Along with histone modifications, RNA interference and delayed replication timing, DNA methylation belongs to the key processes in epigenetic regulation of gene expression. Therefore, reliable information about the methylation level of particular DNA fragments is of major interest. Herein the methylation level at two positions of the promoter region of the gene methylguanine‐O6‐DNA‐Methyltransferase (MGMT) was investigated. Previously, it was demonstrated that the epigenetic status of this DNA region correlates with response to alkylating anticancer agents. An automated CGE method with LIF detection was established to separate the six DNA fragments resulting from combined bisulfite restriction analysis of the methylated and non‐methylated MGMT promoter. In COBRA, the DNA was treated with bisulfite converting cytosine into uracil. During PCR uracil pairs with adenine, which changes the original recognition site of the restriction enzyme Taql. Artificial probes generated by mixing appropriate amounts of DNA after bisulfite treatment and PCR amplification were used for validation of the method. The methylation levels of these samples could be determined with high accuracy and precision. DNA samples prepared by mixing the corresponding clones first and then performing PCR amplification led to non‐linear correlation between the corrected peak areas and the methylation levels. This effect is explained by slightly different PCR amplification of DNA with different sequences present in the mixture. The superiority of CGE over PAGE was clearly demonstrated. Finally, the established method was used to analyze the methylation levels of human brain tumor tissue samples. 相似文献

本研究将开放式微波和直接超声波振荡两种不同的能量方式相结合,研制出超声-微波协同萃取装置。通过萃取土壤中微量多环芳烃(PAHs),对方法和仪器的可行性进行了初步评价。结果表明,在60 mL二氯甲烷-正已烷(1∶1,V/V)的混合萃取剂,100 W微波辐射功率(超声振动功率固定为50 W),萃取9~10m in,土壤中多环芳烃回收率达86.6%,相对标准偏差约4.0%。与索氏抽提、高压密闭和开放式微波等萃取方法相比,本方法具有样品容量大,萃取时间短,萃取效率受样品中含水量和溶剂极性影响小等优点。 相似文献

Carbon fabric (CF) was surface treated with silane-coupling agent modification, HNO3 oxidation, combined surface treatment, respectively. The friction and wear properties of the carbon fabric reinforced phenolic composites (CFP), sliding against GCr15 steel rings, were investigated on an M-2000 model ring-on-block test rig. Experimental results revealed that combined surface treatment largely reduced the friction and wear of the CFP composites. Scanning electron microscope (SEM) investigation of the worn surfaces of the CFP composites showed that combined surface modified CFP composite had the strongest interfacial adhesion and the smoothest worn surface under given load and sliding rate. SEM and X-ray photoelectron spectroscopy (XPS) study of carbon fiber surface showed that the fiber surface became rougher and the oxygen concentration increased greatly after combined surface treatment, which improved the adhesion between the fiber and the phenolic resin matrix and hence to improve the friction-reduction and anti-wear properties of the CFP composite. 相似文献

The thermal decomposition processes of silver behenate have been studied by infrared spectroscopy (IR), X-ray diffraction (XRD), combined thermogravimetry-differential thermal analysis-mass spectrometry (TG-DTA-MS), transmission electron microscopy (TEM) and UV-vis spectroscopy. The TG-DTA and the higher temperature IR and XRD measurements indicated that complicated structural changes took place while heating silver behenate, but there were two distinct thermal transitions. During the first transition at 138 °C, the alkyl chains of silver behenate were transformed from an ordered into a disordered state. During the second transition at about 231 °C, a structural change took place for silver behenate, which was the decomposition of silver behenate. The major products of the thermal decomposition of silver behenate were metallic silver and behenic acid. Upon heating up to 500 °C, the final product of the thermal decomposition was metallic silver. The combined TG-MS analysis showed that the gas products of the thermal decomposition of silver behenate were carbon dioxide, water, hydrogen, acetylene and some small molecule alkenes. TEM and UV-vis spectroscopy were used to investigate the process of the formation and growth of metallic silver nanoparticles. 相似文献

Industrial wastewater streams containing high concentrations of biorefractory materials like cyanides should ideally be treated at source. In the present work, degradation of potassium ferrocyanide (K4Fe(CN)6) as a model pollutant has been investigated using cavitational reactors with possible intensification studies using different approaches. Effect of different operating parameters such as initial concentration, temperature and pH on the extent of degradation using acoustic cavitation has been investigated. For the case of hydrodynamic cavitation, flow characteristics of cavitating device (venturi) have been established initially followed by the effect of inlet pressure and pH on the extent of degradation. Under the optimized set of operating parameters, the addition of hydrogen peroxide (ratio of K4Fe(CN)6:H2O2 varied from 1:1 to 1:30 mol basis) as process intensifying approach has been investigated. The present work has conclusively established that under the set of optimized operating parameters, cavitation can be effectively used for degradation of potassium ferrocyanide. The comparative study of hydrodynamic cavitation and acoustic cavitation suggested that hydrodynamic cavitation is more energy efficient and gives higher degradation as compared to acoustic cavitation for equivalent power/energy dissipation. A procedure for studying the first-passage failure of strongly non-linear oscillators with time-delayed feedback control under combined harmonic and wide-band noise excitations is proposed. First, the time-delayed feedback control forces are expressed approximately in terms of the system state variables without time delay. Then, the averaged Itô stochastic differential equations for the system are derived by using the stochastic averaging method. A backward Kolmogorov equation governing the conditional reliability function and a set of generalized Pontryagin equations governing the conditional moments of first-passage time are established. Finally, the conditional reliability function, the conditional probability density and moments of first-passage time are obtained by solving the backward Kolmogorov equation and generalized Pontryagin equations with suitable initial and boundary conditions. An example is worked out in detail to illustrate the proposed procedure. The effects of time delay in feedback control forces on the conditional reliability function, conditional probability density and moments of first-passage time are analyzed. The validity of the proposed method is confirmed by digital simulation. 相似文献
