Two new acyltetramic acids related to streptolydigin have been isolated from fermentations of Streptomyces lydicus. The principal members of this complex were resolved by TLC on silica gel. However, the methods of detection, permanganate spray or bioautography, were not suitable for both crude fermentation broths and purified extracts. Gas chromatography is unsuitable for the detection of either underivatized or silylated streptolydigins. High performance liquid chromatography (HPLC) particularly on triethylaminoethyl cellulose is rapid and sensitive and is the method of choice for the analysis of both crude and purified samples. Using high performance liquid chromatography, two components were detected in the complex, which are not observed using any of the other chromatographic procedures.  相似文献

Thermal lens detection with a 325.0 nm He-Cd excitation laser is used for thermooptical indirect detection in combination with the capillary electrophoretic separation of organic anions. The optimization of indirect thermooptical detection is discussed. With Mordant Yellow 7 (an azo dye) chosen as a probe ion limits of detection for 1-heptane-, 1-pentane-, 1-butane-, 1-propanesulfonic, and acetic acid at a level of n × 10−7 M were achieved with a separation electrolyte containing 50 μM of the probe ion and 5 mM Tris pH 9.90. A further increase in the detection sensitivity (twofold decrease in the limit of detection ) was obtained with a separation electrolyte containing a volume fraction of 20% acetonitrile.  相似文献

A system for separation of zinc traces from large amounts of cadmium is proposed in this paper. It is based on the solid-phase extraction of the zinc in the form of thiocyanate complexes by the polyurethane foam. The following parameters were studied: effect of pH and of the thiocyanate concentration on the zinc extraction, shaking time required for quantitative extraction, amount of PU foam necessary for complete extraction, conditions for the separation of zinc from cadmium, influence of other cations and anions on the zinc sorption by PU foam, and required conditions for back extraction of zinc from the PU foam. The results show that zinc traces can be separated from large amounts of cadmium at pH 3.0±0.50, with the range of thiocyanate concentration from 0.15 to 0.20 mol l−1, and the shaking time of 5 min. The back extraction of zinc can be done by shaking it with water for 10 min. Calcium, barium, strontium, magnesium, aluminum, nickel and iron(II) are efficiently separated. Iron(III), copper(II) and cobalt(II) are extracted simultaneously with zinc, but the iron reduction with ascorbic acid and the use of citrate to mask copper(II) and cobalt(II) increase the selectivity of the zinc extraction. The anions nitrate, chloride, sulfate, acetate, thiosulphate, tartarate, oxalate, fluoride, citrate, and carbonate do not affect the zinc extraction. Phosphate and EDTA must be absent. The method proposed was applied to determine zinc in cadmium salts using 4-(2-pyridylazo)-resorcinol (PAR) as a spectrophotometric reagent. The result achieved did not show significant difference in the accuracy and precision (95% confidence level) with those obtained by ICP–AES analysis.  相似文献

A simplified procedure based on ion-exchange separation of selenourea (Se-U) and selenocystamine (Se-CM), which have very close half-wave potential when they are simultaneously analyzed by voltammetric techniques, has been developed and optimized. Thus, selenocystamine remains in the cation exchanger Purolite C 100 H, whereas selenourea is found in the effluent and is determined by square wave cathodic stripping voltammetry using Na2CO3 as electrolyte. Selenocystamine is then eluted from the cation exchanger using 4 M HCl and analyzed by square wave cathodic stripping voltammetry in the HCl solution. For each voltammetric determination the corresponding parameters were investigated and optimized; the obtained detection limits were 0.3 ng Se mL–1 for Se-CM and 2 ng Se mL–1 for Se-U. A flow sheet for the separation of inorganic (Se(IV) and Se(VI)) and organoselenium compounds (Se-U, Se-CM, (CH3)2Se2, and (CH3)2Se) developed for their electrochemical detection is presented and it was successfully applied to a certified reference material, an environmental soil sample, and a urine sample.  相似文献

MCM-48 membranes have been prepared on alumina supports of different pore sizes. A battery of characterization techniques has been used to study the physical properties and the quality of the membranes prepared. The highest quality membranes were prepared on supports with pore size of up to 60 nm. The MCM-48 membranes were tested in the separation of gas phase mixtures and a cyclohexane/O2 selectivity higher than 270 was obtained. The selective separation of organic compounds from inert components is a result of the cooperative effects of capillary condensation in MCM-48 pores and of the specific interactions of the permeating compounds and the membrane material.  相似文献

