A polyclonal antibody (Ab) for the progestin levonorgestrel (LNG) was generated, and immunochemical assays for its detection, clean-up and concentration were developed. A highly specific microplate diagnostic assay for the detection of LNG was developed that used the enzyme linked immunosorbent assay (ELISA) method. The LNG ELISA developed was sensitive and reproducible; it exhibited I50 and I20 values of 3.3 ± 1.8 ng mL−1 and 0.6 ± 0.4 ng mL−1, respectively, and the Abs did not cross react with any of the tested steroid hormones. The above Abs were used to develop a sol-gel-based immunoaffinity purification (IAP) method for concentration and clean-up of LNG that is compatible with subsequent immunochemical or instrumental chemical analytical procedures, such as liquid chromatography followed by mass spectrometry (LC-MS/MS). Development of the columns included successful entrapment of Abs within a tetramethoxysilane (TMOS)-based SiO2 polymer network. The Abs could bind the free analyte from solution, and the bound analyte could be easily eluted from the sol-gel matrix at high recoveries. The Ab selectivity towards the antigen was high, in both ELISA and the sol-gel columns, but the entrapped Abs cross-reacted with two other steroid hormones - ethynylestradiol (EE2) and nortestosterone (NT) - which share similar epitopes with LNG, despite the lack of cross reactivity in the ELISA. The validity of the method was investigated by LC-MS/MS and a good analytical correlation was obtained.  相似文献

We have studied the use of wide-band detection in conjunction with saturation of a rovibronic transition of OH within itsA 2 +X 2(0,0) band. For wide-band detection, in which fluorescence is detected from the entire excited rotational manifold, the fluorescence yield is sensitive to collisions in two ways. First, it is sensitive to the ratio of rate coefficients describing rotational energy transfer and electronic quenching; this ratio determines the number of neighboring rotational levels that are populated during the laser pulse. Second, the fluorescence yield can vary with the total collisional rate coefficient; only after a sufficient number of collisions, corresponding to 2.5 ns in an atmospheric flame, does the rotational manifold reach steady state. We also compare measurements employing wide-band (detecting theR 1 andR 2 branches) and narrow-band (detecting a single transition) saturated fluorescence of OH. Over a wide range of conditions — obtained by varying the equivalence ratio, temperature, N2 dilution, and pressure — the wide- and narrow-band fluorescence techniques compare well. Given this good agreement, wide-band saturated fluorescence could be especially useful for analyzing atmospheric flames with XeCl-excimer lasers; one can potentially obtain 2—D images of OH which have a high signal-to-noise ratio and a reduced sensitivity to laser irradiance and quenching.  相似文献

Rayleigh scattering and planar laser induced predissociative fluorescence are used to obtain 2-D images of total and specific densities inside a combustion bomb. The experimental arrangement is the same for both methods except for the selection of laser wavelength and the filtering of the radiated light. The former method yields a distribution of total densities while the latter provides densities of hot O2, i.e., those with v=2, J=17 and 33. Hydrogen-air mixtures of various compositions are used. Because the thermodynamics are well known, a bomb may serve as a reference device for diagnostics for high temperature species, and the results are in accord with calculations. Additional Rayleigh experiments are described which yield a) scattering cross sections at 193 nm, b) a 2-D temperature distribution in a hot air stream, and c) a 2-D temperature distribution, of limited precision, inside the bomb.  相似文献

The thermal degradation of poly(ether sulfone) (PES) and polysulfone (PSF) was studied with a combination of thermogravimetric analysis and stepwise pyrolysis–gas chromatography/mass spectrometry techniques with consecutive heating of the samples at fixed temperature intervals (100 °C) to achieve narrow‐temperature pyrolysis conditions. The individual mass chromatograms of various pyrolysates were correlated with pyrolysis temperatures to elucidate the pyrolysis mechanism. The major mechanism for both PES and PSF was a one‐stage pyrolysis involving main‐chain random scission and carbonization. The major products SO2 and phenol were released from the sulfone and ether groups in PES. The major products SO2, phenol, and 1‐methyl‐4‐phenoxybenzene were released from the sulfone, ether, and isopropylene groups in PSF. In the PES, the thermal stability of the sulfone and ether groups was identical to the maximum thermogravimetric loss rate. In the PSF, the thermal stability was in the following order: sulfone < ether < isopropylene. The temperature of the maximum thermogravimetric loss rate was similar to the maximum evolution of phenol. However, there was a considerable difference in the thermal behavior of both polymers; the correlation of the polymer structure to the degradation mechanism is discussed. © 2000 John Wiley & Sons, Inc. The main goal of this work is to identify polyurethane (PU) building blocks by pyrolysis gas chromatography/mass spectrometry (Py–GC/MS) and matrix-assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF MS). Toluene diisocyanate (TDI) and diphenylmethane diisocyanate (MDI) are widely used polymer building blocks. Py–GC/MS and MALDI-TOF MS were proved to be powerful methods to distinguish TDI-PU and MDI-PU according to the characteristic pyrolysis products and the different repeated units, respectively. In Py–GC/MS, the specific pyrolyzates are TDI for TDI-PU and MDI for MDI-PU. In MALDI-TOF MS, the weights of repeated units are 264?g/mol for TDI-PU and 340?g/mol for MDI-PU.  相似文献

Scattering amplitudes and intensities of partial azimuthal harmonics and the total intensity of the scattered field are calculated for the case of transmission of longitudinal acoustic waves through a liquid rotating with acceleration. It is shown that the intensity of transmitted waves depends on the angular acceleration and does not depend on the angular velocity of the vortex rotation.  相似文献

2 at 1064 nm, vaporization/fragmentation of soot primary particles and aggregates occurs. Optical measurements are performed using a second laser pulse to probe the effects of these changes upon the LII signal. With the exception of very low fluences, the structural changes induced in the soot lead to a decreased LII intensity produced by the second laser pulse. These two-pulse experiments also show that these changes do not alter the LII signal on timescales less than 1 μs for fluences below the vaporization threshold. A new mechanism whereby Neoclassical Tearing Modes (NTMs) can be triggered through toroidal mode coupling to a magnetic perturbation is proposed. The physical picture is the presence of a relatively small “pre-NTM” magnetic island whose frequency is modified by the perturbation, changing polarization current effects from stabilizing to destabilizing.  相似文献
