利用高效液相色谱建立了地黄的色谱指纹图谱。采用反相C1 8柱 (5 μm ,1 5 0mm× 4.6mm) ,流动相甲醇∶水 (V V =5∶95 ) ,检测波长为 2 80nm ,流速 1 .0mL min进行实验。根据相对保留值和相对面积值对色谱指纹图谱进行分析对比研究 ,建立了控制地黄药材质量的新方法。该法为中药样品的鉴定提供了较全面的信息 ,并为HPLC在复杂组分的样品分析领域开拓了新的应用途径。  相似文献

 Four inter-comparison exercises on organic elemental analysis were carried out between 1997 and 2001 by the Department of Analytical Chemistry of the University of Barcelona, together with the Microanalysis Service and the Institute of the Marine Sciences, which both belong to the CSIC in Barcelona, and the University of A Coru?a. More than sixty laboratories participated in these exercises. Here we describe the design and characteristics of the trials, the samples and the homogeneity tests applied. We report the results obtained for the analysis of carbon, hydrogen, nitrogen, sulphur and oxygen, their statistical analysis, and the most relevant aspects of the technical discussion meetings. The storage stability of the occupationally frequently occurring compounds, methylethylketone, methylisobutylketone, benzene, toluene, tetrachloroethylene, n-butylacetate, -pinene, β-pinene, limonene and n-decane, has been investigated on the adsorbents Tenax TA, Chromosorb 106 and Carbotrap using thermally desorbable tube type samplers, commonly utilized in ambient and workroom atmospheric measurements. Fifty and 500 ng of each compound were loaded on the various adsorbents tubes, stored at both ambient (20 °C) and refrigerated (4 °C) temperatures and analysed by means of thermal gas chromatography with mass spectrometric detection on days 0, 7, 14 and 28 after exposure. A 90% storage recovery was chosen as acceptance criteria for storage stability, and statistical testing by Student's t-test, analysis of variance and Bonferroni post hoc tests were employed to investigate the effect of the categorical variables storage time, storage temperature and analyte loading on the different adsorbents. Chromosorb 106 showed the overall best behaviour with recoveries of 90% or better for all analytes during the 28-day test period. Tenax TA and Carbotrap yielded lower recoveries and were more influenced by variations in storage time, storage temperature and analyte loading. Refrigerated temperatures were best avoided for storage on Tenax TA, but may increase the recovery of some compounds on Carbotrap (e.g. n-butylacetate). The blank build-up on the adsorbents was also investigated, and Carbotrap and Tenax TA showed no signs of artefact development over time. Chromosorb 106, however, contained inherently more artefacts that build up over time, which in spite of the excellent storage capability, may limit its use in field studies where long storage times are normal.  相似文献

Knowledge of the equation of state of adsorbed or deposited layers of proteins at the air/water interface is of fundamental interest in the understanding of the surface activity of these molecules. Using scaling laws of current polymer theories, it has been shown that the equation of state of the interfacial layer in the semi-dilute regime should relate the surface pressure to the surface concentration through a power law. The exponent of this power law should reflect the quality of the solvent and the conformation of the adsorbed polypeptide chain. In the case of β-lactoglobulin layers, in the range of surface concentrations that should correspond to the semi-dilute regime, the relationship between surface pressure and surface concentration is expressed as a power law. The exponent of this power law is strongly influenced by the nature of the aqueous substrate and by the net charge of the protein molecule. The use of scaling laws gives a coherent view of the expansion of the polypeptide chain in the interfacial layer and of the relationship between surface concentration and surface pressure in the semi-dilute regime. This result favours a strong similarity between β-lactoglobulin and a polymer chain in the interfacial layer. It is concluded that current theories of polymer adsorption could be applied to interfacial protein layers.  相似文献

In this paper, superhigh reproductive rate strains of MEV with titre more than HA8192* or TCID50 log9.7 10 have been achieved both by cultivation in cell lines with different susceptibility to MEV and by isolating and identifying in field by the author. The systematic tests proved that S18 and L12 strains of MEV are the best strains for vaccine preparation. In this study, the best means for the tissue cultivation of MEV and the most advanced technological process for the production and detection of serum-free cell-cultured MEV fluids with super-high HA titre in batches in large quantities have been established for the first time. Optimum conditions for MEV inactivation were determined, and safe and effective inactivated vaccines with mineral oil or A1(OH)3 gel adjuvant were successfully prepared with serum-free cell-cultured MEV fluids. Both vaccines with different adjuvants can be manufactured in batches in large quantities and have been widely used all over China since 1986. The change laws of the imm  相似文献

本文对不同产地和市售的二十四种厚朴药材及饮片的萃取液进行了高效液相色谱、气相色谱及紫外光谱分析;对所得数据用主成分分析进行分类,概括出不同样品化学成分的异同,认为大叶木兰可做为厚朴的代用品,而在陕西安康购得的样品不应做厚朴使用,后者经生药学鉴定为厚朴伪品凹叶木兰。化学模式识别技术可为寻找药材的代用品、评价中药质量及鉴定伪劣药材提供依据。  相似文献

Traditional Chinese medicine (TCM) is the key to unlock treasures of Chinese civilization. TCM and its compound play a beneficial role in medical activities to cure diseases, especially in major public health events such as novel coronavirus epidemics across the globe. The chemical composition in Chinese medicine formula is complex and diverse, but their effective substances resemble “mystery boxes”. Revealing their active ingredients and their mechanisms of action has become focal point and difficulty of research for herbalists. Although the existing research methods are numerous and constantly updated iteratively, there is remain a lack of prospective reviews. Hence, this paper provides a comprehensive account of existing new approaches and technologies based on previous studies with an in vitro to in vivo perspective. In addition, the bottlenecks of studies on Chinese medicine formula effective substances are also revealed. Especially, we look ahead to new perspectives, technologies and applications for its future development. This work reviews based on new perspectives to open horizons for the future research. Consequently, herbal compounding pharmaceutical substances study should carry on the essence of TCM while pursuing innovations in the field.  相似文献

Differential scanning calorimetry (DSC) is the most widely used thermal analytical technique in food research and it has a great utility in quality assurance of food. Proteins are the most studied food components by thermal analysis including studies on conformation changes of food proteins as affected by various environmental factors, thermal denaturation of tissue proteins, food enzymes and enzyme preparations for the food industry, as well as effects of various additives on their thermal properties. Freezing-induced denaturation of food proteins and the effect of cryoprotectants are also monitored by DSC. Polymer characterization based on DSC of polysaccharides, gelatinization behaviour of starches and interaction of starch with other food components can be determined, and phase transitions during baking processes can be studied by DSC. Studies on crystallization and melting behaviour of fats observed by DSC indicate changes in lipid composition or help characterizing products. Thermal oxidative decomposition of edible oils examined by DSC can be used for predicting oil stability. Using DSC in the freezing range has a great potential for measuring and modelling frozen food thermal properties, and to estimate the state of water in foods and food ingredients. Research in food microbiology utilizes DSC in better understanding thermoadaptive mechanisms or heat killing of food-borne microorganisms. Isothermic microcalorimetric techniques provide informative data regarding microbial growth and microbial metabolism.  相似文献
