Most recently, melamine has raised international concern for its catastrophic health effects stemming from tainted infant formula. So far there is limited information concerning the pharmacokinetics of melamine in mammals. The present report concerns the development and validation of a sensitive HPLC‐ESI‐MS/MS method for the pharmacokinetic study of melamine in rat. The method employed a simple liquid–liquid extraction process for plasma sample cleanup, and the extraction recoveries of melamine from plasma were consistent at different concentrations. There was a linear relationship between chromatographic area and concentration over the range of 10–5000 ng/mL for melamine in plasma (R = 0.995). In this work, for the first time, melamine was administered intravenously and orally to Sprague–Dawley rats and the pharmacokinetic characteristics of this contaminant were investigated. The mean values of major pharmacokinetic parameters of oral availability, the mean steady‐state distribution volume (Vss), clearance, and plasma elimination half‐life (T1/2) of melamine in Sprague–Dawley rats were 72.9 ± 13.2%, 102.5 ± 12.5 mL/kg, 20.1 ± 3.8 mL/h/kg, and 4.9 ± 0.5 h, respectively. The rats pharmacokinetic study results suggested that melamine was predominantly restricted to blood or extracellular fluid and is not extensively distributed to most organ tissues. Meanwhile, melamine should be primarily eliminated by renal filtration for rats and does not undergo significant metabolism. These data should be useful to regulatory for risk assessment. 相似文献

We study the traffic behavior when a vehicle changes from the first lane to the second lane on a two-lane highway. We apply the optimal velocity model to the vehicular motion. If the incoming vehicle does not decelerate successfully, it crashes into the vehicle ahead. On the other hand, if the headway between the incoming vehicle and the vehicle behind on the second lane is not long sufficiently, the rear vehicle may come into collision with the incoming vehicle. The safety-collision transition occurs by changing the lane. The dynamical transition depends highly on the vehicular speed, the sensitivity, and the headway. Eggs containing live Japanese quail embryos were imaged using micro-magnetic resonance imaging (μMRI) at 24-h intervals from Day 0 to 8, the period during which the main body axis is being laid down and organogenesis is taking place. Considerable detail of non-embryonic structures such as the latebra was revealed at early stages but the embryo could only be visualized around Day 3. Three-dimensional (3D) changes in embryo length and volume were quantified and also changes in volume in the extra- and non-embryonic components. The embryo increased in length by 43% and nearly trebled in volume between Day 4 and Day 5. Although the amount of yolk remained fairly constant over the first 5 days, the amount of albumen decreases significantly and was replaced by extra-embryonic fluid (EEF). 1H longitudinal (T1) and transverse (T2) relaxation times of different regions within the eggs were determined over the first 6 days of development. The T2 measurements mirrored the changes in image intensity observed, which can be related to the aqueous protein concentrations. In addition, a comparison of the development of Day 0 to 3 quail embryos exposed to radiofrequency (rf) pulses, 7 T static magnetic fields and magnetic field gradients for an average of 7 h with the development of control embryos did not reveal any gross changes, thus confirming that μMRI is a suitable tool for following the development of live avian embryos over time from the earliest stages. 相似文献

The significant increase in the complexity and autonomy of the hardware systems renders the verification of the functional safety of each individual component as well as of the entire system a complex task and underlines the need for integrated, model based tools that would assist this process. In this paper the authors present such a tool, coupled with an approach to functional safety analysis, based on the integration of functional tests into the model itself. The analysis of the resulting model is done through a stochastic Bayesian model. ObjectivesTo evaluate the safety of ultrasound-guided high-intensity focused ultrasound (HIFU) ablation for patients with diffuse adenomyosis.MethodsThis was a retrospective cohort study. The data was collected from 417 symptomatic adenomyosis patients who underwent ultrasound-guided HIFU between January 2012 and December 2015 at 1st Affiliated Hospital of Chongqing Medical University, Chongqing, China. Among them were 260 patients with diffuse adenomyosis (Group D) and 157 patients with focal adenomyosis (Group F). All patients underwent contrast-enhanced magnetic resonance imaging (MRI) one week before and the day after HIFU treatment. Successful treatment with HIFU was measured by the non-perfused volume ratio (NPVR). Intraprocedural and postprocedural adverse effects and complications were recorded to assess the safety of the procedure. Patients were followed-up for three months post-treatment. Complications were given a grade A through F according to the SIR Standards.ResultsAll patients successfully completed the procedure, non-perfused regions appeared in 415 (99.5%) patients. The non-perfused volume ratio (NPVR) of Group D was significantly lower than that of Group F (P < 0.05). During the procedure, the odds ratio of skin-burning pain was 1.7 (OR = 1.617, 95% CI: 1.103–2.532), when comparing Group D with Group F, while the odds ratio of inguinal pain was equal to 2.0 (OR = 2.038, 95% CI: 1.161–3.580), when Group F was compared to Group D. 97 patients (23.3%) received nominal therapy due to complications ([Society of interventional radiology, SIR]-B grade), among them, there were 62 cases (23.8%) in Group D and 35 cases (22.3%) in Group F. No significant difference was found between the two groups (P > 0.05) and neither of the reported complications of SIR-C-SIR-F occurred within the two groups.ConclusionsBased on our results, ultrasound-guided HIFU is safe for the treatment of diffuse adenomyosis, and controlling the ablation zone is crucial to ensure patients’ safety. 相似文献

We study the dynamic process of the multiple-vehicle collision when a vehicle stops suddenly in a traffic flow. We apply the optimal-velocity model to the vehicular motion. If a vehicle does not decelerate successfully, it crashes into the vehicle ahead with a residual speed. The collision criterion is presented by vi(t)/Δxi(t)→∞ if Δxi(t)→0 where vi(t) and Δxi(t) are the speed and headway of vehicle i at time t.
