为了提高理论力学课堂的趣味性,本文以树枝上小鸟作为刚体平衡的例子,应用力的平行四边形法则、力的可传性、三力平衡汇交定理等基本原理分析了树枝上小鸟的双腿受力,以及啄木鸟尾巴在保持身体平衡中的作用,并讨论了小鸟不同姿态对其受力的影响,期望达到提高学习兴趣、寓教于乐、改善教学效果的目的。     

Developing CRISPR/Cas9-Mediated Fluorescent Reporter Human Pluripotent Stem-Cell Lines for High-Content Screening

Application of the CRISPR/Cas9 system to knock in fluorescent proteins to endogenous genes of interest in human pluripotent stem cells (hPSCs) has the potential to facilitate hPSC-based disease modeling, drug screening, and optimization of transplantation therapy. To evaluate the capability of fluorescent reporter hPSC lines for high-content screening approaches, we targeted EGFP to the endogenous OCT4 locus. Resulting hPSC–OCT4–EGFP lines generated expressed EGFP coincident with pluripotency markers and could be adapted to multi-well formats for high-content screening (HCS) campaigns. However, after long-term culture, hPSCs transiently lost their EGFP expression. Alternatively, through EGFP knock-in to the AAVS1 locus, we established a stable and consistent EGFP-expressing hPSC–AAVS1–EGFP line that maintained EGFP expression during in vitro hematopoietic and neural differentiation. Thus, hPSC–AAVS1–EGFP-derived sensory neurons could be adapted to a high-content screening platform that can be applied to high-throughput small-molecule screening and drug discovery campaigns. Our observations are consistent with recent findings indicating that high-frequency on-target complexities appear following CRISPR/Cas9 genome editing at the OCT4 locus. In contrast, we demonstrate that the AAVS1 locus is a safe genomic location in hPSCs with high gene expression that does not impact hPSC quality and differentiation. Our findings suggest that the CRISPR/Cas9-integrated AAVS1 system should be applied for generating stable reporter hPSC lines for long-term HCS approaches, and they underscore the importance of careful evaluation and selection of the applied reporter cell lines for HCS purposes.     

Studying the Suitability of Nineteen Lignins as Partial Polyol Replacement in Rigid Polyurethane/Polyisocyanurate Foam

In this study, nineteen unmodified lignins from various sources (hardwood, softwood, wheat straw, and corn stover) and isolation processes (kraft, soda, organosolv, sulfite, and enzymatic hydrolysis) were used to replace 30 wt.% of petroleum-based polyol in rigid polyurethane/polyisocyanurate (PUR/PIR) foam formulations. Lignin samples were characterized by measuring their ash content, hydroxyl content (Phosphorus Nuclear Magnetic Resonance Spectroscopy), impurities (Inductively Coupled Plasma), and pH. After foam formulation, properties of lignin-based foams were evaluated and compared with a control foam (with no lignin) via cell morphology, closed-cell content, compression strength, apparent density, thermal conductivity, and color analysis. Lignin-based foams passed all measured standard specifications required by ASTM International C1029-15 for type 1 rigid insulation foams, except for three foams. These three foams had poor compressive strengths, significantly larger cell sizes, darker color, lower closed-cell contents, and slower foaming times. The foam made with corn stover enzymatic hydrolysis lignin showed no significant difference from the control foam in terms of compressive strength and outperformed all other lignin-based foams due to its higher aliphatic and p-hydroxyphenyl hydroxyl contents. Lignin-based foams that passed all required performance testing were made with lignins having higher pH, potassium, sodium, calcium, magnesium, and aliphatic/p-hydroxyphenyl hydroxyl group contents than those that failed.     

Dislocation Analysis for Large-sized Sapphire Single Crystal Grown by SAPMAC Method

In this paper,large-sized sapphire (230×210 mm,27.5 kg) was grown by SAPMAC method (sapphire growth technique with micro-pulling and shoulder-expanding at the cooled center). Dislocation peculiarity in large sapphire boule (0001) basal plane was investigated by chemical etching,scanning electron microscopy and X-ray topography method. The triangular dislocation etch pit measured is 7.6×101～8.0×102 cm-2,in which relative high-density dislocations were generated at both initial and final stages of crystal growth. The analysis of single-crystal X-ray topography shows that there are no apparent sub-grain boundaries; the dislocation lines are isolated and straight. Finally,the origins of low-density dislocation in sapphire crystal are discussed by numerical analysis method.     

