通过广义的Laguerre类型轨道,在组态相互作用框架内,发展了一套从头计算程序,来研究原子中的电子结构.应用该程序,通过计算能量最小值,求出最优波函数.利用优化波函数的单电子密度分布和角双电子密度的最可几分布,研究了处于最低激发态的Be,B,C,N,O,Ne原子的sαpβ组态的几何活性原子态GAAS(geometricallyactive atomic state).结果显示,作为原子波函数的内禀性质,双电子密度最可几角分布的角度与通常由分子杂化轨道理论解释的相关原子的分子的结合键角近似相等.  相似文献

There is an urgent need for development of rapid and inexpensive techniques for detection of microRNAs (miRNAs), which are potential biomarkers of various types of cancer. In this paper, we describe a multiplexed electrochemical platform for determination of three cancer‐relevant miRNAs: miR‐21, let‐7a and miR‐31. The strategy combines the use of magnetic beads (MBs) modified with a commercial antibody for the efficient capture of the heteroduplexes formed by hybridization of the target miRNA with DNA probe. Free non‐hybridized region of the DNA probe was thereafter hybridized with two biotin‐labeled auxiliary DNA probes in a process of hybridization chain reaction (HCR), resulting in a long hybrid bearing a large number of biotin molecules. Labeling of these multiple biotin units with streptavidin‐peroxidase conjugates allowed an amplification of the amperometric signal measured after capturing the modified MBs at a screen‐printed carbon electrode array of eight electrodes. The combined strategy demonstrated in a similar assay time significantly higher sensitivity than those previously described using modified MBs with the same capture antibody (without amplification by HCR) or a HCR strategy implemented on the surface of MBs, respectively. The methodology exhibits a good selectivity for discriminating single mismatches and was applied to the determination of the three target miRNAs in total RNA (RNAt) extracted from various cancer cell lines and from cervical precancerous lesions.  相似文献

We demonstrated recently that large nanomembranes became available from inorganic, organic/inorganic and totally organic precursors. Spin coating of mixtures of acrylate monomers and zirconium butoxide (or tetraethoxysilane), when combined with simultaneous radical polymerization and sol-gel reaction, gave 10–30 nm thick, free-standing nanomembranes composed of metal oxides and crosslinked acrylates as interpenetrating network. This approach was extended to thermosetting resins (epoxy, melamine and phthalic resins), to fabricate robust, free-standing nanomembranes. Mechanical properties of the nanomembranes as measured by the bulge test and by a compression method were comparable to those of the corresponding macroscopic resins. The hybrid nanomembrane was subsequently prepared by supramolecular assembly of the nanofilm with surfactants and lipids. These amphiphiles produced adsorbed monolayers on the nanomembrane and some of the assemblies were stably kept in water.  相似文献

利用第一性原理计算Mn在ZnS(001)表面上几种掺杂位置的形成能、局域分波态密度和磁矩.对Mn在ZnS(001)表面上的三种位置的形成能进行比较,得到两种填隙位置是非常稳定的掺杂位置.分析ZnS(001):Mn各种再构表面的电子态密度和电荷密度分布.结果表明,三种表面模型中,自旋向上的Mn原子的3d态和近邻S原子的3p态都有一定的杂化,并且替代掺杂的Mn和邻近S原子的p-d杂化最明显,形成的共价键最强.而自旋向下的Mn原子的3d态比较局域,受S原子的3p态影响较小.计算了三种掺杂表面的磁矩,并分析计算结果.  相似文献

Alzheimer’s disease (AD) is a progressive neurodegenerative disorder and the leading cause of dementia worldwide. The limited pharmacological approaches based on cholinesterase inhibitors only provide symptomatic relief to AD patients. Moreover, the adverse side effects such as nausea, vomiting, loss of appetite, muscle cramps, and headaches associated with these drugs and numerous clinical trial failures present substantial limitations on the use of medications and call for a detailed insight of disease heterogeneity and development of preventive and multifactorial therapeutic strategies on urgent basis. In this context, we herein report a series of quinoline-thiosemicarbazone hybrid therapeutics as selective and potent inhibitors of cholinesterases. A facile multistep synthetic approach was utilized to generate target structures bearing multiple sites for chemical modifications and establishing drug-receptor interactions. The structures of all the synthesized compounds were fully established using readily available spectroscopic techniques (FTIR, 1H- and 13C-NMR). In vitro inhibitory results revealed compound 5b as a promising and lead inhibitor with an IC50 value of 0.12 ± 0.02 μM, a 5-fold higher potency than standard drug (galantamine; IC50 = 0.62 ± 0.01 μM). The synergistic effect of electron-rich (methoxy) group and ethylmorpholine moiety in quinoline-thiosemicarbazone conjugates contributes significantly in improving the inhibition level. Molecular docking analysis revealed various vital interactions of potent compounds with amino acid residues and reinforced the in vitro results. Kinetics experiments revealed the competitive mode of inhibition while ADME properties favored the translation of identified inhibitors into safe and promising drug candidates for pre-clinical testing. Collectively, inhibitory activity data and results from key physicochemical properties merit further research to ensure the design and development of safe and high-quality drug candidates for Alzheimer’s disease.  相似文献

