应温度、原料空速和苯酚/H2O2摩尔比对苯酚羟基化反应有很大影响.以丙酮为溶剂,在苯酚/H2O2摩尔比为3,反应温度为84℃,WHSV为8.46h-1的反应条件下,苯酚转化率、H2O2转化率、苯二酚选择性以及H2O2的有效利用率可分别达到27.7%,94.8%,97.7%和75.1%. 相似文献

采用改装的霍普金森压杆装置结合数值模拟对伪弹性TiNi合金固支梁的结构动态响应特性进行了研究。结果表明,在子弹冲击下,撞击点和固定端附近首先发生相变,并随着载荷增加,进一步产生相变铰,梁演变为二杆铰接机构。由于轴力作用,此处相变铰为拉伸侧的单边铰。与传统塑性铰不同,卸载后相变铰完全消失,梁回复原状没有残余变形。此外,对固支边界条件的实现及其对实验结果的影响进行了专门研究。 相似文献
