The assigned structure of the dinoflagellate‐derived toxin belizentrin was prepared by total synthesis in form of the corresponding methyl ester for stability reasons. The successful route features an unusual solution for the preparation of a recalcitrant ylide on a C‐glycosidic segment; moreover, it involves an asymmetric hetero‐Diels–Alder reaction en route to the tertiary hemiacetal substructure, a Negishi cross‐coupling of two elaborate building blocks, and a macrocyclization based on an intramolecular aminolysis of a spirolactone. A modified Kocienski olefination ultimately allowed the polyol side chain to be attached to the macrocycle although this transformation faced the exceptional base sensitivity of this polyunsaturated target compound. 相似文献

The molecule is essentially planar, with the largest deviations occurring at bromine (–0.21 Å), carbonyl oxygen O(l) (+0.19 Å) and in ring-A (C(9) –0.15 Å, C(10) –0.15 Å). Total syntheses of the title marine natural products have been achieved and so confirming the structures originally assigned to them. Upon subjecting agesasine A and its corresponding ethyl ester to Mitsunobu conditions, a 1,5-cyclodehydration reaction takes place to give 2-oxazolines. In contrast, on subjecting agesasine B to the same Mitsunobu conditions, a simple dehydration reaction occurs to give the corresponding acrylate. A total synthesis of longamide E was achieved by engaging a 1,2-disubstituted pyrrole in a lactam-forming reaction and this was followed by a two-fold and fully regio-controlled bromination reaction. A distinctly different and possibly biomimetic route was used to synthesize, via the open-chain natural product nakamurine B, longamide B and its methyl ester. Preliminary biological evaluations of the title alkaloids and various analogues against a small human cancer cell line panel reveals cytotoxic properties that vary significantly with structure. 相似文献

The Lacinutrix genus was discovered in 2005 and includes 12 Gram-negative bacterial species. To the best of our knowledge, the secondary metabolite production potential of this genus has not been explored before, and examination of Lacinutrix species may reveal novel chemistry. As part of a screening project of Arctic marine bacteria, the Lacinutrix sp. strain M09B143 was cultivated, extracted, fractionated and tested for antibacterial and cytotoxic activities. One fraction had antibacterial activity and was subjected to mass spectrometry analysis, which revealed two compounds with elemental composition that did not match any known compounds in databases. This resulted in the identification and isolation of two novel isobranched lyso-ornithine lipids, whose structures were elucidated by mass spectrometry and NMR spectroscopy. Lyso-ornithine lipids consist of a 3-hydroxy fatty acid linked to the alpha amino group of an ornithine amino acid through an amide bond. The fatty acid chains were determined to be iso-C15:0 (1) and iso-C16:0 (2). Compound 1 was active against the Gram-positive S. agalactiae, while 2 showed cytotoxic activity against A2058 human melanoma cells. 相似文献

In this work, the motion of a two-dimensional rectangular freely floating body under waves is simulated using Improved Meshless Local Petrov-Galerkin method with Rankine Source function (IMLPG_R) with variable spacing resolutions. The IMLPG_R method is a particle method that solves Navier–Stokes equations using the fractional step method to capture the wave properties. However, many existing particle methods are computationally intensive to model the wave-floating body due to the requirement of fine particles, needing uniform distribution throughout the domain. To improve the computational efficiency and capture the body response properly, variable spaced particle distribution with fine resolution near the floating body and coarse resolution far from the body is implemented. Numerical schemes to handle variable resolutions are reported. An iterative scheme to handle the wave-floating body is implemented in the particle method. Two test cases, one with small wave and another with steep waves, are simulated for uniform particle distribution and the result shows good agreement with literature. Based on this, the performance of the variable spaced particle distribution is tested in coupling with floating body solver. The application of the method for wave impact load from the green water loading of the floating structure is also simulated. 相似文献

Solar panels and bio‐optical sensors play a significant and growing role in a number of applications that are of importance to many organizations. Many of these instruments require a high transmission of radiation into the device for it to work properly. A major issue faced is that harsh marine environments often aid in the growth or development of fouling on the coverglass used to protect the instruments. Over a period of time in an ocean environment, some plant or animal may attach itself to the coverglass, ultimately obscuring the glass and rendering the instrument useless. As such, an antifouling mechanism is needed for these instruments that is inexpensive, long‐lasting, and environment friendly. The approach discussed herein involves the use of known antifouling chemicals which have been incorporated into the polymer matrix. Polymethylmethacrylate (PMMA), bisphenol A polycarbonate (Bis A PC), and a co‐polyterephthalate (CPTE) were examined. The plaques are optically transparent and previous work has shown that, for most samples, the materials display a minimal decrease in mechanical behavior upon the addition of the algaecides. This paper will discuss the effects on the materials' optical properties when exposed to both harsh marine conditions as well as high intensity UV light.
