Several improvements in sample pretreatment for the determination of organic pollutants (i.e. n-alkanes, polycyclic aromatic hydrocarbons, PAHs) in marine biota (mussels) are presented. The use of liquid nitrogen and homogenization of the samples are shown to be an alternative to the time consuming liophilization step required for the analysis of biota samples. Microwave-assisted hydrolysis and extraction are combined to isolate organic pollutants (19 n-alkanes and 27 PAHs) from biota matrices. Experimental design (ED) and artificial neural networks (ANNs) were used to optimize the experimental conditions. NIST-CRM 2978 was used to test the validity of the developed method which shows a good agreement with certified values.  相似文献

A method for determining quadruple points of a two-component system containing a simple hydrate phase is proposed. This method utilizes the quasi-static change of the system along three-phase equilibrium lines and was proved to be able to determine the quadruple points as accurately as the conventional method. By using this method, even though some preparation is necessary, a quadruple point can be determined in just a single experimental run. The behavior of the system near the quadruple points was also examined experimentally, for both the quasi-static and the irreversible change cases. At the quadruple points, the temperature and pressure of the system were kept constant for a while, as at the triple point of water. In both cases, the representative point of the state of the system passed through the quadruple point on a pT diagram.  相似文献

Short-chain alpha-neurotoxins from snakes are highly selective antagonists of the muscle-type nicotinic acetylcholine receptors (nAChR). Although their spatial structures are known and abundant information on topology of binding to nAChR is obtained by labeling and mutagenesis studies, the accurate structure of the complex is not yet known. Here, we present a model for a short alpha-neurotoxin, neurotoxin II from Naja oxiana (NTII), bound to Torpedo californica nAChR. It was built by comparative modeling, docking and molecular dynamics using 1H NMR structure of NTII, cross-linking and mutagenesis data, cryoelectron microscopy structure of Torpedo marmorata nAChR [Unwin, N., 2005. Refined structure of the nicotinic acetylcholine receptor at 4A resolution. J. Mol. Biol. 346, 967-989] and X-ray structures of acetylcholine-binding protein (AChBP) with agonists [Celie, P.H., van Rossum-Fikkert, S.E., van Dijk, W.J., Brejc, K., Smit, A.B., Sixma, T.K., 2004. In complex with the receptor, NTII was located at about 30 A from the membrane surface, the tip of its loop II plunges into the ligand-binding pocket between the alpha/gamma or alpha/delta nAChR subunits, while the loops I and III contact nAChR by their tips only in a 'surface-touch' manner. The toxin structure undergoes some changes during the final complex formation (for 1.45 rmsd in 15-25 ps according to AMBER'99 molecular dynamics simulation), which correlates with NMR data. The data on the mobility and accessibility of spin- and fluorescence labels in free and bound NTII were used in MD simulations. The binding process is dependent on spontaneous outward movement of the C-loop earlier found in the AChBP complexes with alpha-cobratoxin and alpha-conotoxin. Among common features in binding of short- and long alpha-neurotoxins is the rearrangement of aromatic residues in the binding pocket not observed for alpha-conotoxin binding. Being in general very similar, the binding modes of short- and long alpha-neurotoxins differ in the ways of loop II entry into nAChR.  相似文献

A procedure is proposed in which the determination of hydroquinone using a flow injection system with electrochemical detection is described. Size and coefficient of variation of the signal are optimised by a desirability function and a central composite design. The robustness of the optimum reached in the optimisation step is evaluated by means a Plackett-Burman design. The optimised FIA system is able to determine hydroquinone with a minimum detectable net concentration of 10 μg l−1 with a false positive probability of 0.05 and a false negative probability less than 0.05. In samples of bleaching cream, the proposed procedure has a recovery of 102.2% with standard deviation of 4.4% and a relative error of 6.2%.  相似文献

