Immunoglobulins in bovine colostrum were separated and fractionated from other proteins using the method and instrumentation developed in our laboratory. The proposed separation was based on bidirectional isotachophoresis/moving boundary electrophoresis with electrofocusing of the analytes in a pH gradient from 3.9 to 10.1. The preparative instrumentation included the trapezoidal non-woven fabric that served as separation space with divergent continuous flow. The defatted and casein precipitate-free colostrum supernatant was loaded directly into the instrument without any additional colostrum pre-preparation. Immunoglobulin G was fractionated from other immune proteins such as bovine serum albumin, β-lactoglobulin, and α-lactalbumin, and was continuously collected in separated fractions over 3 h. The fractions were further processed, and isolated immunoglobulin G in the liquid fractions was confirmed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and by re-focusing in gel isoelectric focusing. Separated immunoglobulin G was detected in seven fractions by sodium dodecyl sulfate-polyacrylamide gel electrophoresis with a gradually decreased concentration in the fractions. Re-focusing of the proteins in the fractions by gel isoelectric focusing revealed multiple separated zones of immunoglobulin G with the isoelectric point values covering the range from 5.4 to 7.2. Each fraction contained distinct zones with gradually increased isoelectric point values and decreased concentrations from fraction to fraction.  相似文献

为了得到壁面温度在不同来流速度、不同湍流强度条件下对边界层转捩与减阻的影响规律,本文采用Transitionk-kl-ω模型对低来流速度下无压力梯度的光滑平板进行了数值模拟。结果表明,随着来流速度的升高,壁温升高所起到的减阻效果更好,即高来流速度对壁面温度更为敏感。当来流处于中高湍流强度下时,壁温升高能起到推迟转捩的作用,且随着湍流强度的升高,转捩推迟的效果越好,但减阻效果正好相反;当来流处于低湍流强度下时,壁温升高会使得转捩提前发生。壁温升高抑制了边界层内流体的脉动程度,使得层流的稳态不易被破坏,流动更加稳定;同时,壁温升高使得边界层内流体的速度梯度减小,从而降低了壁面摩擦系数,故壁温升高能起到推迟边界层转捩与减阻的作用。  相似文献

In this work it is experimentally investigated a size effect in temperature coefficient of resistance (TCR) of Ni films with Cu and SiO2 thin overlayer. The parameters of electrical transfer (the mean‐free path of electron, the reflectivity coefficient of the external surfaces, the reflection and transmission coefficients at the grain boundary) were calculations. Decreasing of the value of the reflectivity coefficient is due to the change of the surface microrelief. This work introduces a mathematical model for laser cutting taking account of spatially distributed laser radiation. The model involves two coupled nonlinear partial differential equations describing the interacting dynamical behaviors of the free boundaries of the melt during the process. The model will be investigated by linear stability analysis to study the occurence of ripple formations at the cutting surface. We define a measurement for the roughness of the cutting surface and introduce an optimal control problem for minimizing the roughness with respect to the laser beam intensity along the free melt surface. Necessary optimality conditions will be deduced. Finally, a numerical solution will be presented and discussed by means of the necessary conditions. physical considerations.  相似文献

A direct numerical simulation (DNS) dataset of a turbulent boundary layer (TBL) with a step change from a smooth to a rough surface is analyzed to examine the characteristics of a spatially developing flow. The roughness elements are periodically arranged two-dimensional (2-D) spanwise rods, with the first rod placed 80θin downstream from the inlet, where θin denotes the inlet momentum thickness. Based on an accurate estimation of relevant parameters, clear evidence for mean flow universality is provided when scaled properly, even for the present roughness configuration, which is believed to have one of the strongest impacts on the flow. Compared to previous studies, it is shown that overshooting behavior is present in the first- and second-order statistics and is locally created either within the cavity or at the leading edge of the roughness depending on the type of statistics and the wall-normal measurement location. Inspection of spatial two-point correlations of the streamwise velocity fluctuations shows a continuous increase of spanwise length scales of structures over the rough wall after the step change at a greater growth rate than that over smooth wall TBL flow. This is expected because spanwise energy spectrum shows presence of much energetic wider structures over the rough wall. Full images of the DNS data are presented to describe not only predominance of hairpin vortices but also a possible spanwise scale growth mechanism via merging over the rough wall.  相似文献

In this paper we consider an inverse scattering problem from an open arc with impedance boundary conditions on both sides of the crack. Our aim is to recover both the impedance function and the unknown crack simultaneously from the far-field pattern with only one incident wave. Making the most out of the direct problem, a straightforward method of iterative nature is developed for the inverse problem. The ill-posedness of this problem is considered by incorporating the Tikhonov regularization. Numerical examples are provided at the end of the paper to show the feasibility of our method.  相似文献

A static, purely flexural mechanical analysis is presented for a Kirchhoff solid circular plate, deflected by a transverse central force, and bilaterally supported along a single periphery arc, the remaining part of the boundary being free. The contact reaction is assumed to be formed by a distributed reaction force accompanied by a distributed moment with radial axis. This plate problem is formulated in terms of an integral equation of the Prandtl type, supplemented by a vertical and a rotational equilibrium condition. It is shown that the Prandtl equation coupled to the equilibrium condition possesses a unique solution in a proper scale of weighted Sobolev‐type spaces, with certain smoothness properties, and that no solution exists if the distributed moment is absent. Finally, the behaviour of the solution at the endpoints of the support is clarified.  相似文献

A boundary layer analysis is used to investigate the effect of lateral mass flux on mixed convection heat and mass transfer over inclined permeable surfaces in porous media. The conservation equations that govern the problem are reduced to a system of non-linear ordinary differential equations and then the resulting equations is solved by numerical method. The numerical results for heat and mass transfer in terms of Nusselt and Sherwood number are presented in x-y plots for the buoyancy ratio (N) and Lewis number (Le) with mass flux pammeter (Fw). The obtained results are validated against previously published results with on special case of the problem.  相似文献

Quasi‐bubble finite element approximations to the shallow water equations are investigated focusing on implementations of the surface elevation boundary condition. We first demonstrate by numerical results that the conventional implementation of the boundary condition degrades the accuracy of the velocity solution. It is also shown that the degraded velocity leads to a critical instability if the advection term is present in the momentum equation. Then we propose an alternative implementation for the boundary condition. We refer to this alternative implementation as a discontinuous boundary (DB) implementation because it introduces at each boundary node two independent mass–flux values that result in a discontinuity at the boundary. Numerical results show that the proposed DB implementation is consistent, stabilizes the quasi‐bubble scheme, and leads to second‐order accuracy at the surface elevation specified boundary.
