On‐line LC‐EC/ESI‐MS has been established as a fast and simple method to mimic some types of oxidation reaction of various drugs and to study the formation and structure of the resulting products. This technique has been applied to a 2,6,9‐trisubstituted purine, R‐roscovitine, which is known to be an inhibitor of some cyclin‐dependent kinases (CDKs) and a potential anticancer drug. Oxidation of R‐roscovitine in an electrochemical cell (EC), operated under various conditions, resulted in appearance of 6 major products. These were further analyzed by high‐resolution mass spectrometry, their structures were elucidated by accurate mass measurement and compared to previously identified R‐roscovitine in vitro/in vivo metabolites. Although none of the observed products was structurally identical to those identified previously in vitro/in vivo, all of them, except for the methoxylated products, resembled similarity due to appearing through the same reaction type. R‐roscovitine in the EC cell underwent N‐dealkylation of the isopropyl moiety, hydroxylation of the aromatic side‐chain, dihydroxylation, methoxylation and dimer formation. The hydroxylation product was identified as Olomoucine II, a R‐roscovitine derivative, which displays 10‐times higher CDK‐inhibiting activity than R‐roscovitine and the occurrence of which, as R‐roscovitine product, has not yet been observed in vitro/in vivo.  相似文献

The applicability of the headspace coupled to mass spectrometry for evaluation of the sensory quality of virgin olive oil samples is presented. The volatiles of the oil are directly transferred from the sample vial to the detector without chromatographic separation. The mass spectrum obtained can be considered as a fingerprint of the oil sample and can be used for classification purposes. After a training step with samples previously qualified following the official method, a classification model was created using the supervised technique soft independent modeling of class analogy (SIMCA). Eight negative (rancid, winey-vinegary, muddy sediment, hay-wood, vegetable water, earthy, fusty and musty-humidity) and three principal positive attributes (fruity, bitter and pungent) have been included in this study. With them, a classification model consisting of two main classes (extra- and lampante-virgin olive oil) was constructed. In addition, the unsupervised technique cluster analysis permited the discrimination among oils with different negative attributes. The proposed procedure has been applied to the classification of commercial samples (as extra- or lampante-virgin olive oils) and the results were compared with those provided by the expert's panel with acceptable correlation.  相似文献

A new platinum(II) complex with methionine sulfoxide was synthesized and characterized by chemical and spectroscopic techniques. Elemental analyses, mass spectrometric measurements (electrospray ionization quadrupole time-of-flight mass spectrometry), and thermal analyses of the solid compound fit the composition [(C5H10NO3S)Pt(µ-Cl)2Pt(C5H10NO3S)]?·?2.5H2O. Infrared spectroscopic data indicate coordination of the ligand to Pt(II) through the nitrogen of NH2 and the sulfur of the S=O group. 1H-15N nuclear magnetic resonance spectroscopic data confirm nitrogen coordination. Antibacterial activities were evaluated by antibiogram assays using the disc diffusion method. The platinum(II) complex showed antibacterial activity against Gram-negative Pseudomonas aeruginosa bacterial cells.  相似文献

Breast cancer (BC) remains a major cause of mortality, and early detection is considered important for reducing BC‐associated deaths. Early detection of BC is challenging in young women, due to the limitations of mammography on the dense breast tissue of young women. We recently reported results of a pilot proteomics study, using one‐dimensional polyacrylamide gel electrophoresis (1D‐PAGE) and mass spectrometry (MS) to investigate differences in milk proteins from women with and without BC. Here, we applied two‐dimensional polyacrylamide gel electrophoresis (2D‐PAGE) and MS to compare the protein pattern in milk from the breasts of a single woman who was diagnosed with BC in one breast 24 months after donating her milk. Statistically different gel spots were picked for protein digestion followed by nanoliquid chromatography tandem MS (nanoLC‐MS/MS) analysis. The upregulated proteins in BC versus control are alpha‐amylase, gelsolin isoform a precursor, alpha‐2‐glycoprotein 1 zinc isoform CRA_b partial, apoptosis‐inducing factor 2 and vitronectin. Several proteins were downregulated in the milk of the breast later diagnosed with cancer as compared to the milk from the healthy breast, including different isoforms of albumin, cholesterol esterase, different isoforms of lactoferrin, different proteins from the casein family and different isoforms of lysozyme. Results warrant further studies to determine the usefulness of these milk proteins for assessing risk and detecting occult disease. Dissociation of molecular hydrogen (H2) is extensively studied to understand the mechanism of hydrogenation reactions. In this study, H2 dissociation by Au1-doped closed-shell titanium oxide cluster anions AuTi3O7- and AuTi3O8- has been identified by mass spectrometry and quantum chemistry calculations. The clusters were generated by laser ablation and massselected to react with H2 in an ion trap reactor. In the reaction of AuTi3O8- with H2, the ion pair Au+-O22- rather than Au+-O2- is the active site to promote H2 dissociation. This finding is in contrast with the previous result that the lattice oxygen is usually the reactive oxygen species in H2 dissociation. The higher reactivity of the peroxide species is further supported by frontier molecular orbital analysis. This study provides new insights into gold catalysis involving H2 activation and dissociation.  相似文献

