β-Glucosidase was covalently immobilized alone and coimmobilized with cellulase using a hydrophilic polyurethane foam (Hypol®FHP 2002). Immobilization improved the functional properties of the enzymes. When immobilized alone, the Km for cellobiose of β-glucosidase was decreased by 33% and the pH optimum shifted to a slightly more basic value, compared to the free enzyme. Immobilized β-glucosidase was extremely stable (95% of activity remained after 1000 h of continuous use). Coimmobilization of cellulase and β-glucosidase produced a cellulose-hydrolyzing complex with a 2.5-fold greater rate of glucose production for soluble cellulose and a four-fold greater increase for insoluble cellulose, compared to immobilized cellulase alone. The immobilized enzymes showed a broader acceptance of various types of insoluble cellulose substrates than did the free enzymes and showed a long-term (at least 24 h) linear rate of glucose production from microcrystalline cellulose. The pH optimum for the coimmobilized enzymes was 6.0. This method for enzyme immobilization is fast, irreversible, and does not require harsh conditions. The herbaceous crops that may provide fermentable carbohydrates for production of fuels and chemicals also contain 10–20%
protein. Protein coproduction with biomass-derived fuels and chemicals has important advantages: (1) food and fuel production
can be integrated, and (2) protein is a high-value product that may significantly improve overall process economics. We report
the results of an integrated approach to producing protein and fermentable sugars from one herbaceous species, Coastal Bermudagrass
(CBG). The ammonia fiber explosion (AFEX) process makes possible over 90% conversion of cellulose and hemicellulose to simple
sugars (about 650 mg reducing sugars/g dry CBG) at 5 IU cellulase/g vs < 20% conversion for untreated CBG. The AFEX treatment
also improves protein extraction from CBG; over 80% protein recovery is possible from AFEX-treated CBG vs about 30% recovery

Yellow poplar wood sawdust consists of 41% cellulose and 19% hemicellulose. The goal of pressure cooking this material in
water is to hydrate the more chemically resistive regions of cellulose in order to enhance enzymatic conversion to glucose.
Pretreatment can generate organic acids through acid-catalyzed degradation of monosaccharides formed because of acids released
from the biomass material or the inherent acidity of the water at temperatures above 160°C. The resulting acids will further
promote the acid-catalyzed degradation of monomers that cause both a reduction in the yield and the formation of fermentation
inhibitors such as hydroxymethyl furfural and furfural. A continuous pH-monitoring system was developed to help characterize
the trends in pH during pretreatment and to assist in the development of a base (2.0 M KOH) addition profile to help keep
the pH within a specified range in order to reduce any catalytic degradation and the formation of any monosac-charide degradation

Photosynthetic suspension cultures of higher plants offer an alternative approach to biomass production, potentially yielding
cellulosic material and protein on a continuous year-round basis. A bench-top hybrid photobioreactor was developed to study
photomixotrophic and photoautotrophic growth of Glycine max as a model system. Maximum biomass doubling times for photomixotrophic
and photoautotrophic growth were 1.87 and 3.92 d, respectively. The presence of exogenous sugars resulted in photomixotrophic
growth, reduced chlorophyll levels, and a reduction in photosynthetically-evolved oxygen. Depletion of carbohydrates from
the medium coincided with the beginning of stationary phase and an increase in oxygen evolution by the cells. A second growth
phase, prolonging cell viability, could be initiated by increasing the carbon dioxide from 2 to 5%, just before the onset
of stationary phase. Biomass from bioreactor cultured cells proved resistant to enzymatic attack without pretreatment. Composition
of the biomass was 7.8% lignin, 20.7% α-cellulose, 23% hemicellulose, 5.5% starch, 14.5% protein and 6.5% nucleic acids. 相似文献

