Fluorescence resonance energy transfer has been studied between lanthanide(III) chelates as donors and protein-coupled CdTe semiconductor nanoparticles as acceptors. Wide excitation spectra and large Stokes shift of semiconductor nanoparticles and timeresolved fluorescence detection were shown to provide a combination for successful energy transfer assay. Different intrinsically fluorescent europium(III) and terbium(III) chelates coupled to single biotin molecules were studied for optimal energy transfer with streptavidin labeled semiconductor nanoparticles. No significant differences between the studied chelates were observed. The strength of the methodology was demonstrated in a clinically relevant competitive and separation-free immunoassay of estradiol, where subnanomolar limit of detection was achieved with the coefficient of variation 2-11%. The data suggested that relatively short distance was needed to obtain adequate energy transfer. Therefore, biomolecules were coupled onto the semiconductor nanoparticles without any spacers. 相似文献

The potential for a simultaneous two-colour diagnostic scheme for nucleic acids operating on the basis of fluorescence resonance energy transfer (FRET) has been demonstrated. Upon ultraviolet excitation, two-colours of CdSe/ZnS quantum dots with conjugated oligonucleotide probes act as energy donors yielding FRET-sensitized acceptor emission upon hybridization with fluorophore (Cy3 and Alexa647) labeled target oligonucleotides. Energy transfer efficiencies, Förster distances, changes in quantum yield and lifetime, and signal-to-noise with respect to non-specific adsorption have been investigated. The dynamic range and limit-of-detection are tunable with the concentration of QD-DNA conjugate. The Cy3 and Alexa647 acceptor schemes can detect target from 4 to 100% or 10 to 100% of the QD-DNA conjugate concentration, respectively. Nanomolar limits of detection have been demonstrated in this paper, however, results indicate that picomolar detection limits can be achieved with standard instrumentation. The use of an intercalating dye (ethidium bromide) as an acceptor to alleviate non-specific adsorption is also described and increases signal-to-noise from S/N < 2 to S/N = 9-10. The ethidium bromide system had a dynamic range from 8 to 100% of the QD-DNA conjugate concentration and could detect target in a matrix containing an excess of non-complementary nucleic acid. 相似文献

Eighteen previously unknown γ-ray transitions were identified in the T
z = - 1/2 nucleus 51Fe following the fusion-evaporation reaction 32S(28Si,2α1n)51Fe. The level scheme reaches the fully aligned I
π = 27/2- terminating state of the five holes in the f
7/2 shell. The 17/2- state was found to be isomeric, and the lifetime was measured to be 2.87 ns. The mirror symmetry of 51Fe and 51Mn is discussed, and the level scheme of 51Fe is compared to shell-model calculations.

A chemically selective laser ion source has been used in a β-decay study of heavy Ag isotopes into even-even Cd nuclides.
Gamma-spectroscopic techniques in time-resolving event-by-event and multiscaling modes have permitted the identification of
the first 2+ and 4+ levels in 126Cd78, 128Cd80, and tentatively the 2+ state in 130Cd82. From a comparison of these new states in 48Cd with the E(2+) and E(4+)/E(2+) level systematics of 46Pd and 52Te isotopes and several recent model predictions, possible evidence for a weakening of the spherical N = 82 neutron-shell below double-magic 132Sn is obtained.

The decay of the 17 μs isomer of 133Sb was re-investigated experimentally. It was produced by thermal neutron induced fission of 241Pu. Its detection is based on time correlation between fission fragments selected by the LOHENGRIN spectrometer at ILL (Grenoble),
and the γ-rays, and conversion electrons from the isomer. The interpretation of the level scheme is based on shell model calculations,
where empirical two-body matrix elements were employed. The good agreement between theory and experiment suggests that the
isomer is the 21/2+ member of the 2p-1h πg
7/2ν(f

Gamow-Teller (GT) and spin-dipole (SD) states in 208Bi are studied by using self-consistent Hartree-Fock + Tamm-Dancoff approximation taking into account the coupling to the
continuum. Most of SD strength is found at the excitation energy E
x≈ 25MeV with a very broad width, which agrees with recent experimental observations. It is shown that Landau damping effect is responsible
for the large width of SD peak, while the escape width is found to be at most 1MeV. We study also electric dipole (E1) transitions between GT and SD states in 208Bi. Main E1 transitions for 0- and 1- states are found near excitation energy expected from Brink's hypothesis in which SD states are considered as E1 resonances built on the GT state. Calculated E1 transition strengths between GT and SD states are compared with the analytic sum rules within one-particle one-hole (1p-1h)
configuration space and within both 1p-1h and 2p-2h model space.
