An amperometric method for the determination of the neurotoxic amino acid β‐N‐oxalyl‐L ‐α,β‐diaminopropionic acid (β‐ODAP) using a screen printed carbon electrode (SPCE) is reported. The electrode material was bulk‐modified with manganese dioxide and used as a detector in flow injection analysis (FIA). The enzyme glutamate oxidase (GlOx) was immobilized in a Nafion‐film on the electrode surface. The performance of the biosensor was optimized using glutamate as an analyte. Optimum parameters were found as: operational potential 440 mV (vs. Ag/AgCl), flow rate 0.2 mL min?1, and carrier composition 0.1 mol L?1 phosphate buffer (pH 7.75). The same conditions were used for the determination of β‐ODAP. The signal was linear within the concentration range 53–855 μmol L?1 glutamate and 195–1950 μmol L?1 β‐ODAP. Detection limits (as 3σ value) for both analytes were 9.12 and 111.0 μmol L?1, respectively, with corresponding relative standard deviations of 3.3 and 4.5%. The biosensor retained more than 73% of its activity after 40 days of on‐line use.  相似文献

A potentiometric sensor constructed from a mixture of 25% (m/m) spinel‐type manganese oxide (lambda‐MnO2), 50% (m/m) graphite powder and 25% (m/m) mineral oil is used for the determination of lithium ions in a flow injection analysis system. Experimental parameters, such as pH of the carrier solution, flow rate, injection sample volume, and selectivity for Li+ against other alkali and alkaline‐earth ions and the response time of this sensor were investigated. The sensor response to lithium ions was linear in the concentration range 8.6×10?5–1.0×10?2 mol L?1 with a slope 78.9±0.3 mV dec?1 over a wide pH range 7–10 (Tris buffer), without interference of other alkali and alkaline‐earth metals. For a flow rate of 5.0 mL min?1 and a injection sample volume of 408.6 μL, the relative standard deviation for repeated injections of a 5.0×10?4 mol L?1 lithium ions was 0.3%.  相似文献

An amperometric detector for hydrophobic ions based on a plasticized poly(vinyl) chloride (PVC) membrane incorporated in a flow‐injection system was developed. A four‐electrode potentiostat with ohmic drop compensation was used, while a flow‐through cell incorporated the four electrodes and the membrane, which contained tetrabutylammonium tetraphenylborate. When the influence of the applied potential and of the flow‐injection variables on the determination of tetrabutylammonium was studied, a linear relationship was observed between current peak height and ion concentration over a range of 5×10?6–6×10?5 M tetrabutylammonium. Good repeatability and between‐day reproducibility and high sample frequency were obtained. The effect of other ions was studied. Two different amperometric methods, indirect and direct, were also developed for the determination of dodecylsulfate in the concentration range 3×10?5–9×10?4 M.  相似文献

The preparation and the electrochemical study of Disperse Blue 1‐chemically modified electrodes (DB1‐CME), as well as their efficiency for the electrocatalytic oxidation of NADH is described. The proposed mediator was immobilized by physical adsorption onto graphite electrodes. The electrochemical behavior of DB1‐CME was studied with cyclic voltammetry. The electrochemical redox reaction of DB1 was found to be reversible, revealing two well‐shaped pair of peaks with formal potentials 152 and ?42 mV, respectively, (vs. Ag/AgCl/3M KCl) at pH 6.5. The current Ip has a linear relationship with the scan rate up to 800 mV s?1, which is indicative for a fast electron transfer kinetics. The dissociation constants of the immobilized DB1 redox couple were calculated pK1=4 and pK2=5. The electrochemical rate constants of the immobilized DB1 were calculated k1°=18 s?1 and k2°=23 s?1 (Γ=2.36 nmol cm?2). The modified electrodes were mounted in a flow injection manifold, poised at +150 mV (vs. Ag/AgCl/3M KCl) and a catalytic current due to the oxidation of NADH was measured. The reproducibility was 1.4% RSD (n=11 for 30 μM NADH) The behavior of the sensor towards different reducing compounds was investigated. The sensor exhibited good operational and storage stability.  相似文献