Xylan backbones in native plant cell walls are extensively acety-lated. Previously, no direct investigations as to their role
in cellulolytic enzyme resistance have been done, though indirect results point to their importance. An in vitro deesterification
of aspen wood and wheat straw has been completed using hydroxylamine solutions. Yields of 90% acetyl ester removal for both
materials have been accomplished, with little disruption of other fractions (i.e., lignin). Apparently, as the xylan becomes
increasingly deacetylated, it becomes 5–7 times more digestible. This renders the cellulose fraction more accessible, and
2–3 times more digestible. This effect levels off near an acetyl removal of 75%, where other resistances become limiting. 相似文献

Aspen wood substrates with varying degrees of deacetylation, xylan, and lignin removal have been prepared and submitted to
enzymatic hydrolysis with a cellulase/hemicellulase preparation for an extended constant period of hydrolysis. Controlled
deacetylation has been achieved by treating wood with various alkali metal hydroxide solutions, at various alkali/wood ratios.
It has been found that samples with the same extent of deacetylation produce the same sugar yields upon enzymatic hydrolysis.
Increased degree of deacetylation increases the yield of sugars obtained from enzymatic hydrolysis, all other compositional
parameters held constant. The acetyl group removal is proportional to the stoichiometric relation between added base and wood
acetyl content, i.e., the same number of milliequivalents of base/weight of wood remove the same extent of acetyl groups,
regardless of the concentration of the base solution. No cation effects are found among Li, Na, and K alkali hydroxide solutions,
suggesting that swelling is not as important a parameter as is the removal of the acetyl groups from the xylan backbone in
determining the extent of hydrolyzability of the resulting sample. 相似文献

We have computed a state-of-the-art benchmark potential energy surface (PES) for the archetypal oxidative addition of the ethane C-C bond to the palladium atom and have used this to evaluate the performance of 24 popular density functionals, covering LDA, GGA, meta-GGA, and hybrid density functionals, for describing this reaction. The ab initio benchmark is obtained by exploring the PES using a hierarchical series of ab initio methods [HF, MP2, CCSD, CCSD(T)] in combination with a hierarchical series of five Gaussian-type basis sets, up to g polarization. Relativistic effects are taken into account either through a relativistic effective core potential for palladium or through a full four-component all-electron approach. Our best estimate of kinetic and thermodynamic parameters is -10.8 (-11.3) kcal/mol for the formation of the reactant complex, 19.4 (17.1) kcal/mol for the activation energy relative to the separate reactants, and -4.5 (-6.8) kcal/mol for the reaction energy (zero-point vibrational energy-corrected values in parentheses). Our work highlights the importance of sufficient higher angular momentum polarization functions for correctly describing metal-d-electron correlation. Best overall agreement with our ab initio benchmark is obtained by functionals from all three categories, GGA, meta-GGA, and hybrid DFT, with mean absolute errors of 1.5 to 2.5 kcal/mol and errors in activation energies ranging from -0.2 to -3.2 kcal/mol. Interestingly, the well-known BLYP functional compares very reasonably with a slight underestimation of the overall barrier by -0.9 kcal/mol. For comparison, with B3LYP we arrive at an overestimation of the overall barrier by 5.8 kcal/mol. On the other hand, B3LYP performs excellently for the central barrier (i.e., relative to the reactant complex) which it underestimates by only -0.1 kcal/mol. 相似文献

3-Acetamido-3,6-dideoxy-d-galactopyranose (d-Fucp3NAc) is an aminosugar almost exclusively found in phytopathogenic O-antigens. The glycosylation reaction involving d-Fucp3NAc donors was studied with several rhamnosyl acceptors, revealing that the best yields and highest α-stereoselectivity were obtainable by coupling a N-phenyl trifluoroacetimidate glycosyl donor in a ternary mixture (dioxane/DME/toluene 4:1:1) as solvent. For the first time a synthetic access to α-d-Fucp3NAc containing oligorhamnans, that are interesting molecules for studying the effects of O-antigen model oligosaccharides on the modulation of plant response to bacteria, was reported.
