海洋天然产物研究新进展

海洋天然产物化学是目前天然产物化学中最活跃的研究领域之一。近年来,从海洋生物中分离到非常多的化学结构和生理活性上令人注目的新化合物,引起了有机化学家和药物化学家的关注。本文按萜类、太环内酯、聚醚类、生物碱,环肽、含氰化合物、甾醇、聚丙酸酯类化合物等化学结构来概述海洋天然产物研究近年来的进展。     

Xyloketal G,a Novel Metabolite from the Mangrove Fungus <Emphasis Type="Italic">Xylaria sp.</Emphasis> 2508

A novel metabolite 1, named xyloketal G, was isolated from cultures of marine derived mangrove fungus Xylaria sp. 2508. Its structure was elucidated by analysis of spectroscopic data.__________Published in Khimiya Prirodnykh Soedinenii, No. 1, pp. 22–23, January–February, 2005.     

3β-Methoxyolean-18-ene (miliacin) from the marine fungus Chaetomium olivaceum

A pentacyclic tritepenoid, 3-methoxyolean-18-ene (miliacin), was isolated for the first time from the marine fungus Chaetomium olivaceum. Its structure was determined on the basis of 2D NMR spectroscopy and X-ray diffraction data.     

Total synthesis of the marine alkaloid halitulin

The structure of the strongly cytotoxic marine alkaloid halitulin (1) has been confirmed by total synthesis and its absolute configuration determined as (15S). The synthesis follows a strategy previously reported by one of us and uses an efficient preparation of the quinoline-7,8-diol unit by modified Baeyer-Villiger and Skraup reactions. The O-benzyl protecting groups were removed in the last step of the synthesis by transfer hydrogenolysis without concomitant reduction of the quinoline ring. The method can be applied for the synthesis of halitulin analogues.     

SARS-CoV protease inhibitors design using virtual screening method from natural products libraries

Two natural products databases, the marine natural products database (MNPD) and the traditional Chinese medicines database (TCMD), were used to find novel structures of potent SARS-CoV protease inhibitors through virtual screening. Before the procedure, the databases were filtered by Lipinski's ROF and Xu's extension rules. The results were analyzed by statistic methods to eliminate the bias in target-based database screening toward higher molecular weight compounds for enhancing the hit rate. Eighteen lead compounds were recommended by the screening procedure. They were useful for experimental scientists in prioritizing drug candidates and studying the interaction mechanism. The binding mechanism was also analyzed between the best screening compound and the SARS protein.     

八十年代海洋天然有机化学的进展

八十年代以来,海洋天然有机化学仍然继续发展,大量新的化合物,尤其是具有生理活性的化合物不断出现,海洋天然物的生物合成和人工合成研究也有新的进展。在海洋生物中,研究得最多的是海藻和海绵,其次便是珊瑚。     

Influence of media nutrients on synthesis of lignin peroxidase from Aspergillus sp

The effect of carbon and nitrogen sources, lignocellulosic substrates, and metal ions on lignin peroxidase (LiP) activity of Aspergillus sp., which was isolated from a mangrove area, was studied. Glucose (1%) was found to be the best carbon source. Among the various lignocellulosic substrates used, coir pith at 3% concentration increased LiP activity twofold on the second day of incubation. Peptone and KNO₃ completely inhibited the enzyme synthesis while (NH₄)₂SO₄ at 12.5 mM produced maximum activity. Since seawater contained all the requisite metal ions, any added ions had a negative effect on activity. Cu²⁺ had the most inhibiting effect while K⁺ the least. When all the optimized conditions were provided, in nitrogen- and carbon-sufficient medium, a maximum LiP activity of 345 U/mL was obtained on the second day of incubation.     

Iterative synthesis of the ABCDEF-ring system of yessotoxin and adriatoxin

A stereocontrolled linear synthesis of the ABCDEF-ring system of yessotoxin and adriatoxin, diarrhetic shellfish toxins, is described. Iterative application of a tetrahydropyran synthesis by reaction of the alkylation of a sulfonyl-stabilized oxiranyl anion followed by 6-endo cyclization of a 4,5-epoxy alcohol led to the synthesis of the trans-fused hexacyclic ether system, and the seven-membered E ring was constructed by ring expansion reaction.     

In Situ Correlations Between Polycyclic Aromatic Hydrocarbons (PAH) and Pah Metabolizing System Activities in Mussels and Fish in the Mediterranean Sea: Preliminary Results

Abstract

Biochemical indices based on enzymatic activities have been determined in fish and mussels sampled in various different coastal locations in the Mediterranean Sea. Preliminary results show a good agreement between biochemical measurements in marine organisms and chemical analyses of polycyclic aromatic hydrocarbons present in sediments. The results obtained suggest the use of biochemical indices for application in chemical contaminant biomonitoring.