Summary The elution behavior of alkali and earth alkaline cations has been studied on a newly synthesized weak cationexchanger (WCX). Applying this stationary phase isocratic separation of these cations is possible. The dependence of retention on competing ion concentration is described via a simple model. The influence of crown ethers as well as organic eluent additives on selectivity and peak efficiency is demonstrated. With optimized separation such cations can be determined with detection limits of about 0.5 ppm using a conductivity detector without ion suppression.  相似文献

Proposed in this paper is an improved method for removal of tin as the matrix element by volatilization in an anhydrous medium as mixed halides of different composition. Any one of them boils at a lower temperature than SnBr4. This ternary system enhances the eventual recombination of temporarily dissociated species to volatile ones, thus making the process quicker and more efficient. The presence of 2 mg of Pb(II) suffices to prevent Bi(III) from loss due to volatilization.  相似文献

This study reports on the direct HPLC stereoisomer separation of selected pyrethroic acids employing commercial cinchona alkaloid derived chiral stationary phases (CSPs). cis/trans-Chrysanthemic acid (cis/trans-CA), cis/trans-chrysanthemum dicarboxylic acid (cis/trans-CDCA), cis/trans-permethrinic acid (cis/trans-PA), and fenvaleric acid (FA) were resolved into the individual stereoisomers, i.e. enantiomers and diastereomers as well. To achieve satisfactory baseline separation an optimisation of the variables of the chromatographic method including chemical structure of the cinchona carbamate CSP, mobile phase composition, and flow rate was required. All four stereoisomers of PA were successfully separated in a single run (alphacis = 1.20, alphatrans = 1.26, critical Rs = 1.65) with an acetonitrile (ACN)-based polar-organic eluent. The complete baseline resolution of all CA stereoisomers succeeded in polar-organic (alphacis = 1.20, alphatrans = 1.35, critical Rs = 3.03) as well as in acetonitrile-based reversed-phase media (alphacis = 1.24, alphatrans = 1.22, critical Rs = 2.73). The latter elution mode was also found to be suitable for the enantio- as well as diastereoselective resolution of CDCA (alphacis = 1.09, alphatrans = 1.50, critical Rs = 1.43), which is to the best of our knowledge the first reported enantiomer separation of this analyte. The enantiomers of FA could be baseline separated employing also reversed-phase mode (alpha = 1.16, Rs = 2.91). These separation methods may be applied for quality control processes in the production of stereoisomerically pure insecticides as well as stereoselective toxicokinetic studies, as CDCA, PA, and FA are suitable biomarkers for monitoring human pyrethroid burden.  相似文献

A novel PBI/P84 co-polyimide dual-layer hollow fiber membrane has been specifically fabricated through the dry-jet wet phase inversion process, for the first time, for the dehydration pervaporation of tetrafluoropropanol (TFP). Polybenzimidazole (PBI) was chosen as the outer selective layer because of its superior hydrophilic nature and excellent solvent-resistance together with robust thermal stability, while P84 co-polyimide was employed as the inner supporting layer because of its good solvent-resistance and thermal stability. The PBI/P84 membrane exhibits superior water selectivity and relatively high permeation flux. At 60 °C, the PBI/P84 dual-layer hollow fiber membrane shows a permeation flux of 332 g/(m2 h) and a separation factor of 1990 for a feed solution containing of 85 wt% TFP. The preferential water sorption and the significant diffusivity difference between TFP and water are the main causes of high separation factor. However, an increase in feed temperature will greatly increase the permeation flux but seriously decrease the water selectivity. The activation energy data verify that water can preferentially permeate the PBI membrane due to the strong water affinity of PBI and a much smaller molecular size of water.  相似文献

As a model for a binary alloy undergoing an unmixing phase transition, we consider a square lattice where each site can be either taken by an A atom, a B atom, or a vacancy (V), and there exists a repulsive interaction between AB nearest neighbor pairs. Starting from a random initial configuration, unmixing proceeds via random jumps of A atoms or B atoms to nearest neighbor vacant sites. In the absence of any interaction, these jumps occur at jump rates A and B, respectively. For a small concentration of vacancies (c v=0.04) the dynamics of the structure factorS(k,t) and its first two momentsk 1(t),k 2 2 (t) is studied during the early stages of phase separation, for several choices of concentrationc B of B atoms. Forc B=0.18 also the time evolution of the cluster size distribution is studied. Apart from very early times, the mean cluster sizel(t) as well as the moments of the structure function depend on timet and the ratio of the jump rates (= B/ A) only via a scaled timet/(). Qualitatively, the behavior is very similar to the direct exchange model containing no vacancies. Consequences for phase separation of real alloys are briefly discussed.  相似文献
