Ionic liquid and buffer mixture media are first reported in the peroxidase‐catalyzed polymerization of phenol. Yield of 100% with molecular weights of 7000 KDa, as assessed by size‐exclusion chromatography (SEC), were attained using 1‐butyl‐3‐methylimidazolium tetrafluoroborate–buffer mixtures with added hydrogen peroxide. The simplicity of the process and the low vapor pressure of the solvent media allow an eco‐friendly alternative to the general synthesis of polyphenolic‐type biopolymers. Evidence for the consequent polyphenol (PPO) was obtained from solid‐state 13C cross‐polarization magic angle spinning (CP‐MAS) NMR spectroscopy and FT‐IR. 采用聚乙二醇单甲醚(mPEG)为亲水段,聚赖氨酸(PzLL)为疏水段,通过二硫键和碳氮双键串联桥连合成了两嵌段共聚物(mPEG-CN-SS-PzLL),其中的二硫键具有还原敏感性,碳氮双键具有pH酸敏感性。通过红外光谱和核磁共振谱等手段测试分析了产物的化学结构。将聚合物通过透析法自组装制备得到双刺激响应型纳米载药粒子。结果表明:该纳米载药粒子的药物包封率较高,达到52%。该载药系统在还原环境或酸性环境下具有良好的体外释药性能。  相似文献

Small peptides in serum are potential biomarkers for the diagnosis of cancer and other diseases. The identification of peptide biomarkers in human plasma/serum has become an area of high interest in medical research. However, the direct analysis of peptides in serum samples using mass spectrometry is challenging due to the low concentration of peptides and the high abundance of high-molecular-weight proteins in serum, the latter of which causes severe signal suppression. Herein, we reported that porous semiconductor-noble metal hybrid nanostructures can both eliminate the interference from large proteins in serum samples and significantly enhance the matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) yields of peptides captured on the nanostructure. Serum peptide fingerprints with high fidelity can be acquired rapidly, and successful discrimination of colorectal cancer patients based on peptide fingerprints is demonstrated.  相似文献

Present work demonstrates the utilization of surface modified polycarbonate (PC) membrane as solid phase and antibody conjugated CdSe/ZnS quantum dots (QDs) as fluorescent label for the sensitive and selective detection of Salmonella typhi (S. typhi) in water in a period of 2.5 h. PC membrane was surface modified with glycine and activated by EDC/NHS for immobilization of S. typhi specific IgG. Antibody immobilized porous PC membrane was incubated with bacteria contaminated water for immunocapturing of S. typhi. Antibody conjugated QDs were also prepared by using carbodiimide chemistry. Both modified PC membrane and quantum dots were characterized by using various modern analytical tools. It was estimated that 1.95 molecules of QDs were successfully bio-conjugated per unit of IgG. PC membrane with captured bacteria was incubated with prepared IgG conjugated QDs for the formation of sandwich complex. Analysis of the regions of interest (ROI) in fluorescent micrographs showed that newly developed method based on PC and fluorescent QDs has 100 times higher detection sensitivity (100 cells/mL) as compared with detection using conventional dye (FITC) based methods.  相似文献

A novel matrix, gold nanoparticles-bacterial cellulose nanofibers (Au-BC) nanocomposite was developed for enzyme immobilization and biosensor fabrication due to its unique properties such as satisfying biocompatibility, good conductivity and extensive surface area, which were inherited from both gold nanoparticles (AuNPs) and bacterial cellulose nanofibers (BC). Heme proteins such as horseradish peroxidase (HRP), hemoglobin (Hb) and myoglobin (Mb) were successfully immobilized on the surface of Au-BC nanocomposite modified glassy carbon electrode (GCE). The immobilized heme proteins showed electrocatalytic activities to the reduction of H2O2 in the presence of the mediator hydroquinone (HQ), which might be due to the fact that heme proteins retained the near-native secondary structures in the Au-BC nanocomposite which was proved by UV-vis and IR spectra. The response of the developed biosensor to H2O2 was related to the amount of AuNPs in Au-BC nanocomposite, indicating that the AuNPs in BC network played an important role in the biosensor performance. Under the optimum conditions, the biosensor based on HRP exhibited a fast amperometric response (within 1 s) to H2O2, a good linear response over a wide range of concentration from 0.3 μM to 1.00 mM, and a low detection limit of 0.1 μM based on S/N = 3. The high performance of the biosensor made Au-BC nanocomposite superior to other materials as immobilization matrix.  相似文献

Trenbolone (TRE) is a steroid used by veterinarians on livestock to increase appetite and body weight. The use of TRE has been restricted because of its harmful side effect for consumers. To effectively control TRE residue in food and food product, a rapid and convenient immunoassay was developed by preparing an anti-TRE monoclonal antibody. The immunogen and coating antigen were prepared by coupling TRE hapten with carrier proteins via 1-ethyl-3-(dimethylaminopropyl)carbodiimide hydrochloride (EDC) method. The optimized method gave an average IC50 value of 0.323 ng mL−1 towards TRE and an average detection limit (LOD) of 0.06 ng mL−1, which is much lower than the maximum residue levels (2.0 ng g−1) accepted by the Joint FAO/WHO Expert Committee on Food Additives (JECFA). The specificity of the antibody was evaluated by measuring cross-reactivity of six structurally related compounds, including 19-nortestosterone (9.7%), testosterone (0.13%), methyltestosterone (<0.01%), methandrostenolone (<0.01%), (+)-dehydroisoandrosterone (<0.001%) and β-estradiol (<0.001%). The recovery rates of the test in detection of TRE-fortified animal tissue, urine and animal feed samples were in the range of 81.3-89.4%, while the intra- and inter-assay coefficients of variation were less than 12.0%.  相似文献

将金纳米棒(AuNRs)和辣根过氧化物酶(HRP)以自组装的方式依次修饰到Au电极表面,构建了响应过氧化苯甲酰(BPO)生物传感器。采用循环伏安法和电流时间法研究了传感器的电化学性质和最佳工作条件;由于HRP/AuNRs复合膜的协同效应,电极性能得到显著改善。在最佳工作条件下(工作电压-0.02V,pH 7的磷酸盐缓冲体系),BPO浓度在5.0×10-6~1.0×10-4 mol/L范围内与电极的电流响应值呈良好的线性关系,线性回归方程为:i(μA)=12.6796C(mmol/L)+0.2406,R=0.9993。电极的检出限为8.5×10-7 mol/L。电极用于面粉中BPO的测定,平行测定6次,平均回收率为97.9%~100.1%,相对标准偏差(RSD)为0.5%~2.1%。本方法用于商品面粉中的BPO测定,取得满意结果。  相似文献