基于分子信标(MB)识别和荧光纳米粒子探针技术,建立了均相体系中李斯特菌目标DNA的高灵敏检测新方法.首先以羊抗人免疫球蛋白(IgG)标记的异硫氰酸荧光素(FITC)为核材料,成功制备了FITC-IgG@SiO2核壳荧光纳米粒子,有效防止了传统方法中采用单一FITC制备纳米颗粒时泄露严重的问题.随后以FITC-IgG@SiO2荧光纳米粒子和纳米金分别标记单核细胞增生李斯特菌序列特异性分子信标探针5'端和3'端,成功构建了单核细胞增生李斯特菌序列特异性分子信标荧光纳米探针.在实验优化条件下,α(令α=F/F0,F代表MB和目标DNA杂交以后的荧光强度,F0代表MB完全闭合时的荧光强度)与目标DNA浓度在1~200pmol/L浓度范围内呈良好的线性关系,检出下限为0.3pmol/L,相对标准偏差为2.6%(50pmol/L,n=11).将该方法应用于食品样品中单核细胞增生李斯特菌的检测,结果与国标法一致.  相似文献

从胡椒酸出发,通过还原、甲磺酸酯化、偶联和成盐四步反应合成了一系列新型的胡椒基咪唑盐类化合物,其结构经1HNMR, 13CNMR,HRMS以及X射线单晶衍射确定.对合成的新化合物进行了体外抗肿瘤细胞毒活性筛选,结果表明,1-((苯并[d][1,3]二氧杂环戊烯-5-基甲基)-3-(2-萘甲基))-5,6-二甲基-1H-苯并[d]咪唑-3-溴盐(30)具有显著的细胞毒活性,对HL-60、SMMC-7721、A-549、MCF-7和SW-480肿瘤细胞株的活性均优于顺铂(DDP),尤其对HL-60肿瘤细胞株表现出较好的选择性细胞毒活性,其IC50值约为顺铂的7.2倍.进一步研究表明,化合物30具有诱导SMMC-7721细胞株在细胞周期G0/G1期阻滞和细胞凋亡的作用.  相似文献

Spin-polarized photoemission with circularly polarized light represents a relatively new technique in surface science. It became feasible with the increased availability of circularly polarized light in the vacuum ultraviolet and soft X-ray regime, generated in dedicated synchrotron radiation facilities. Another important ingredient was the development of efficient electron spin polarization detectors. Supported by group theoretical considerations, this technique was first employed to study the process of optical spin orientation in single crystalline non-magnetic materials. In these samples, spin-dependent effects arise solely from spin-orbit coupling. The experiments revealed the strong influence of spin-orbit coupling on the details of the electronic band structure, such as the symmetry character of the electronic wave functions, hybridization phenomena, and the behavior of degeneracies. The results opened the way to a detailed understanding of the electronic structure and permit a rigorous test of both state-of-the-art bulk band theories and fully relativistic photoemission calculations. Since 1990, spin-polarized photoemission with circularly polarized light is also used to investigate ferromagnetic systems. These experiments led to the discovery of the counterpart of optical spin orientation in ferromagnets, namely, the magnetic dichroisms. Magnetic dichroism means that the spin-dependent photoexcitation from a ferromagnetically ordered system manifests itself already in the photoelectron intensity distributions. Because of this particular property, techniques based on magnetic dichroisms are currently receiving wide interest in the spectroscopic investigation of magnetic systems and phenomena.

The following article reviews the current status of the field of spin- and angle-resolved photoemission from solids using circularly polarized radiation. We survey the development of this technique over the last 10 years, covering its applications to both non-magnetic and ferromagnetic systems.  相似文献