家禽肌肉组织中硝基咪唑类药物残留的液相色谱串联质谱测定

建立了同时检测家禽肌肉组织中3种硝基咪唑类药物残留的液相色谱-串联质谱法。样品用乙酸乙酯提取,经浓缩、过滤后直接进入液相色谱-质谱分析。采用监测母离子及其相应子离子的多反应监测技术,根据离子流峰面积进行定量。3种药物的定量检出限分别为甲硝唑0．8μg／kg、洛硝哒唑0．1μg／kg、二甲硝唑0．1μg／kg。1次分析的运行时间为5min。是一种快速、灵敏、可靠的分析方法。     

液相色谱-串联质谱联用法测定蜂蜜及水产品中硝基呋喃类抗生素代谢物残留量

采用微量化样品前处理技术,以邻硝基苯甲醛为衍生剂,电喷雾正离子多反应监测方式建立了蜂蜜及水产品中四种硝基呋喃类抗生素代谢物残留量同时测定的液相色谱-串联质谱联用法。方法的检出限为0.02～0.3 ng/g,线性范围均>10~2,线性方程的相关系数>0.997,回收率为79.0％～95.6％。     

尿样及动物组织中甲氧苄氨嘧啶残留量的高效液相色谱-串联质谱测定法

采用微量化样品前处理技术,以固相萃取为净化方法,电喷雾正离子多反应监测方式建立了尿样及动物组织中甲氧苄氨嘧啶残留量的液相色谱-串联质谱联用测定法.检出限为0.1 ng/g,线性范围均大于3个数量级,线性方程的相关系数大于0.999,组织样品和尿样的回收率分别为89%和93%.     

苯二甲酸衍生物及含氮配体构筑的两个镍配位聚合物的合成、晶体结构与热稳定性研究

使用硝酸镍分别与2个含取代基的苯基二羧酸3-硝基邻苯二甲酸（3-NPAH₂）和5-溴间苯二甲酸（H₂BIPA）以及2个刚性含氮配体4,4’-联吡啶（4,4’-bipy）、1,4’-二咪唑基二苯（L=1,4’-bis（imidazol-yl）benzene）在水热条件下合成了2个镍配位聚合物,即,[Ni（3-NPA）（4,4’-bipy）（H₂O）]_n（1）和[Ni（BIPA）（L）]_n（2）。表征了2个化合物的红外光谱、元素分析、热重、X-射线单晶衍射以及X-射线粉末衍射。化合物1是三斜晶系,P1空间群,含有[Ni（OCO）₄Ni]双核单元,该双核单元再通过4,4’-联吡啶连接而成一个二维单层结构。层与层之间通过羧酸氧原子与配位水分子之间产生氢键作用。化合物2是单斜晶系,P2₁/c空间群,含有[Ni₂（COO）₂]双核单元,该双核单元进一步通过BIPA^2-构成了一个双链结构,该双链再通过1,4’-二咪唑基二苯桥联配体连接形成了一个三维框架结构。此外,热重分析结果显示化合物2具有较高的热稳定性。     

超高效液相色谱-串联质谱法鉴定猪肉的特异性肽生物标志物

以亲缘关系较近的猪、牛和羊3个物种的肌肉组织为研究对象,采用超高效液相色谱-串联质谱(UPLC-MS),筛选并确认了猪物种肉特异性肽生物标志物.3种纯肉样品经蛋白质提取、胰蛋白酶消化和UPLC-TripleTOF-MS分离鉴定,得到的总离子流图谱(TIC)与Uniprot蛋白质数据库对比分析,筛选出3个物种肉的3种高丰度同源蛋白和8种潜在的肽生物标志物;潜在的肽生物标志物经QTRAP-MS质谱的多反应模式(MRM)分析,最终确认了猪物种肉的5种肽生物标志物,其中3种肽生物标志物未见文献报道.
