The investigation of multi‐site ligand–protein binding and multi‐step mechanisms is highly demanding. In this work, advanced NMR methodologies such as 2D 1H–15N line‐shape analysis, which allows a reliable investigation of ligand binding occurring on micro‐ to millisecond timescales, have been extended to model a two‐step binding mechanism. The molecular recognition and complex uptake mechanism of two bile salt molecules by lipid carriers is an interesting example that shows that protein dynamics has the potential to modulate the macromolecule–ligand encounter. Kinetic analysis supports a conformational selection model as the initial recognition process in which the dynamics observed in the apo form is essential for ligand uptake, leading to conformations with improved access to the binding cavity. Subsequent multi‐step events could be modelled, for several residues, with a two‐step binding mechanism. The protein in the ligand‐bound state still exhibits a conformational rearrangement that occurs on a very slow timescale, as observed for other proteins of the family. A global mechanism suggesting how bile acids access the macromolecular cavity is thus proposed. 相似文献

The absorption spectra of methane at different path lengths and different pressures for three temperatures 180, 240 and 296 K have been recorded in the 5556-6166 cm−1 region using the Bruker IFS 120 HR and 125 HR high-resolution Fourier transform spectrometers. The multispectrum fitting procedure has been applied to these spectra to recover the spectral line parameters. The main goal of this procedure was the determination of self-broadening and self-pressure-induced shift coefficients and the exponents of their temperature dependences. These parameters have been derived for 406 assigned lines with good values of the signal to noise ratio. The rotational dependence of these parameters is discussed. 相似文献

The proposed satellite mission ACCURATE consists of a small constellation of satellites in low Earth orbit, combining microwave occultation for thermodynamic state profiling with infrared-laser occultation for greenhouse gas and line-of-sight wind profiling. The mission aims to detect six greenhouse gas molecules with four additional isotopologues (H2O, CO2, CH4, N2O, O3, CO, 13CO2, OC18O, HDO, and H218O) in the upper troposphere and lower stratosphere in the 4000-5000 cm−1 spectral region. Greenhouse gas profiles will be retrieved to within 1-2% accuracy using a ‘differential’ method, requiring two spectral points for each species - one to sample the spectral line and the other nearby to sample the baseline.An estimation of retrieval errors for the ACCURATE mission reveals that errors in spectroscopic line parameters dominate all other error sources. Poor knowledge of the spectroscopy introduces systematic errors into the retrieved greenhouse gas profiles. Using a simple approach, it was shown that the best line parameters currently available are too large to allow retrievals of greenhouse gases to within the stated ACCURATE mission goals of 1% accuracy for CO2 and 2% for all other species. Therefore, spectroscopic line parameters for targeted lines need to be improved before the ACCURATE mission can be launched. Requirements have been formulated in this direction, and laboratory experiments outlined that could meet these requirements. 相似文献

A transmission line model for subwavelength metallic grating with single cut is presented. The model is based on analogy between the subwavelength metallic structure and the microwave transmission line theory. The analytical expression for the transmission is derived by the transfer matrix method. To confirm our model, the finite-difference time-domain (FDTD) method simulations are carried out. The influences of the position of the cuts on the higher modes of slit are presented. It is found that the transmission line model gives analytical prediction about the evolution of the transmission spectra. 相似文献

According to the principle of double-beam interference imaging using an infrared broadband light source, a novel method based on the optical fiber low-coherence Young's interferometer for the measurement of programmable optical fiber delay line is proposed. By measuring the small length increments of delay fiber through the evaluation of the central positions of zero-order interference fringes, we successfully construct a system with a variety of important functions, such as real-time controlling, measurement and displaying. This system offers the benefit that it can be regulated precisely to generate interference stripes once again after the length of fiber segment changes. When the locations of central stripes are moved to coincide with the original markers, the determination of the corresponding fiber optical delay time becomes really simplified, as just by reading out the tinny displacement of the reference arm. Multiple groups of experimental results demonstrate the outstanding performance of the proposed system since it bears a measuring accuracy of μm and a measuring range from 1 mm to 20 cm. 相似文献

为了提高超宽带系统的辐射因子,对超宽带脉冲整形技术进行了深入研究,介绍了采用Blumlein线产生双极脉冲的高功率双极脉冲产生技术。对采用Blumlein线产生双极脉冲的原理进行了讨论,通过数值模拟分析了影响双极脉冲形成的主要因素。设计了一套Blumlein高功率双极脉冲形成线,在800 kV脉冲源上开展了高压实验研究,分析了开关及形成线长度对形双极脉冲的影响。在输入单极脉冲电压为652.0 kV、脉宽为2.1 ns的情况下,Blumlein双极脉冲形成线可以产生负峰电压为571.9 kV、正峰电压为550.4 kV、半周期为740 ps的双极脉冲,峰-峰值电压是入射脉冲峰值电压的1.72倍,辐射因子为4.54 MV。 相似文献