Fluorenone (C13H8O) was inserted into the channels of zeolite L by using gas-phase adsorption. The size, structure, and stability of fluorenone are well suited for studying host-guest interactions. The Fourier transform IR, Raman, luminescence, and excitation spectra, in addition to thermal analysis data, of fluorenone in solution and fluorenone/zeolite L are reported. Normal coordinate analysis of fluorenone was performed, based on which IR and Raman bands were assigned, and an experimental force field was determined. The vibrational spectra can be used for nondestructive quantitative analysis by comparing a characteristic dye band with a zeolite band that has been chosen as the internal standard. Molecular orbital calculations were performed to gain a better understanding of the electronic structure of the system and to support the interpretation of the electronic absorption and luminescence spectra. Fluorenone shows unusual luminescence behavior in that it emits from two states. The relative intensity of these two bands depends strongly on the environment and changes unexpectedly in response to temperature. In fluorenone/zeolite L, the intensity of the 300 nm band (lifetime 9 micros) increases with decreasing temperature, while the opposite is true for the 400 nm band (lifetime 115 micros). A model of the host-guest interaction is derived from the experimental results and calculations: the dye molecule sits close to the channel walls with the carbonyl group pointing to an Al3+ site of the zeolite framework. A secondary interaction was observed between the fluorenone's aromatic ring and the zeolite's charge-compensating cations.  相似文献

A fast multi‐residue screening method for determining pesticides in tea is described. Pesticides are extracted from tea with acetone and methylene chloride, then enriched and cleaned up with solid phase extraction (SPE) prior to gas chromatographic determination. The fast screening is achieved by a gas chromatograph system equipped with dual‐column, dual‐tower auto‐sampler and both electron capture detector (ECD) and flame photometric detector (FPD). Optimal conditions are investigated for the prospective pesticides including column selection, detection mode, the retention behaviors, quantitative calibration, as well as the recoveries and repeatability of pesticides from tea samples. Under the optimal conditions, with the FPD‐P detector accompanied CP‐SIL 13CB column, 48 pesticides can be separated well and detected within 38 min; and with a DB‐5 column, 35 ECD‐detectable pesticides can be separated and detected within 46 min. The recoveries of 84 pesticides in tea samples are 65–120% with 0.34–16% RSD for spiking 0.02–3.0 mg/kg standard species. Because of the thermal instability of most pesticides, direct cold extraction of pesticides from a tea sample is recommended. The proposed method provided a very fast and efficient procedure to screen 84 pesticides from a complicated tea sample matrix.  相似文献

For rough heterogeneous samples, the contrast observed in XPS images may result from both changes in elemental or chemical composition and sample topography. Background image acquisition and subtraction are frequently utilized to minimize topographical effects so that images represent concentration variations in the sample. This procedure may significantly increase the data acquisition time. Multivariate statistical methods can assist in resolving topographical and chemical information from multispectral XPS images. Principal component analysis (PCA) is one method for identification of the highest correlation/variation between the images. Topography, which is common to all of the images, will be resolved in the first most significant component. The score of this component contains spatial information about the topography of the surface, whereas the loading is a quantitative representation of the topography contribution to each elemental/chemical image. The simple‐to‐use self‐modelling mixture analysis (Simplisma) method is a pure variable method that searches for the source of most differences in the data and therefore has the potential to distinguish between chemical and topographical phases in images. The mathematical background correction scheme is developed and validated by comparing results to the experimental background correction for samples with differing degrees of topography. An error analysis for numerically evaluating random uncertainties in x‐ray photoelectron spectroscopy has been implemented in version 2003 of the spectra treatment and analysis software UNIFIT in order to improve the understanding of the statistical basis and the reliability of the model parameters for photoelectron spectra. The theoretical basis as well as two approaches to obtain error limits of the fit parameters have been considered. Several test spectra have been analysed and discussed. A representative example has been chosen to demonstrate the relevance of the error estimation for practical surface analysis. A novel hybrid optimization method called quantum stochastic tunneling has been recently introduced. Here, we report its implementation within a new docking program called EasyDock and a validation with the CCDC/Astex data set of ligand-protein complexes using the PLP score to represent the ligand-protein potential energy surface and ScreenScore to score the ligand-protein binding energies. When taking the top energy-ranked ligand binding mode pose, we were able to predict the correct crystallographic ligand binding mode in up to 75% of the cases. By using this novel optimization method run times for typical docking simulations are significantly shortened.  相似文献
