6-Hydroxy chromone-3-carbaldehyde-(4′-hydroxy) benzoyl hydrazone (L) and its Ln (III) complexes, [Ln = La, Nd, Eu and Tb]
have been prepared and characterized on the basis of elemental analyses, molar conductivities, mass spectra, 1H NMR, thermogravimety/differential thermal analysis (TG-DTA), UV-vis spectra, fluorescence spectra and IR spectra. The formula
of the complex is [Ln L·(NO3)2]·NO3. Spectrometric titration, ethidium bromide displacement experiments and viscosity measurements indicate that Eu (III) complex
bind with calf-thymus DNA, presumably via an intercalation mechanism. The intrinsic binding constant of Eu (III) with DNA

Gold-coated iron oxide nanoparticle Hepatitis B virus (HBV) DNA probes were prepared, and their application for HBV DNA measurement
was studied. Gold-coated iron oxide nanoparticles were prepared by the citrate reduction of tetra-chloroauric acid in the
presence of iron oxide nanoparticles which were added as seeds. With a fluorescence-based method, the maximal surface coverage
of hexaethiol 30-mer oligonucleotides and the maximal percentage of hybridization strands on gold-coated iron oxide nanoparticles
were (120 ± 8) oligonucleotides per nanoparticle, and (14 ± 2%), respectively, which were comparable with those of (132 ± 10)
and (22 ± 3%) in Au nanoparticle groups. Large network aggregates were formed when gold-coated iron oxide nanoparticle HBV
DNA gene probe was applied to detect HBV DNA molecules as evidenced by transmission electron microscopy and the high specificity
was verified by blot hybridization. Our results further suggested that detecting DNA with iron oxide nanoparticles and magnetic

We study the elastic responses of double-(ds) and single-stranded (ss) DNA at external force fields. A double-strand-polymer
elastic model is constructed and solved by path integral methods and Monte Carlo simulations to understand the entropic elasticity,
cooperative extensibility, and supercoiling property of dsDNA. The good agreement with experiments indicates that short-ranged
base-pair stacking interaction is crucial for the stability and the high deformability of dsDNA. Hairpin-coil transition in
ssDNA is studied with generating function method. A threshold force is needed to pull the ssDNA hairpin patterns, stabilized
by base pairing and base-pair stacking, into random coils. This phase transition is predicted to be of first order for stacking

We have studied the dynamic process of interactions between a DNA chain and a histone octamer by numerical simulations.It is found that DNA indeed may wrap around the histone octamer about two turns as in the actual situations.The simulation shows that the interaction potential between DNA and histone is a key factor for the wrapping of DNA,and the temperature is also an important parameter in the process. 相似文献

采用6种化学发光体系和两种比色体系研究了芳香植物罗勒和香柠檬的非挥发性组分(简称罗勒、香柠檬)直接清除活性氧、活性氮和对^.OH引起的DNA损伤的保护作用。结果表明,罗勒和香柠檬不仅能直接清除O2,而且可抑制黄嘌呤氧化酶的活性从而减少O2的产生。罗勒和香柠檬也能有效清除^.OH、H2O2、ONOO^.和DPPH^.,两者相比,香柠檬除对^.OH的清除作用弱于罗勒外,清除以上各种自由基的能力均强于罗勒。结果还显示,罗勒和香柠檬都能抑制脂质过氧化,明显地抑制和延缓DNA损伤,起到预防型抗氧化剂和断链型抗氧化剂的双重作用。这些结果表明,罗勒和香柠檬的非挥发性组分是有效的、多功能的天然抗氧化剂和自由基清除剂。 相似文献

荧光探针技术可以用来测定药物-核酸相互作用的强度。通过药物与核酸相互作用,使DNA与探针键合的程度减小,反映在探针荧光光谱的改变,从而可以了解药物和核酸的作用机理。相互作用常数D为DNA 探针中加入药物后探针的荧光强度相对于DNA 探针荧光强度减去纯探针荧光强度的下降百分数。文章利用盐酸小檗碱(Berberine)作为探针,测定几种抗癌药物加入DNA与荧光探针(盐酸小檗碱)混合溶液的荧光谱,探讨它们与DNA的相互作用。并根据相同浓度的不同药物与核酸相互作用的常数D确定作用强弱。 相似文献

采用时间分辨的发光光谱技术,分别测量了两种新合成的钌配合物[Ru(bpy)2 (dpbpd(NH2)2)]2 和[Ru(phen)2(dpbpd(NH2)2)]2 与小牛胸腺脱氧核糖核酸(ctDNA)相互作用时的瞬态发光动力学过程,并与以往对[Ru(phen)2dppz]2 等的研究结果进行对比,从而研究带-NH2的嘧啶环对配合物与DNA作用动力学过程的影响.结果表明:这两种含有带-NH2的嘧啶环的钌配合物与DNA相互作用时的发光按双指数规律衰减,发光寿命为几十纳秒,比dppz类钌配合物与DNA作用时的发光衰变寿命(几百纳秒)小一个数量级.归因于嘧啶环上的N和-NH2可能与水分子、DNA的碱基对或磷酸骨架形成氢键,从而加快激发态的无辐射弛豫,削弱发射光强,缩短发光寿命.该结论为进一步研究配合物分子与DNA的相互作用的机理提供了一定的依据. 相似文献

We report model calculations on DNA single strands which describe the equilibrium dynamics and kinetics of hairpin formation
and melting. Modeling is at the level of single bases. Strand rigidity is described in terms of simple polymer models; alternative
calculations performed using the freely rotating chain and the discrete Kratky-Porod models are reported. Stem formation is
modeled according to the Peyrard-Bishop-Dauxois Hamiltonian. The kinetics of opening and closing is described in terms of
a diffusion-controlled motion in an effective free-energy landscape. Melting profiles, dependence of melting temperature on
loop length, and kinetic time scales are in semiquantitative agreement with experimental data obtained from fluorescent DNA
beacons forming poly(T) loops. Variation in strand rigidity is not sufficient to account for the large activation enthalpy
of closing and the strong loop length dependence observed in hairpins forming poly(A) loops. Implications for modeling single
