In a previous paper, we report a preliminary DSC study on bovine (BSA) and human (HSA) serum albumins. However, at accurate HPLC analysis the commercial proteins show three peaks: Fraction V-I, probably globulins (as declared by the producers), Fraction V-II (about 15–18% of the product) and Fraction V-III that represents pure BSA or HSA. A hypothesis is that the Fraction II is a covalent dimer, or trimer or a mixture of both, generated during the scalf-life of the commercial product. Denaturation enthalpies of the purified Fraction V-III and Fraction V-II of BSA, have been determined calorimetrically, at changing thepH, and the results of both compared with those obtained on the untreated protein. Few calorimetric experiments have been also carried on a BSA monomer derivative with sulphidril group protected. Computer program have been developed for the deconvolution of exo- and endothermic effects and for the analysis of thermal denaturation profiles.  相似文献

Oxygen dependence of a tyrosinase‐based electrochemical biosensor for determination of phenol in aqueous and organic media was systematically investigated. The result demonstrated that the enzymatic coupling reaction rate of tyrosinase (deoxy form) and O2 to regenerate tyrosinase (oxy form) is a kinetically fast reaction, and the significant change of O2 concentration in aqueous solution did not affect the coupling reaction. The further increase of O2 concentration did not increase the overall oxidation reaction rate of the substrate at low substrate concentration (e.g.,<10 μM phenol) when O2 concentration was greater than 8.9 ppm. The oxygen dependence was observed in the case of high substrate concentration due to insufficient amount of O2 available for the regeneration of tyrosinase. In other words, the upper linear range is oxygen dependent for tyrosinase biosensors. The phenol biosensors employing microelectrodes had wider upper linear ranges than macroelectrodes in both aqueous and organic phase, which can be explained by the oxygen dependence.  相似文献

The effect of temperature on the π∗←π transition band in the UV absorption spectrum of NO3(aq) centered at ≈200 nm was studied in the temperature range 10–70 C. The observed temperature independence of this band was in contrast to the significant influence of temperature on the nitrate π∗←n transition reported recently by us. However, taking into account the electronic states involved in both the transitions, it was concluded that this finding was in accordance with our previous assumption that interconversion between spectrally distinct (with respect to π∗←n band) nitrate species included the rupture/formation of hydrogen bond(s) in the hydration shell of the nitrate ion.  相似文献

Ammonium persulfate (APS), 2,2′‐azobis(amidinopropane) dihydrochloride (V50) and 4,4′‐azobis(4‐cyanovaleric acid) (ACVA) were utilized to prepare temperature‐sensitive poly(N‐isopropylacrylamide) (PNIPAM) microgels by precipitation polymerization under various reaction pH conditions. Their particle sizes and swelling ratios depended on the reaction pH due to the pH dependence on the ionization degree of the decomposed fragments originating from the initiators and their hydrophilicity‐hydrophobicity. The more hydrophobic initiator, under the reaction pH conditions used, could be partitioned to a greater extent into the microgel particles due to the hydrophobicity of PNIPAM chains at the reaction temperature, which led to a more cross‐linked structure inside the microgels resulting in their smaller swelling ratio. pH dependence of surface charge density of the microgels with amidino groups or carboxylic acid groups on their surfaces was evidenced by the variation of their zeta potentials as a function of pH.  相似文献

A flow microcalorimeter/densimeter system has been commissioned to measure heat capacities and densities of solutions containing radioactive species as a function of temperature. Measurements were made for NaTcO4(aq) at six temperatures (189.15 K to 373.15 K for the heat capacities, 287.43 K to 396.67 K for the densities) over the molality range 0.01 to 0.29 mol-kg–1. Measurements for NaReO4(aq) (NaReO4 is a common nonradioactive analogue for NaTcO4) were made under similar conditions, but for eight temperatures and a more extensive range of molalities, 0.05 to 0.65 mol-kg–1. Heat capacities of NaCl(aq) reference solutions were also measured from 293.15 K to 398.15 K.The heat capacity and density data are analysed using Pitzer's ioninteraction model. Equations for the apparent molar heat capacities and volumes are reported. Values of the NaReO4(aq) partial molar heat capacities are compared to literature values based on integral heats of solution. The agreement between the two sets of NaReO4 results is good below 330 K, but only fair at the higher temperatures. Values of the partial molar volumes have also been derived. Using literature values and the results of our experiments, it is calculated that the disproportionation of hydrated TcO2(s) to form TcO 4 (aq) and Tc(cr) occurs more readily at high temperatures. The uncertainties introduced by using thermodynamic values for ReO 4 (aq), in the absence of values for TcO 4 (aq), are discussed.  相似文献

Some cationic surfactants such as cetyltrimethylammonium bromide (CTAB) show a very spectacular viscoelasticity in solution, in even very diluted states, with some aromatic substances added, such as salicylic acid. Formerly, the authors established that the remarkable solution behaviour was the result of the entanglement of such enormously elongated giant micelles, based on recognition of the existence of large aggregates, through direct observation of them as substantial images under the electron microscope.In this report, we further confirm the relation between micelle length and solution viscoelasticity after arbitrarily obtaining different size aggregates by altering the media pH, or by admixing shorter chain length surfactants than CTAB, which alone did not induce viscoelasticity.  相似文献

A series of rheological experiments was performed for a polypropylene (PP) melt to explore its elongation behavior through a capillary die. Using an advanced twin-bore capillary rheometer with dies measuring 1.0, 0.5, and 0.25 mm in diameter, the experiments were performed at 210, 220, and 230 °C. The results indicated that the temperature of the PP melt had a significant effect on its extensional viscosity. The different decreases in the extensional viscosity values in the tested dies revealed the geometry dependence of the extensional viscosity. In the case of PP in the 0.25 mm die at 210 °C, the extensional viscosity values under different extensional strain rates were much higher than those in the other dies. Only in the 1.0 mm die did the relationship between the extensional viscosity of PP and its temperature obey the Arrhenius equation due to the larger die size which related to a slight size effect on its elongation behavior. The calculated deviations of the extensional viscosity in the tested dies demonstrated that the increasing pressure applied to the PP melt in the micro channel was related to the geometry dependence of the elongation behavior of the PP melt. The change in the extensional viscosity eventually relied on the interaction of the die geometry, the temperature, and the extensional stress of the PP melt.  相似文献
