The root explants of the germinated seedlings of Podophyllum hexandrum were grown in MS medium supplemented with indole acetic acid (IAA) (2 mg/L) and activated charcoal (0.5%), and healthy callus
culture was obtained after incubation for 3 wk at 20°C. The cultivation of plant cells in shake flask was associated with
problems such as clumping of cells and browning of media, which were solved by the addition of pectinase and polyvinylpyrrolidone.
The effect of major media components and carbon source was studied on the growth and podophyllotoxin production in suspension
culture. It was found that glucose was a better carbon source than sucrose and that NH4
+:NO3
− ratio (total nitrogen concentration of 60 mM) and PO4
3− did not have much effect on the growth and product formation. The relative effect of culture parameters (inoculum level,
pH, IAA, glucose, NH4
+:NO3
− ratio, and PO4
3−) on the overall growth and product response of the plant cell suspension culture was further investigated by Plackett-Burman
design. This indicated that inoculum level, glucose, IAA, and pH had significant effects on growth and production of podophyllotoxin.
To identify the exact optimum concentrations of these parameters on culture growth and podophyllotoxin production, central
composite design experiments were formulated. The overall response equations with respect to growth and podophyllotoxin production
as a function of these culture parameters were developed and used to determine the optimum concentrations of these parameters,
which were pH 6.0, 1.25 mg/L of IAA, 72 g/L of glucose, and inoculum level of 8 g/L. 相似文献

A molecularly imprinted polymer was prepared using a water-soluble crosslinking agent. An ionic complex was utilized as the assembly for the template molecule and the functional monomer, and water as porogenic solvent during preparation of the imprinted polymer. The results of chromatographic evaluations for the prepared polymer suggested that the polymer had much lower hydrophobicity compared with usual octadecyl group bonded silica or the usual molecular imprinted polymer prepared from ethyleneglycol dimethacrylate, and the selective recognition ability for template molecule in the completely aqueous condition. 相似文献

AbstractHerein the bioaccessibility of Mn, Cu, Zn, Cd, and Pb, selected from essential and toxic elements, was determined in commercially sold vegetable purees intended for infant and toddler consumption. Chemical fractionation studies using water, acetone, diethyl ether, chloroform:methanol, and n-hexane were employed to predict the importance of the protein and lipid parts of a matrix to assess the bioaccessibility data. In addition, in-vitro gastrointestinal digestion was performed to determine the bioaccessibility of the elements using a five level, three factor central composite design (CCD) to maximize the elemental solubility. The total elemental concentrations in all of the fractions were determined by inductively coupled plasma – mass spectrometry (ICP-MS). Based on the consumption of one jar of vegetable/rice-based baby food, Zn was 1.3% of the recommended dietary allowance and Mn was 4.2% of the adequate intake level, while Cu was almost 100% of the adequate intake level. Additionally, Pb was always below the detection limit and Cd was sometimes under the detection limit for the percent bioaccessibility. Cu2MnGeS4 crystallizes orthorhombic in a wurtzite superstructure type while Cu2MnSnS4 crystallizes in a tetragonal sphalerite superstructure type. Lattice constants and thermal analyses of the solid solution series Cu2MnGexSn1−xS4 are presented. A two-phase region is found from Cu2MnGe0.3Sn0.7S4 to Cu2MnGe0.5Sn0.5S4. The cell volume of the mixed crystals increases with increasing Sn content. The melting points increase smoothly with increasing Ge content to x=0.5 and then steeply for higher Ge contents. The single crystal X-ray structure analysis of Cu2MnGe0.55Sn0.45S4 is presented. The refinement converges to R=0.0270 and wR2=0.0586, Z is 2. The volumes of the tetrahedra [MS4] (M=Cu, Mn, Ge, Sn) are calculated. From these volumes the differences in size of the tetrahedra are derived and compared with the corresponding differences in the end members of the solid solution series. It turns out that the resulting structure type in these materials depends on the volume differences of the constituting tetrahedra [MS4]. 相似文献

The Green's function is constructed, its trace and the trace of its first iteration are calculated, and the ability of the traces to provide upper and lower bounds for the ground eigenvalue is examined. A delayed coincidence method, time-interval analysis (TIA), has been applied to successive – decay events on the millisecond time-scale. Such decay events are part of the 220Rn216Po (T1/2 145 ms) (Th-series) and 219Rn215Po (T1/2 1.78 ms) (Ac-series). By using TIA in addition to measurement of 226Ra (U-series) from -spectrometry by liquid scintillation counting (LSC), two natural decay series could be identified and separated. The TIA detection efficiency was improved by using the pulse-shape discrimination technique (PSD) to reject -pulses, by solvent extraction of Ra combined with simple chemical separation, and by purging the scintillation solution with dry N2 gas. The U- and Th-series together with the Ac-series were determined, respectively, from alpha spectra and TIA carried out immediately after Ra-extraction. Using the 221Fr217At (T1/2 32.3 ms) decay process as a tracer, overall yields were estimated from application of TIA to the 225Ra (Np-decay series) at the time of maximum growth. A series of Ti/Mg supported catalysts are prepared by using ball-milled mixtures of MgCl2-ethanol adducts and NaCl as supports, and 1-hexene polymerizations catalyzed by the novel catalysts are studied. It is found that the molecular weight distribution of poly(1-hexene) becomes apparently narrower when catalysts with doped supports are used, indicating that changing the structure of the support is an effective way to regulate the active center distribution of heterogeneous Ziegler-Natta catalyst. 相似文献