An analytical method was utilized to detect ppt levels of VOCs in air. The method was based on the US-EPA method TO-14, consisting of canister sampling, three module enrichment and GC/MS analysis. Target compounds included chlorofluorocarbons (four kinds), benzene and its derivatives (14), halogenated hydrocarbons (20), and others (three). The minimum detection limits of the method for the target compounds ranged from 0.016 to 0.040 ppb (0.06–0.23 μg/m3). The recoveries of the target compounds ranged from 77 to 113% and relative coefficients of variation (n=4) were 3.0–9.0%. The sampled air was stable for at least 14 days after pressurizing with humidified nitrogen gas at 200 kPa (absolute pressure). The method was applied to analyze the VOCs in the air of Yakushima, a remote island of south-west Japan where no distinct local pollution source is considered.  相似文献

Formation and Reactions of the CH2Li‐Derivatives of tBu2P–P=P(CH3)tBu2 and (Me3Si)tBuP–P=P(CH3)tBu2 With nBuLi, (Me3Si)tBuP–P=P(CH3)tBu2 ( 1 ) and tBu2P–P=P(CH3)tBu2 ( 2 ) yield (Me3Si)tBuP–P=P(CH2Li)tBu2 ( 3 ) and tBu2P–P=P(CH2Li)tBu2 ( 4 ), wich react with Me3SiCl to give (Me3Si)tBuP–P=P(CH2–SiMe3)tBu2 ( 5 ) and tBu2P–P=P(CH2–SiMe3)tBu2 ( 6 ), respectively. With tBu2P–P(SiMe3)–PtBuCl ( 7 ), compound 3 forms 5 as well as the cyclic products [H2C–P(tBu)2=P–P(tBu)–PtBu] ( 8 ) and [H2C–P(tBu)2=P–P(PtBu2)–P(tBu)] ( 9 ). Also 3 forms 8 with tBuPCl2. The cleavage of the Me3Si–P‐bond in 1 by means of C2Cl6 or N‐bromo‐succinimide yields (Cl)tBuP–P=P(CH3)tBu2 ( 10 ) or (Br)tBuP–P=P(CH3)tBu2 ( 11 ), resp. With LiP(SiMe3)2, 10 forms (Me3Si)2P–P(tBu)–P=P(CH3)tBu2 ( 12 ), and Et2P–P(tBu)–P=P(CH3)tBu2 ( 13 ) with LiPEt2. All compounds are characterized by 31P NMR Data and mass spectra; the ylide 5 and the THF adduct of 4 additionally by X‐ray structure analyses.  相似文献

Me2Sn(O2PPh2)2 ( 1 ), Ph2Pb(O2PMe2)2 ( 2 ), and Ph2Pb(O2PPh2)2 ( 3 ) have been synthesized by the reactions of Me2SnCl2 or Ph3PbCl with the corresponding diorganophosphinic acid in methanol. X‐ray diffraction studies show that the diorganophosphinate groups behave as double bridges between the metal atoms leading to polymeric ring‐chain structures with M2O4P2 (M = Pb, Sn) eight‐membered rings. The organic groups bonded to the metal atoms are in trans‐position in the resulting octahedral arrangement around the metal atoms. The IR and the mass spectra were reported and discussed.  相似文献

The fate of the crude oil under irradiation was studied. After the UV irradiation, the fraction present in the highest percentage shifted from C8–C9 fraction to C13 one, in GC–MS analysis. An increase of the relative amount of the C13–C25 fraction was observed, while a decrease in the relative amount of the C7–C12 fractions was present. The synchronous fluorescence spectrum showed a maximum at 396 nm. Two hours irradiation of the sample induced an increase of the fluorescence emission in the region 420–550 nm. After 20, 40, 60, and 100 h irradiation we observed a decrease of the fluorescence emission.  相似文献