A simple and rapid microwave-assisted extraction (MAE) procedure was developed and optimized for the extraction of paclitaxel (Taxol) from the needles of yew trees Taxus baccata L. grown in Iranian habitats. The samples, immersed in a methanol-water mixture, were irradiated with microwaves in a closed-vessel system. The method was evaluated using a factorial design approach based on parameters such as extraction time, temperature, methanol concentration in water (v/v), and the ratio of grams of sample to 10 mL of solvent. Statistical treatment of the results revealed that the selected parameters were all significant except the extraction time. Optimum conditions would be 1.5 g samples in 10 mL solvent (90% methanol), an extraction temperature of 95 degrees C, and an extraction time of 7 min. The extracts has been analyzed by reverse-phase high-performance liquid chromatography with UV detection (LC/UV) at 227 nm for quantification. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) was used for confirmation. The main advantage of the proposed MAE method versus conventional solvent extraction (CSE) are the considerable reductions in time (7 min versus 16 h) and in solvent consumption (20 mL versus 150 mL). The MAE procedure yielded extracts that could be analyzed directly without any preliminary clean-up or solvent exchange steps. Both extraction methods show RSDs lower than 10% and lead to comparable recoveries of paclitaxel (87-92%).  相似文献

Summary Experimental Design methodology allows the modelling and optimization of the chromatographic separation of similar pesticides (triazine family) by GC and HPLC. The GC separation of simazin and atrazin is well modelled by a first degree equation, involving injected volume, carrier gas pressure and rising oven temperature. The LC is modelled by a second degree equation, depending on injected volume, eluent flow and composition. These calculated models allow easy optimization of the separations, using isoresponse curves.  相似文献

The vapor-liquid polycondensation of cyclohexylphosphoric dichloride with hydroquinone was investigated. The influence of the temperature, reaction time, base concentration, and molar ratio of reagents on the yield, inherent viscosity, and molecular weight of the obtained polymer was studied. Second order, central, composite, rotatable experimental design was used in order to carry out this work and to mark limits of the experimental field for optimal yields and high inherent viscosities.  相似文献

A survey about students' laboratory safety and environmental protection consciousness was performed in the freshmen majored in chemistry at Sun Yat-Sen University. Results showed that laboratory safety and environmental protection consciousness of most students' are weak. After analyzing the reasons, we put forward some suggestions. Cultivating students' laboratory safety and environmental protection consciousness should be carried out from several aspects such as emphasizing and regulating from school level, training in laboratory teaching, maintaining good cultural atmosphere of laboratory, as well as providing perfect laboratory safety and environmental protection facilities. We should guide students to actively learn knowledge and skills of laboratory safety and environmental protection in order to develop their good behavior and raise their consciousness.  相似文献

Robust design problems in aerodynamics are associated with the design variables, which control the shape of an aerodynamic body, and also with the so‐called environmental variables, which account for uncertainties. In this kind of problems, the set of design variables, which leads to optimal performance, taking into account possible variations in the environmental variables, is sought. One of the possible ways to solve this problem is by means of the second‐order second‐moment approach, which requires first‐order and second‐order derivatives of the objective function with respect to the environmental variables. Should the minimization problem be solved using a gradient‐based method, algorithms for the computation of up to third‐order sensitivity derivatives (twice with respect to the environmental variables and once with respect to the shape controlling design variables) must be devised. In this paper, a combination of the continuous adjoint variable method and direct differentiation to compute the third‐order sensitivities is proposed. This is shown to be the most efficient among all alternative methods provided that the environmental variables are much less than the design ones. Apart from presenting the method formulation, this paper focuses on the assessment of the so‐computed up‐to third‐order mixed derivatives through comparison with costly finite‐difference schemes. To this end, the robust design of a two‐dimensional duct is performed. Then, using the validated method, the robust design of a two‐dimensional cascade airfoil is demonstrated. Although both cases are handled as inverse design problems, the method can be extended to other objective functions or three‐dimensional problems in a straightforward manner. A finite point method for solving compressible flow problems involving moving boundaries and adaptivity is presented. The numerical methodology is based on an upwind‐biased discretization of the Euler equations, written in arbitrary Lagrangian–Eulerian form and integrated in time by means of a dual‐time steeping technique. In order to exploit the meshless potential of the method, a domain deformation approach based on the spring network analogy is implemented, and h‐adaptivity is also employed in the computations. Typical movable boundary problems in transonic flow regime are solved to assess the performance of the proposed technique. In addition, an application to a fluid–structure interaction problem involving static aeroelasticity illustrates the capability of the method to deal with practical engineering analyses.
